Project description:Genome-wide DNA methylation profiles in liquid based cytology (LBC) cervical scrapes samples was assessed using the Illumina Infinium Methylation850 BeadChip V1.0B4. The purpose of this study was to identify new candidate genes that are differentially methylated in squamous cell carcinoma compared to the DNA samples from cervical intraepithelial neoplasia grade (CIN) and normal cervical scrapes.
Project description:The Illumina Infinium 27k Human DNA methylation Beadchip v1.2 was used to obtain DNA methylation profiles across approximately 27,000 CpGs in normal cells from the uterine cervix (liquid based cytology samples), obtained from 152 women in a nested prospective case-control study within the ARTISTIC trial. Out of the 152 women, 75 developed a cervical intraepitelial neoplasia of grade 2 or higher (CIN2+) after 3 years of sample collection. The rest of women (77) remained disease free (CIN2-).
Project description:We profiled 798 miRNAs using NanoString technology in plasma, urine, and liquid-based cytology (LBC) from 24 women with and without high-grade cervical lesions (CIN2/3). LBC showed the highest detection efficiency and consistency, with the lowest proportion of low-count miRNAs and the highest expression levels, supporting its utility as the most effective specimen type for miRNA-based screening.

Project description:Human papillomavirus infection is the cause of essentially all cases of cervical premalignant lesions and cervical cancer. Primary screening using HPV DNA testing with cytology triage has been shown to be more sensitive and, importantly, more specific than conventional testing based on cytology among women more than 35 years of age. <br><br><br><br>The aim of the experiment was to evaluate a HPV genotyping microarray method using patient sample DNA. HPV genotyping is based on multiplex PCR (PGMY-t primers) followed by ligation step where two probes are ligated together if a matching template is present in the mixture. The ligated probes are then detected on microarray.
