Project description:Despite the dramatic changes in morphology occurring in the seminal vesicles during aging, the transcriptomic characteristics are poorly understood. Thus, we performed a bulk RNA sequencing on seminal vesicle samples from 3-month-old, 13-month-old, and 21-month-old mice to elucidate transcriptomic changes in this study. Comparing 3-month-old mice with the 21-month-old, inflammatory and immune response were identified as the most significantly biological process in upregulated differentially expressed genes (DEGs). Meanwhile, “chemokine signaling pathway” was identified as a key pathway. Indeed, a series of DEGs were enriched in “chemokine signaling pathway”, including Ccl6, Ccl8, Ccl9, Ccr2, Ccr5 and Ccr7. Hub genes such as Ptprc, Adgre1, H2-Ab1 and Ctss were obtained through Protein-Protein interaction network analysis among the continuously up-regulated differentially expressed genes in mouse seminal vesicles at three different ages. These analyses revealed an increased status of immune and inflammatory response in the aged seminal vesicles. In addition, multiple aging-associated DEGs were identified, including Vcam1, Apoe and Cdkn1a. To our knowledge, this study is the first to investigate the general transcriptome landscape of the seminal vesicles in a murine model of natural aging, which may provide novel insights involved in aging-related seminal vesicle dysfunction.

Project description:BACKGROUND: Prostate cancer is the most frequently diagnosed cancer among men in the United States. In contrast, cancer of the seminal vesicle is exceedingly rare, despite that the prostate and seminal vesicle share similar histology, secretory function, androgen dependency, blood supply, and (in part) embryonic origin. We hypothesized that gene-expression differences between prostate and seminal vesicle might inform mechanisms underlying the higher incidence of prostate cancer. METHODS: Whole-genome DNA microarrays were used to profile gene expression of 11 normal prostate and 7 seminal vesicle specimens (including 6 matched pairs) obtained from radical prostatectomy. Supervised analysis was used to identify genes differentially expressed between normal prostate and seminal vesicle, and this list was then cross-referenced to genes differentially expressed between normal and cancerous prostate. Expression patterns of selected genes were confirmed by immunohistochemistry using a tissue microarray. We identified 32 genes that displayed a highly statistically-significant expression pattern with highest levels in seminal vesicle, lower levels in normal prostate, and lowest levels in prostate cancer. Among these genes was the known candidate prostate tumor suppressor GSTP1 (involved in xenobiotic detoxification). The expression pattern of GSTP1 and four other genes, ABCG2 (xenobiotic transport), CRABP2 (retinoic acid signaling), GATA3 (lineage-specific transcription) and SLPI (immune response), was confirmed by immunohistochemistry. CONCLUSIONS: Our findings identify candidate prostate cancer genes whose reduced expression in prostate (compared to seminal vesicle) may be permissive to prostate cancer initiation. Such genes and their pathways may inform mechanisms of prostate carcinogenesis, and suggest new opportunities for prostate cancer prevention. Project description:The seminal vesicles synthesise bioactive factors that support gamete function, modulate the female reproductive tract to promote implantation, and influence developmental programming of offspring phenotype. Despite the significance of the seminal vesicles in reproduction, their biology remains poorly defined. Here, to advance understanding of seminal vesicle biology, we analyse the mouse seminal vesicle transcriptome under normal physiological conditions and in response to acute exposure to the reproductive toxicant acrylamide. Mice were administered acrylamide (25 mg/kg bw/day) or vehicle control daily for five consecutive days prior to collecting seminal vesicle tissue 72 h following the final injection.

Project description:During mammalian reproduction, sperm are delivered to the female reproductive tract bathed in a complex medium known as seminal fluid, which plays key roles in signaling to the female reproductive tract and in nourishing sperm for their onwards journey. The majority of seminal fluid is produced by a somewhat understudied organ known as the seminal vesicle. Here, we report a single-cell RNA-Seq atlas of the mouse seminal vesicle, generated using tissues obtained from 23 mice of varying ages, exposed to a range of dietary challenges. We define the transcriptome of the secretory epithelial cells in this tissue, identifying a relative homogeneous population of the epithelial cells responsible for producing the majority of seminal fluid. We also define the immune cell populations – including large populations of macrophages, dendritic cells, T cells, and NKT cells – which have the potential to play roles in producing various immune mediators present in seminal plasma. Together, our data provide a resource for understanding the composition of an understudied reproductive tissue with potential implications for paternal control of offspring development and metabolism.

