Project description:Most physiological and molecular mechanisms of salinity stress are researched based on salt shock conditions. However, salt shock doesn’t occur in agricultural practice or natural ecosystem. In the fields, salts accumulate gradually through high salt or sodium irrigation water and poor managements that allow ground water to rise to soil surface. Therefore, it is more reasonable to research salinity stress in a mild way (stepwise salt addition). The objective of this study is to select marker genes to differentiate between salt shock (Phase 0) and salt stress (Phase 1). Three replicates were used for all RNA-Seq experiments conducted on control, Phase 0 and Phase 1 samples in Arabidopsis thaliana. Phase 0 samples (rosette leaves) were harvested 1 hour later from 105 mM NaCl salt shock treatment; Phase 1 samples (rosette leaves) were harvested 2 days later after reaching 90 mM NaCl by a stepwise addition (15 mM NaCl per day).

Project description:The experiment was performed in a commercial sweet cherry (cv. Tsolakeika, Prunus avium L.) orchard in North Greece (Edessa) during 2017 growing season. The orchard contained 10-years old trees, planted at 5x5 m spacing between rows and along the row, grafted onto Mahaleb cherry (Prunus mahaleb L.) rootstock, trained in open vase and subjected to standard cultural practices. Three foliar sprays (0.5% or 35 mM CaCl2) were performed at 15, 27 and 37 days after full blossom (DAFB). Cherry fruits (exocarp plus mesocarp tissues) were sampled in two developmental stages, namely at full red color (44 DAFB, S4 stage) and at commercial harvest (55 DAFB, S5 stage). Three biological replicates of 20-fruit sub-lots in control and Ca-treated fruits were frozen in liquid nitrogen, grinding in fine powder and stored at -80 ⁰C for proteomic processing.

Project description:Parasitoid wasps are one of the most species-rich groups of animals on Earth, due to their ability to successfully develop as parasites of nearly all types of insects. Unlike most known parasitoid wasps that specialize within one or a few host species, Diachasmimorpha longicaudata is a generalist that can survive within multiple genera of tephritid fruit fly hosts, including many globally important pest species. D. longicaudata has therefore been widely released to suppress pest populations as part of biological control efforts in tropical and subtropical agricultural systems. In this study, we investigated the role of a mutualistic poxvirus in shaping the host range of D. longicaudata across three genera of agricultural pest species: two of which are permissive hosts for D. longicaudata parasitism and one that is a nonpermissive host. We found that permissive hosts Ceratitis capitata and Bactrocera dorsalis were highly susceptible to virus infection, displaying rapid virus replication and abundant fly mortality. However, the nonpermissive host Zeugodacus cucurbitae largely overcame virus infection, exhibiting substantially lower mortality and no virus replication. Further investigation of transcriptional dynamics during virus infection demonstrated hindered viral gene expression and limited changes in fly gene expression within the nonpermissive host compared to the permissive species, indicating that the host range of the viral symbiont may dictate the host range of D. longicaudata wasps. These findings also reveal that viral symbiont activity may be a major contributor to the success of D. longicaudata as a generalist parasitoid species and a globally successful biological control agent.

Project description:We have performed a transcriptome analysis of genes at three different ripening stages of the pink-white fruits and the ripe stage of the red fruits of Chinese bayberry. This analysis provided a total of 119,701 unigenes, of which 41.43% were annotated in the Nr database. Our results showed that the formation of the pink-white color in Chinese bayberry fruits depended on the anthocyanin metabolic pathway, regulated by MYB1. Downregulated expression of key anthocyanin biosynthetic pathway genes, such as UFGT, F3’H, and ANS at the late stage of fruits development compared with DK3 fruits resulted in the failure to form red fruits. Our findings shed light on the regulatory mechanisms and metabolic processes that control color development in the fruits of Chinese bayberry.

Project description:Dwarfed stature is a desired trait for modern orchard production systems. One effective strategy of dwarfing is exogenously applying plant growth retardants (PGRs) to plants. However, for many economic fruit trees, the current knowledge on the regulatory mechanisms underlying the dwarfing effect of PGRs were limited, which largely restricts their agricultural application. In this study, we exogenously applied three kinds of PGRs (paclobutrazol, B9 and mannitol) to the seedlings of pomegranate (Punica granatum L.) and performed comparative transcriptome analysis to elucidate the molecular features of PGR-induced dwarfing in pomegranates. Our results showed that all the three PGRs could significantly suppress auxin biosynthetic and metabolic processes, as well as auxin-mediated shoot development, which may be the main reason for the dwarfing. Besides, different PGRs were also found to induce dwarfing via specially mechanisms. Cellular response to strigolactone were downregulated by the application of paclobutrazol, while carbohydrate homeostasis and metabolism were specifically suppressed in conditions of either B9 or mannitol treatments. Furthermore, exogenous PGR application was supposed to causes adverse impacts on the normal physiological process of pomegranate seedlings, which may bring extra burden to stress adaptation of pomegranate plants. These novel findings unveiled the genetic basis underlying the dwarfing in pomegranates.
