Project description:In nature, moderate innate defensive behaviors (e.g. escaping and freezing) are commonly triggered by imminent threats in human and animal, accompanied with series of physiological responses related with activation of autonomic nervous system (e.g. tachycardia，increased blood pressure, tachypnea). According to many prior researches, posterior hypothalamic nucleus (PHN) with highly cell diversity is recognized to be connected tightly with panic attack. However, the neural substrate of sub-clusters of this important hypothalamic nucleus in panic attack-related defensive behavior has not yet elucidated. In order to justify the hypothesis, we adopted snDNA-seq to screen out a main population, Cbln2+ neurons, manipulating panic attack behaviors in PHN. Subsequently, we illustrated the neural coding mechanism of Cbln2+ PHN neurons for threats stimulus, and indicated the neurobiological functions of neural network of PHN neurons in defensive behaviors. In conclusion, our data suggested that Cbln2+ PHN neurons is sufficient and necessary to regulate panic attack related defensive behaviors in mice.

Transcription Factors Dlx1 and Otp Segregate the Identity of Orexigenic/Anti-thermogenic- and Growth Promoting-Neurons in the Arcuate Nucleus

Project description:The hypothalamic arcuate nucleus contains multiple types of neurons controlling critical physiological processes. However, the gene regulatory network directing their development remains rudimentary. Here we report that two transcription factors Otp and Dlx1 segregate the identity of orexigenic/anti-thermogenic NPY/AgRP- and growth-promoting GHRH-neurons in the developing arcuate nucleus. Otp-null and Dlx1-null mice lose NPY/AgRP and GHRH expression, respectively. Dlx1-null mice also show enhanced expression of Otp/NPY/AgRP, which is normalized in Dlx1;Otp-double mutant mice. Correspondingly, Dlx1-null mice exhibit decreased growth, lower body temperature and increased feeding. Furthermore, our genome-wide studies identify Otp as a negative target gene of Dlx1. Therefore, Otp is critical for NPY/AgRP-neuronal development, while Dlx1 promotes GHRH-neuronal development and antagonizes NPY/AgRP-neuronal development. These results identify a mechanism for segregating the identity of two functionally related neurons, and the Dlx1-Otp axis likely contributes to coordinating energy balance and growth by maintaining a proper ratio of NPY/AgRP- to GHRH-neurons.

Seq libraries are provided from STAT1 competent (Stat1+/+ x Rpl22HA/+) or STAT1-deficient (Stat1fl/fl x Rpl22HA/+) rLCMV-Cre carrier mice which have been challenged intravenously with LCMVwt at 5 weeks of age and then sacrificed after 9 days. These seq libraries represent the response of rLCMV-Cre infected neurons to CD8+ T cell attack following challenge with LCMVwt.

Project description:A substantial proportion of basal amygdala (BA) glutamate neurons project to nucleus accumbens (NAc). The evidence that these neurons are activated by reward and/or aversion is equivocal. Social stimuli are highly salient, and in male mice we conducted a detailed analysis of the responsiveness of BA-NAc neurons to estrous female (social reward, SR) or aggressive male (social aversion, SA). Both SR and SA activated c-Fos expression in a relatively high number of BA-NAc neurons in intermediate (int) BA. Using Fos-TRAP2 mice, the majority of social int-BA-NAc neurons were activated by either SR or SA, i.e. were monovalent, and in similar numbers. Fiber photometry provided corroborative evidence that int-BA-NAc neural pathway activity was similar in response to SR or SA. These findings contribute substantially to understanding the topography and valence-specificity of BA-NAc neurons with respect to highly salient stimuli, and to identifying molecular targets for treatment of reward- or aversion-specific psychopathologies.
