Project description:Natural product cinnamaldehyde has significant antifungal activities, and its inhibitory activity against Candida albicans has been reported. Up to now, there are few reports on the changes in the transcriptome of Candida albicans after cinnamaldehyde treatment. Here, RNA sequencing analysis of samples obtained from Candida albicans cells grown in the presence or absence of cinnamaldehyde was performed. A total of 123 differentially expressed genes were identified, of which 15 genes were up-regulated and 108 genes were down-regulated.

Transcriptomic characterization of Candida albicans in response to Aureobasidin treatment in vitro.

Project description:Purpose: Cellular response of Candida albicans to Aureobasidin A at transcriptomic level. Method: The transcriptome of C. albicans SC5314 upon exposure to 1 µg/ml AbA (0.5X Minimum Inhibitory Concentration) for three hours was analyzed by RNA sequencing (RNA-Seq) on Illumina HiSeq 4000 instrument using the 2 X 150 bp chemistry. Result: Following STAR-alignment of the quality-checked sequence reads against SC5314 reference genome and DESeq2 analysis of the read counts data, we identified 85 differentially-expressed genes between the AbA-treated cells and DMSO-treated controls, with false discovery rate <0.001 and a log2 fold change ≥ 1. Among them, 42 were significantly up-regulated and 43 were significantly down-regulated in AbA-treated cells as compared to the controls. The analysis of biological processes and molecular functions using Gene Ontology (GO) Term Finder of the Candida Genome Database (CGD) showed down-regulation of genes associated with cellular lipid biosynthetic process, and up-regulation of genes associated with regulation of membrane lipid distribution and floppase activity. The genes associated with cell aggregation, biofilm formation and DNA packaging were also affected. Conclusion: In summary, the analysis of transcriptomic data in this study provides beneficial and critical information contributing to our understanding of the cellular response of C. albicans to AbA and its antifungal activity.

Global transcriptomic analyses of the candida albicans response to treatment with a novel inhibitor of filamentation

Project description:The opportunistic pathogenic fungus Candida albicans can cause devastating infections in severely compromised patients. Its ability to undergo a morphogenetic transition from yeast to filamentous forms allows it to penetrate tissues and cause damage, and the expression of a number of pathogenetic mechanisms are also coordinately regulated with this yeast-to-hyphae conversion. Therefore, it is widely considered that filamentation represents one of the main virulence factors of C. albicans. We have previously identified N-[3-(allyloxy)-phenyl]-4-methoxybenzamide (compound 9029936) as the lead compound in a series of small molecule inhibitors of C. albicans filamentation, and characterized its activity, both in vitro and in vivo, as a promising candidate for the development of alternative anti-virulence strategies for the treatment of C. albicans infections. Here we have performed RNA sequencing analysis of samples obtained from C. albicans cells grown under filament-inducing conditions in the presence or absence of this compound. Overall treatment with compound 9029936 resulted in 618 up-regulated and 702 down-regulated genes. Not surprisingly some of the top down-regulated genes included well characterized genes associated with filamentation and virulence such as SAP5, ECE1 (candidalysin), and ALS3, as well as genes that impact metal chelation and utilization. Gene ontology analysis revealed an over-representation of cell adhesion, iron transport, filamentation, biofilm formation, and pathogenesis processes among down-regulated genes as result of treatment with this leading compound. Interestingly, the top up-regulated genes pointed to an enhancement of vesicular transport pathways, and in particular SNARE interactions.
