Project description:Background/Issue and Objectives: Health risks from chronic low dose radiation exposures encountered in environmental and occupational settings are uncertain. A key aspect to understanding health risks is precise and accurate modelling of the dose-response relationship. Benchmark dose (BMD) modeling is an approach used in chemical hazard assessments to identify the dose at which a pre-defined (e.g., 10%) change relative to background occurs. Herein, this method is being explored for radiation hazard assessments along with Adverse Outcome Pathways (AOPs), a knowledge framework of causally linked chains of key events (KEs) from a molecular initiating event to an adverse outcome (AO). Design/Method/Description: Blood was drawn from human participants (6 females and 8 males), and lymphocytes were isolated, cultured and X-irradiated at a lower dose-rate (LDR: 0.05 Gy/minute) and higher dose-rate (HDR: 1 Gy/minute) across nine different doses (0, 0.05, 0.10 0.25, 0.5, 1, 2, 4 and 6 Gy). Transcriptomic and proteomic changes were then assessed 24 hours post-exposure. Concurrently, cell membrane integrity and cellular ATP level were also measured, which are markers of radiation injuries. BMD values were then derived for each endpoint and pathways enriched in omics data were compared with Kes in an existing AOP to leukemia (www.aopwiki.org/aop/432). Conclusions/Impacts/Outcomes/Implications/Next Steps: By using AOPs as an organizational framework and BMD modeling for dose-response analysis, it provides a pragmatic platform to analyse complex data for radiation hazard assessment. Future work will entail in-depth analysis of the responses specific to LDR and HDR and effects of confounding factors on BMD values.

Project description:We have previously shown that low doses of ionizing radiation (LDIR) induce angiogenesis. In the present study we investigated their action in experimentally induced hindlimb ischemia. We demonstrated that 0.3 Gy, administered for four consecutive days, significantly improves blood perfusion in the murine ischemic limb by stimulating angiogenesis and arteriogenesis. This is achieved through durable and simultaneous up-regulation of a repertoire of pro-angiogenic factors and their receptors in endothelial cells, as evident in cells isolated from the irradiated gastrocnemius muscles. Moreover, we demonstrated that this mechanism is mediated via VEGFR signaling, since VEGFR inhibition abrogated the LDIR-mediated gene up-regulation and impeded the increase in vessel density. Importantly, the vasculature in an irradiated non-ischemic bed is not affected and no adverse effects associated to the use of LDIR were seen. These findings disclose an innovative, non-invasive strategy to induce therapeutic angiogenesis in a murine model of severe hindlimb ischemia, emerging as a novel approach in the treatment of Critical Limb Ischemia patients.

Project description:Gene expression profiles of peripheral blood samples from C57BL/6 mice exposed with ionizing radiation. We used mice as model animal to study biologial recovery response after radiation damage. Therefore, we obtained gene expression profiles from C57BL/6 mice exposed with various levels of ionizing radiation, including low and high doses and control groups. In order to measure recovery rate, we collected peripheral blood samples at different time durations after the exposure. In order to obtain robust signatures specific to radiation response, we are interested in knowing if the radiation signarures will be present in the presence of confounders. Therefore, mice were given intraperitoneal injections of lipopolysaccharide endotoxin (LPS), or treated with granulocyte colony-stimulating factor (GCSF), otherwise no treatment after ionizing radiation exposure. The underlying mechanism of confounder treatment is that LPS induces strong immune response resembling the effect of infection, and GCSF stimulates mobilization of HSCs. Exploratory analysis shows that the confounding effects did affect the radiation signature to some extent. This study provides insights into the molecular basis of time- and dose- dependent response to ionizing radiation in mice hematopoietic system.

Project description:In the context of radiotherapy, the effects elicited by ionizing radiation on the supportive stroma of tumors remains understudied. Tumor infiltrating fibroblasts are abundantly found in the stromal tissue of most tumor types and are key determinants of tumor progression and metastasis. Given the clinical attention that ablative radiotherapy is gaining nowadays, and the paramount role played by CAFs in cancer sustainability, in this study the effect of high-dose ionizing radiation on primary CAF?S have been investigated. We have used high-throughput transcriptome analysis to ascertain in which degree tumor stromal fibroblast become pro-tumorigenic or anti-tumorigenic after ablative irradiation. Hence, genome-wide transcriptome analysis has been carried out on CAFs isolated from NSCLC tumors after receiving a single dose of 18 Gray. The expressions of relevant genes have been validated by real-time PCR, and transformed cellular pathways have been checked by functional assays. Overall, our data reveals profound changes in biological pathways including cellular stress, DNA damage, cell cycle, aging, apoptosis, oxidative stress, and matrix remodeling. Ablative radiation doses may exert both pro-and anti-tumorigenic effects on CAFs. The ultimate effect of such disturbances merits further investigation in more complex in vivo settings.

A total of 536 C57BL/6 mice peripheral blood gene expression profiles were measured in 3 different batches using the Affymetrix mouse 430A 2.0 microarray. The experiment is designed to assess blood gene expression changes after exposure to ionizing radiation of 0, 100, 150, 200, 300, 450, 600, 800 and 1050 cGy. Samples were collected at 6, 24, 48, 72, 120 and 168hrs after a single dose exposure.
