The objective of this study was: to evaluate the anti-pancreatic cancer activity of erianin and explore its underlying mechanisms.To further explore the potential molecular mechanisms of erianin in pancreatic cancer, we performed RNA Sequencing analysis to identify DEGs under erianin stimulation in SW1990 cells.

Project description:Erianin has been reported to play key roles in suppressing a wide variety of tumors. Howbeit, the mechanism underlying the erianin in anti-carcinogenesis remains to be fully addressed. Half-maximal inhibitory concentration (IC50) was detected to determine the influence of erianin on melanoma in vitro. Subcutaneous xenografts were established to observe the effect of erianin on the proliferation of melanoma cells in vivo. Transcriptome screening was performed to identify the meaningful target. Molecular docking and western blot were performed to verified the GSK3α-NF-κB cascade reaction. We reported the finding of Erianin as an efficient inhibitor of melanoma. Erianin selectively inhibited the growth of melanoma cells, rather than normal human melanocyte cell line. Specifically, erianin downregulated the cellular GSK3α-NF-κB pathway to induce melanoma cell apoptosis. Collectively, we initially proved that erianin inhibited melanoma effectively, which was caused by inactivation of GSK3α-NF-κB signaling inducing melanoma cells apoptosis. Thus, the study implies that erianin provides a novel therapeutic strategy for melanoma therapy.

Erianin abrogates vasculogenic mimicry through targeting m5C methylase NSUN2

Project description:Purpose: The precise temporal and spatial regulation of N5-methylcytosine (m5C) RNA modification plays essential roles in RNA metabolism. Targeting m5C regulation in cancer cells may be a potential strategy for cancer therapy. Erianin is a natural product isolated from Dendrobium chrysotoxum Lindl. Howbeit, the in‐depth understanding of interaction between erianin and m5C modification remains indistinct. Methods: Natural product library screening was used to explore the effects of natural product monomers on uveal melanoma (UM) cells. Intraocular xenografts model was established to examine the effect of erianin. Immunoprecipitation mass spectrometry (IP-MS) and molecular docking analyses were performed to identify NSUN2 as the target of erianin. m5C-meRIP-seq and m5C-meRIP-qPCR analyses were performed to identify the functional target of NSUN2. Tube formation assay and CD31-PAS double staining were used to detect vasculogenic mimicry (VM) in UM. Results: Herein, we report the discovery of erianin as an effective inhibitor of uveal melanoma. Mechanistically, the targeted inhibition of NSUN2 function by erianin results in a decrease in the m5C modification and expression levels of CHAC1 in UM, thereby curtailing the formation of VM. Conclusions: Collectively, our data suggested that erianin significantly inhibited UM progression in vitro and in vivo. Our study unveils a novel therapeutic strategy for combating UM.

Project description:Anaplastic thyroid carcinoma (ATC) is an aggressive thyroid malignancy characterized by rapid progression, resistance to conventional therapies, and poor patient prognosis. There is an urgent need for innovative therapeutic strategies. Erianin, a natural compound derived from Dendrobium, has demonstrated significant anti-tumor effects in various cancers, yet its role in ATC remains unexplored.In this study, we confirmed the significant anticancer effects of erianin on ATC through both in vitro and in vivo models, including the induction of apoptosis and GSDME-dependent pyroptosis in ATC cells.

ASK1-induces FGF21 expression in the liver

Herein we identify apoptosis signal regulating kinase 1 (ASK1) as an inducer of hepatic FGF21 production. High fat diet (HFD)-fed liver-specific ASK1 overexpressing mice (ASK1+hep) revealed increased hepatic expression and circulating levels of FGF21.

Expression data from inflorescence (unopened flower buds) of Arabidopsis thaliana - comparison between Ler and ask1 mutant

Project description:ARABIDOPSIS SKP1-LIKE1 (ASK1) protein is involved in regulating flower development. We have compared ask1 mutant floral transcriptome with wild-type Ler to identify the role of ASK1-containing E3 ubiquitin ligases in regulating flower transcriptome. In this dataset, we include the expression data obtained from Arabidopsis thaliana Ler and ask1 mutant flower buds. We identified 42 genes and 74 genes that are down-regulated and up-regulated, respectively. Totally 5 samples were analyzed: 3 samples of ask1 and 2 samples of Ler. The average values were compared between ask1 and Ler.

RNA-seq analysis of macrophage NFATc3 or ASK1 effects on metabolic dysfunction-associated steatohepatitis

Project description:To explore the roles of macrophage NFATc3 and ASK1 in metabolic dysfunction-associated steatohepatitis, we generated macrophage-specific Nfatc3 knockout mice (Nfatc3-MKO), macrophage-specific Ask1 knockout mice (Ask1-MKO),macrophage-specific double knockout (MDKO) mice, macrophage-specific Nfatc3 knockin mice (Nfatc3-MKI), and macrophage-specific Ask1 knockin mice (Ask1-MKI). Lyz2-Cre mice were used as control. Eight-week-old male Lyz2-Cre, Nfatc3-MKO, Ask1-MKO, and MDKO mice were treated with methionine- and choline-deficient (MCD) diet for 4 weeks. Subsequently, mouse liver tissues were isolated and subjected to RNA sequencing analysis. In addition, mouse bone marrow-derived macrophages (BMDMs) were isolated from Lyz2-Cre, Nfatc3-MKI, and Ask1-MKI mice and were also subjected to RNA sequencing analysis.
