Project description:gamma delta intraepithelial lymphocytes were isolated from the colons of DSS-treated and untreated mice. Total RNAs were isolated and compared by Affymetrix DNA microarray. Duplicate comparisons were made between DSS-treated and untreated gamma delta intraepithelial lymphocytes.

Project description:Intestinal intraepithelial gamma-delta T cells (iIEL gd T cells) constitute a peculiar subset of T lymphocytes located in the gut epithelial barrier involved in intestinal homeostasis. To date, type 1 and type 17 TCR gamma-delta T cells have been well characterized into two functional categories based on phenotypic and functional markers e.g. CD44 and CD27. However, It remains to be determined how to classify iIEL gamma T cells based on commonly used markers. We addressed this question by comparing the transcriptomes of mouse iIEL gamma-delta T cells with distinct functionally well-described gamma-delta T cells present in the periphery.
Project description:Gamma-delta T cells prevent CRC. To understand the criticial role of Gamma-delta T cells in CRC, AOM-DSS tumors from mice lacking TCRγδ and with TCRγδ cells were used to isolate epithelial cells to study gene expression by bulk RNA sequencing.
Project description:To investigate the detailed molecular mechanisms for the regulatory role of Nik in colitis, microarray gene expression analysis was performed on colon tissue RNA isolated from 3-month-old untreated control and DSS treated Nik+/+ and NikΔIE mice.
Project description:Adamts12-deficient mice undergo more severe colitis than WT mice after induction with DSS. We used microarrays to determine the gene expression differences between Adamts12-deficient and WT mice during ulcerative colitis induced with DSS (dextran sodium sulfate) Fragments of distal colon from DSS-treated (2% DSS during 7 days and 1 day of recovery) and untreated Adamts12-deficient and WT mice were obtained for RNA extraction and hybridiztion with Affymetrix microarrays
Project description:Expression profile of colon mucosa of F344 rats treated with 5% DSS during five days vs colon mucosa of F344 rats pre-treated for 20 days with resveratrol 1mg/kg/day and during the five days of 5% DSS administration Keywords: Inflammation experiment Two conditions experiment, colon mucosa of F344 rats treated 5% DSS for 5d vs colon mucosa of F344 rats pretreated with 1mg/kg/day resv + 5% DSS for 5d. 8 Biological samples for each group
Project description:To discover possible molecular mechanisms mediated by GM-CSF that may be involved in regeneration of the colon epithelium after DSS-induced injury we performed whole-genome wide mRNA microchip analysis of isolated crypt epithelial cells from unchallenged and DSS-challenged WT and GM-CSF-/- mice. Total RNA was prepared from isolated crypts obtained from colons of GM-CSF deficient mice and WT littermates treated or untreated with 1.5%DSS for 6 days. Three biological replicates were performed for each experimental condition.
Project description:Interferon alpha (IFNa) is a pro-inflammatory cytokine that is rapidly upregulated as part of an innate immune response. Gamma delta intraepithelial lymphocytes (gd IEL) mount a rapid antimicrobial response to luminal microorganisms and previous report indicate that the T cell receptor (TCR) of these cells is constantly triggered at steady-state. We assessed the contribution of tonic TCR activation in response to acute, systemic IFNa administration among murine gd IELs. Our data demonstrate that inhibiting basal TCR signaling has a minimal effect on the transcriptional profile of gd IELs and acute IFNa exposure induces antiviral gene programs independent of TCR signaling.
