Project description:Light intensity and spectral quality are two important factors that affect the cryopreservation success of plant germplasm. In a previous study, we found that LEDs with 90% red and 10% blue (RB) light have a positive effect on potato (Solanum tuberosum L.) shoot tip recovery during post-cryopreservation regeneration. In the present study, transcriptome profiles of non-cryopreserved (-) and cryopreserved (+) potato shoot tips exposed to RB, cool white fluorescent tubes (CW) and full darkness (D) were compared. Results indicated that light spectrum significantly affects transcript numbers in post-cryopreserved samples resulting in 3,284 differentially expressed genes of recovering shoot tips exposed to CW, RB and D. Gene ontology (GO) enrichment analysis revealed that several stress-related terms were more abundant in CW+, whereas morphogenesis-related processes were activated under RB+. Overall, cryopreservation had an extensive effect on gene expression, as 12,017 genes were differentially expressed in non-cryopreserved and cryopreserved shoot tips. Among all cryopreserved samples, enriched GO terms included various defense and stress responses to biotic stimuli and oxidative stress, whereas suppressed responses were mainly related to organ morphogenesis. The present study emphasizes that the optimization of light spectral properties to decrease light-related stress may greatly benefit the cryopreservation efficiency of plant germplasm.

Project description:Cryopreservation of mature oocytes is a critical means for female fertility preservation. Despite clinical advances using vitrification preservation method, the high concentrations of toxic penetrating cryoprotectant agents (CPA, up to 4.3 M) and low throughput (only one every experiment) put forward a challenge. Here, we report a synergetic ice inhibition platform via PVA/Fe3O4/GO nanoparticles (PFG NPs), that achieves sharp ice morphology, ice recrystallization, and devitrification inhibition, thus reducing cryodamage both in cooling and warming stages. Oocyte cryopreservation experiment demonstrates the survival rate can attain 98.6% using 2.5 M penetrating CPA while ensuring the scalability (ten oocytes each cryopreservation procedure). In contrast, recovered oocytes via PFG platform show 85 gene variation compared with fresh oocytes, whereas tradition CPA (TCPA) formulation induces 1396 gene changes. Meanwhile, the PFG-cryopreservation oocytes maintain normal ability of fertilization, development, and birth of offspring.

Project description:BackgroundPreservation of plant virus is a fundamental requirement in all types of virus-related research and applied applications. Development of efficient, reliable strategies for long-term preservation of plant virus would largely assist these studies.ResultsThe present study reported a novel biotechnology allowing cryopreservation of Apple stem grooving virus (ASGV) in living shoot tips. Following cryopreservation by droplet-vitrification or encapsulation-dehydration, about 62-67% of shoot regrowth and 100% of ASGV cryopreservation were obtained. Although shoot proliferation and virus concentration were reduced in cryopreserved diseased shoots after 8 weeks of shoot regeneration, continuous subculture for 4 times (16 weeks) increased shoot proliferation and virus concentration to comparative levels as those produced by shoot tip culture (as a control to shoot tip cryopreservation). Cryopreserved ASGV was efficiently transmitted to a woody plant by micrografting and to a herbaceous indicator by mechanical inoculation. Gene sequencing in three fragments of ASGV genome including coat protein and movement protein showed that cryopreserved ASGV shared 99.87% nucleotide identities with shoot tip culture-preserved virus, indicating cryopreserved virus is genetically stable.ConclusionsThe present study demonstrates ASGV, a representative virus that can infect meristematic cells of shoot tips, can be efficiently cryopreserved in shoot tips. To the best of our knowledge, this is the first report on plant virus cryopreservation in living tissues, and has great potential applications to long-term preservation of plant viruses.

Project description:Potato wild relatives (Solanum section Petota) are a source of genetic diversity for improving traits in modern cultivars (S. tuberosum) to meet climate challenges. Potatoes are susceptible to multiple abiotic and biotic stresses and have undergone constant improvement through breeding programs worldwide. The allotetraploid S. acaule Bitter has been used to introgress cold tolerance into potato breeding germplasm. The cold challenged transcriptome of S. acaule was compared with that of autotetraploid S. tuberosum cv. Atlantic, and was found to have fewer differentially expressed genes than the latter. Specifically, subgenome 1 has less downregulated alleles compared to subgenome 2 and S. tuberosum.

Project description:Cryopreservation is a key method for long-term storage of biological specimens. The development of optimal cryopreservation condition will reduce the damage from the storage as least as possible. The effect of the cryopreservation condition we used on peripheral blood mononuclear cells (PBMCs) has been previously evaluated with microarray analysis. The emerging single-cell RNA sequencing (scRNA-seq) technology enable deeper exploring cellular heterogeneity and function. In the current study, we further optimized the cryopreservation procedure using FACS analysis, and the method were further evaluated by ScRNA-Seq for two different length of storage time: six months and 12 months in comparison to fresh cells. We identified major immune cell types from both fresh and recovered cryopreserved PBMCs, including monocytes, dendritic cells (DCs), natural Killer (NK) cells, CD4+ T cells, CD8+ T cells, and B cells. The cell viability of all identified immune cell types was relatively stable during the initial 6-month of cryopreservation, while showed10 ~ 40 % decline after cryopreserved for 12 months for different cell types. Furthermore, transcriptome profile of cryopreserved samples did not show obvious perturbation during whole 12 months testing time, although a few key genes involved in stress response or apoptosis, exhibited significant change, especially in monocytes, DC and NK cells. Our results validate that the optimized cryopreservation is a reliable procedure for maintaining these major immune cell types in PBMCs, also highlight the importance of careful assessment of individual sample as variation could be introduced by cryopreservation.
