Project description:Chronic kidney disease (CKD) is a progressive condition characterized by sustained alterations in kidney structure and function. Long-term kidney fibrosis is marked by glomerulosclerosis, vascular sclerosis, and tubulointerstitial fibrosis. Therefore we investigated the therapeutic potential of BMSC-CM on RPTEC/TERT1 in a fibrotic environment. Using PAA gel platforms, we mimicked the stiffness typical of chronic kidney disease patients' kidneys.

Project description:Human renal proximal tubule cells (RPTEC/TERT1) were exposed to one of two nephrotoxic chemicals, ochratoxin A (OA) or potassium bromate (KB), or a vehicle control (CT). The exposure regime was a repeated dosing every 24h for 5 days followed by chemical washout and 3 days of recovery with medium renewal every 24h. Total experimental duration was 8 days. Cell lysates were prepared for microarray analysis after 1 day, 3 days and 5 days of exposure, and after 3 days of recovery (day 8).

High throughput targeted RNA sequencing on RPTEC/TERT1 cell line exposed to multiple concentration levels at various different time points

Project description:This experiment was carried out to investigate the time dynamics of the proximal tubule cells upon the exposure of cisplatin in multiple severity scenario by varying the concentration. The RPTEC/TERT1 cells which are immortalized human proximal tubule cells were differentiated for 14 days to reach the optimal maturity where the cells express the proximal tubule markers and transporter similarly to human proximal tubule cells. The cells were then exposed to cisplatin at multiple concentration levels ranging from 0-50 uM for 0,8,16, 24,48, and 72 hours. The cells were lysed and the lysates were collected for high throughput targeted RNA-seq with the human whole genome library. The gene expression profiles were analyzed to identify the modulated responses of the cells.This dataset is part of the TransQST project.

Identification of drug repurposing candidates for the treatment of polycystic kidney disease - RPTEC/TERT1 proteome

Project description:Autosomal dominant polycystic kidney disease (ADPKD) is a leading cause of end-stage renal disease with limited treatment options. Drug repurposing offers a promising strategy to find effective treatments. We identified birinapant, bardoxolone methyl and salicylic acid as repurposing candidates for ADPKD and applied thermal proteome profiling to identify disease-relevant targets. Our results uncovered shared effects between the three drugs, including a thermal shift in ADP/ATP translocases (SLC25A4/5/6) caused by reduced ATP production. As expected, salicylic acid displayed a polypharmacological profile but also engaged enzymes along the N-linked glycosylation pathway and inhibited PTK2, an activator of the proliferative PI3K/Akt/mTOR pathway. Orthogonal assays and computational modelling confirmed these findings and located the binding site (499-504) of salicylic acid within PTK2’s ATP-binding pocket. Engagement of several targets was achieved at clinically relevant concentrations.We have identified novel targets of three repurposing candidates that contribute to the cyst growth reducing effects in preclinical ADPKD models and identify salicylic acid, the metabolite of aspirin, as the repurposing candidate with the most promising mechanisms of action.This dataset contains files from the global protein analysis of RPTEC/TERT1 cells cultured on TC-inserts under physiological (10%; PO) and atmospheric (21%; AO) oxygen tension.