Multiple models of human metabolism have been reconstructed, but each represents only a subset of our knowledge. Here we describe Recon 2, a community-driven,
consensus 'metabolic reconstruction', which is the most comprehensive representation of human metabolism that is applicable to computational modeling. Compared
with its predecessors, the reconstruction has improved topological and functional features, including ~2x more reactions and ~1.7x more unique metabolites. Using
Recon 2 we predicted changes in metabolite biomarkers for 49 inborn errors of metabolism with 77% accuracy when compared to experimental data. Mapping metabolomic
data and drug information onto Recon 2 demonstrates its potential for integrating and analyzing diverse data types. Using protein expression data, we automatically
generated a compendium of 65 cell type-specific models, providing a basis for manual curation or investigation of cell-specific metabolic properties. Recon 2 will
facilitate many future biomedical studies and is freely available at http://humanmetabolism.org/.

Project description:To investigate the identity of the tissue origin for metastases, we compared gene expression profiles of the prostate tissues and seminal vesicles from PB-Pten-NICD mice and those of 7 lung metastases from different mice. Three respective cell lines established from primary seminal vesicle tumors, primary prostate tumors, and lung metastases in PB-Pten-NICD mice were also included in the microarray analysis. We hypothesized that the gene expression profiles of primary and metastatic tumors should resemble those of their tissue origins. We identified genes differentially expressed between the seminal vesicle and prostate tumors. The expression profiles of the two cell lines established from primary tumors resembled those of their respective tissue origins, which supports our hypothesis. The expression profile of the metastatic cell line resembled those of the prostate tumor tissues, indicating the prostate as its tissue of origin. The expression profiles of 2 metastatic specimens each resembled those of the seminal vesicle tumors and the prostate tumors, respectively, while the expression profiles of the remaining 3 samples displayed a mixed pattern. This study suggests that metastatic tumors in this model may have originated from either the prostate, or the seminal vesicles, or both.

Project description:The seminal vesicles are an integral part of the male accessory gland system, and are responsible for producing a complex array of bioactive moieties that support gamete function and promote reproductive success. However, despite their physiological significance, the biology of the seminal vesicles remains poorly defined. Here, we exploit an established reproductive toxicant model, where mice were administered acrylamide (25 mg/kg bw/day) or control each morning for 5 consecutive days prior to collecting seminal vesicle tissue 72 h following the final injection, to complete the first proteomic assessment of mouse seminal vesicles. A total of 5,013 proteins were identified in the core seminal vesicle proteome with bioinformatics analysis of identifying cell proliferation, transcript/protein production and cellular death and survival pathways as prominent biological processes. Comparison of our acrylamide to control seminal vesicle proteome revealed 311 differentially regulated (-1.5 ≤ FC ≥ 1.5, p ≤ 0.05, 205 up-regulated, 106 down-regulated) proteins with immunoblotting analysis confirming proteomic data. Pathways that initiate protein synthesis to promote cellular survival were prominent amongst the dysregulated cellular pathways and RICTOR (Z-score = -3.64, p = 6.69E-07) was a top-ranked upstream driver. Oxidative stress was implicated as a contributor to protein changes with acrylamide leading to a significant increase in 8-OHdG in epithelial cells (5-fold increase, p = 0.016). Direct effects of acrylamide on seminal vesicle function were observed through a reduction in seminal vesicle secretion weight and overall protein content. Together these finding support the emerging concept that in addition to sperm, paternal exposures exert significant influence on the seminal vesicles, and identify new pathways to develop targeted interventions with which to manipulate fertility and reproductive outcomes.

Project description:The seminal vesicles are male accessory sex glands that contribute the bulk of the seminal plasma in which mammalian spermatozoa are bathed during ejaculation. In addition to helping convey sperm through the ejaculatory duct, seminal vesicle secretions support sperm survival after ejaculation and influence the female reproductive tract to promote receptivity to pregnancy. Despite its biological importance, there is limited understanding of seminal vesicle fluid (SVF) composition with only 75 proteins having been identified in publicly available proteomic analyses. To address this limitation, here we report new preparative methodology involving sequential solubilization of mouse SVF in guanidine hydrochloride, prior to acetone precipitation and analysis by label-free quantitative mass spectrometry. This strategy identified 126 proteins, including 83 previously undetected in SVF. Members of the seminal vesicle secretory protein family were the most abundant, accounting for 79% of all peptide spectrum matches. Functional analysis of SVF proteins identified inflammation and formation of the vaginal plug as the two most prominent biological processes. Notable additional processes included modulation of sperm function and regulation of the female reproductive tract immune environment. Together, these findings provide a robust methodological framework for future SVF studies and identify novel proteins with the potential to influence both male and female reproductive health.
