Expression analysis of the auditory cortex after various behavior treatments

Project description:Auditory cued fear conditioning is a widely used behavioral paradigm to investigate associative learning and memory formation. During conditioning, an animal learns that a particular auditory stimulus predicts an aversive stimulus, a foot shock. When later confronted again with this conditional auditory stimulus rodents typically respond with a cessation of movement. The purpose of this study was to analyze the effects of auditory fear conditioning on gene expression in the auditory cortex in a number of different paradigms that control for the effect of several behavioral factors. The different paradigms were M-^SpairedM-^T were the sound and the shock are paired so that the mouse associates the tone with the noxious stimulus the shock, M-^SsoundM-^T were only the sound was presented, M-^SshockM-^T were only the shock was presented, M-^SunpairedM-^T were the sound and the shock were presented one minute apart so that the mouse cannot form an association between the sound and the shock, M-^ScontextM-^T were no sounds or shocks were presented but the mouse was in the behavior chamber and M-^SHCM-^T were the mouse was taken directly from the cage.

PTSD mouse anterior cingulate cortex proteome

Project description:Posttraumatic stress disorder (PTSD) is a prevalent psychiatric disorder. Several studies have attempted to characterize molecular alterations associated with PTSD, but most findings were limited to the investigation of specific cellular markers in the periphery or defined brain regions. In the current study, we aimed to unravel affected molecular pathways/mechanisms in the fear circuitry associated with PTSD. We interrogated a foot shock induced PTSD mouse model by integrating proteomics and metabolomics profiling data. Alterations at the proteome level were analyzed using in vivo 15N metabolic labeling combined with mass spectrometry in prelimbic cortex (PrL), anterior cingulate cortex (ACC), basolateral amygdala (BLA), central nucleus of amygdala (CeA) and CA1 of hippocampus between shocked and non-shocked (control) mice, with and without fluoxetine treatment.

Molecular diversity of auditory critical period plasticity revealed by single-nucleus RNA sequencing

Project description:Auditory experience drives neural circuit refinement during auditory circuit development, but little is known about the genetic regulation of this developmental process. The primary auditory cortex (A1) exhibits a critical period for thalamocortical connectivity between postnatal days P12 and P15, during which tone exposure alters the tonotopic topography of A1. We hypothesized that a coordinated, multicellular transcriptional program governs this window for patterning of the auditory cortex. To test this idea, we generated a multicellular map of gene expression by performing droplet-based, single-nucleus RNA sequencing (snRNA-seq) of A1 across three developmental time points spanning the tonotopic critical period (P10, P15, P20). We also tone-reared mice (7 kHz pips) during the 3-day critical period and carried out snRNA-seq of A1 at P15 and P20. Using semi-supervised clustering and marker genes, we identified and profiled neuronal (glutamatergic and GABAergic) and non-neuronal (oligodendrocytes, microglia, astrocytes, and endothelial) cell types in A1 under these different conditions to identify candidate genes that might regulate auditory critical period plasticity. By comparing normally reared and tone-reared mice, we identified hundreds of genes in both glutamatergic and GABAergic cells with altered expression as a result of sensory manipulation in the critical period. In addition, we identified previously unknown effects of developmental tone exposure on interneuron developmental trajectories. This single-cell transcriptomic resource of the developing auditory cortex will provide a powerful discovery platform for future characterization of mediators of tonotopic plasticity.

Spatio-molecular gene expression reflects dorsal anterior cingulate cortex structure and function in the human brain [snRNA-seq]

Project description:In the human brain, the dorsal anterior cingulate cortex (dACC) plays a key role in the reward pathway, which is connected to drug addiction, substance abuse, and some psychiatric disorders. Specific cell types in the dACC are associated with these disorders, but the breakdown of cell populations and their relative positions are unknown. Uniquely, the dACC has agranular laminar structure due to a lack of layer IV, which relates to distinct biological functions. We generated a human dACC spatial transcriptomic map by integrating 10x Genomics Visium Spatial Gene Expression data from ten neurotypical donors and combining this map with corresponding 10x Genomics Chromium Single Cell Gene Expression data. We utilized nonnegative matrix factorization (NMF) to discover various gene programs, which shed light on von Economo neurons (VENs) in Layer V of the dACC.To better understand the molecular landscape of this region, we compared it with the dorsolateral prefrontal cortex (dlPFC) region in the same ten donors. The brain’s structure and function are closely connected, so our precise pairing of gene expression to spatial information sheds light on how dysregulation in the dACC relates to psychiatric disorders.
