Project description:Seeds of Pisum sativum L. cv Alaska Early (purchased from the ‘Abundant Life Seed Foundation’, Port Townsend, USA) were equilibrated for approximately 5 weeks in tightly sealed boxes over 300 g-1 non-saturated LiCl (Hay et al. 2008), producing 60% relative humidity, in a temperature-controlled room (20 ± 1 ºC) until their water potential was stable at 12 % MC. Relative humidity was recorded with a Rotronic AWVC-D10 Hygropalm. These equilibrated seeds are referred to as 0 day ‘non-aged controls’. Following equilibration, accelerated ageing was carried out by placing seeds in sealed bottles at 50 °C until viability loss, thereafter referred to as ‘ageing’. Samples were taken at intervals after 8, 12, and 15 days of ageing. At each interval, germination tests were performed on 1 % water agar at 25 °C under warm white fluorescent light (15 micromol m-2 s-1) at a day / night cycle (8/16h). Germination was defined as radical emergence by at least 2 mm. To summarize, we have performed a transcriptional profiling of Pisum sativum seeds comparing 0 day ‘non-aged controls’ with 8, 12 and 15 days of artificially aged seeds.

Project description:We studied potentially amyloidogenic proteins (e.g. protein forming polymers and complexes that are resistant to treatment with ionic detergents) in root nodules formed by two lines of garden pea (P. sativum L.): Sprint-2 (Fix+ phenotype) and Sprint-2Fix- (sym31) (Fix- phenotype) inoculated with the Rhizobium leguminosarum bv. viciae RCAM1026 root nodule bacteria. The Fix+ phenotype is characterized by effective (ability to fix nitrogen) root nodules formation. The Fix- line is a descendant of the Fix+ line and forms ineffective root nodules (unable to fix nitrogen) with undifferentiated bacteroids. We demonstrated the presence of both plant and bacterial proteins in detergent resistant fractions, including previously identified amyloid proteins RopA and RopB of R. leguminosarum and vicilin of P. sativum L.

Project description:Symbiotic legume nodules and lateral roots arise away from the root meristem via dedifferentiation events. While these organs share some morphological and developmental similarities, whether legume nodules are modified lateral roots is an open question. We dissected emerging nodules (EN), mature nodules (MN), emerging lateral roots (ELR) and young lateral roots (YLR), and constructed strand-specific RNAseq libraries using polyA-enriched RNA preparations. Root sections above and below these organs devoid of any lateral organs were used to construct respective control tissue libraries (ABEN, ABMN, ABELR, ABYLR respectively). High sequence quality, predominant mapping to coding sequences, and consistency between replicates indicated that the RNAseq libraries were of very high quality. We identified genes enriched in emerging nodules, mature nodules, emerging lateral roots and young lateral roots in soybean by comparing global gene expression profiles between each of these organs and adjacent root segments. Potential uses for this high quality transcriptome data set include generation of global gene regulatory networks to identify key regulators; metabolic pathway analyses and comparative analysis of key gene families to discover organ-specific biological processes; and identification of organ-specific alternate spliced transcripts. When combined with other similar datasets especially from leguminous plants these analyses can help answer questions on the evolutionary origins of root nodules and relationships between the development of different plant lateral organs.

Project description:Pea (Pisum. sativum L.) is a traditional and important edible legume that can be sorted into grain pea and vegetable pea according to their harvested maturely or not. Vegetable pea by eating the fresh seed is becoming more and more popular in recent years. These two type peas display huge variations of the taste and nutrition, but how seed development and nutrition accumulation of grain pea and vegetable pea and their differences at the molecular level remains poorly understood. To understand the genes and gene networks regulate seed development in grain pea and vegetable pea, high throughput RNA-Seq and bioinformatics analysis were used to compare the transcriptomes of vegetable pea and grain pea developing seed. RNA-Seq generated 18.7 G raw data, which was then de novo assembled into 77,273 unigenes with a mean length of 930 bp. Functional annotation of the unigenes was carried out using the nr, Swiss-Prot, COG, GO and KEGG databases. There were 459 and 801 genes showing differentially expressed between vegetable pea and grain pea at early and late seed maturation phases, respectively. Sugar and starch metabolism related genes were dramatically activated during pea seed development. The up-regulated of starch biosynthesis genes could explain the increment of starch content in grain pea then vegetable pea; while up-regulation of sugar metabolism related genes in vegetable pea then grain pea should participate in sugar accumulation and associated with the increase in sweetness of vegetable pea then grain pea. Furthermore, transcription factors were implicated in the seed development regulation in grain pea and vegetable pea. Thus, our results constitute a foundation in support of future efforts for understanding the underlying mechanism that control pea seed development and also serve as a valuable resource for improved pea breeding.
