Project description:Strigolactone (SL) signalling plays many roles in plants, however, the downstream responsive genes remain unclear. SUPPRESSOR OF MORE AXILLARY GROWTH2-LIKE 7 (SMXL7) is degraded upon SL signalling. Here, we generated Arabidopsis carrying a transgene SMXL7pro:SMXL7d53-GR, where a SL-insensitive version of SMXL7 is tagged with the ligand binding domain of the glucocorticoid receptor (GR). SMXL7d53-GR will be translocated to the nucleus upon dexamethasone (dex), and we profiled the gene expression in mock or dex treated plants and revealed the downstream responsive genes of SL signalling.

Strigolactone and cytokinin act antagonistically in regulating rice mesocotyl elongation in darkness

Project description:Strigolactones have been defined as a new group of phytohormones that regulate shoot branching. Tthe phenotypes of strigolacton-related rice (Oryza sativa) dwarf (d) mutants demonstrated that SLs inhibit mesocotyl elongation by controlling cell division. Moreover, the trans-zeatin, one of active cytokinins, content of mesocotyls was increased in the SL-deficient d10-1 and SL-insensitive d14-1 mutants. To examine if there are genes related to cytokinin-biosynthesis or -degradation among the strigolactone-responsive genes expressed in the mesocotyl, we carried out microarray analyses using d10-1, d14-1 and the wild-type grown under dark conditions, with or without pretreatment of a synthetic strigolactone analog, GR24.

A PIF-SAUR module safeguards hypocotyl elongation from ABA inhibition in the dark

Project description:Driven by cell elongation, hypocotyl growth is tightly controlled by light and responds to both external stimuli and endogenous hormonal pathways. Hypocotyls are responsive to the stress signalling hormone abscisic acid (ABA), which effectively inhibits cell elongation. However, how this regulation is connected to light signalling has been a subject of limited studies. Here, we show that, contrary to light-grown seedlings, hypocotyl elongation of dark-grown seedlings is ABA-insensitive. In the dark, hypocotyl sensitivity to ABA is restored in the constitutive photomorphogenic pifq and cop1-4 mutants, suggesting that an active light signalling pathway is necessary for hypocotyl responsiveness to ABA. Surprisingly, etiolated hypocotyls retain an intact ABA signalling pathway, as could be detected by the induction of ABI1 and RD29B transcripts in response to exogenous ABA, suggesting that inhibition of hypocotyl elongation occurs through a divergent path that can be inhibited by PIFs. Here, using RNA-seq analysis we identified a number of ABA differentially expressed genes (DEGs) that correlate with ABA inhibition of hypocotyl elongation. Among these DEGs a number of SMALL AUXIN-UP RNA (SAUR) genes that are known to directly repress D-CLADE TYPE 2C PROTEIN PHOSPHATASES (PP2C.D) were found. The abrogation of PP2CD.D2-SAUR interaction in the pp2c.d2/PP2C.D2M331K -GFP line, restored ABA sensitivity in the dark, suggesting that SAUR proteins, upregulated by PIFs, are sufficient to render hypocotyls insensitive to ABA. This hypocotyl-specific mechanism enables to promote growth towards the light at all costs, overriding ABA inhibition of cell elongation, to ensure subsequent seedling establishment and survival.

Strigolactone biosynthesis requires the symbiotic GRAS-type transcription factors NSP1 and NSP2

Project description:Legume GRAS-type transcription factors NSP1 and NSP2 are essential for Rhizobium Nod factor-induced nodulation. Both proteins are considered to be Nod factor response factors regulating gene expression upon symbiotic signalling. However, legume NSP1 and NSP2 can be functionally replaced by non-legume orthologs; including rice (Oryza sativa) OsNSP1 and OsNSP2. This shows that both proteins are functionally conserved in higher plants, suggesting an ancient function that was conserved during evolution. Here we show that NSP1 and NSP2 are indispensable for strigolactone biosynthesis in the legume Medicago truncatula as well as rice. Mutant nsp1-nsp2 plants hardly produce strigolactones. The lack of strigolactone biosynthesis coincides with strongly reduced DWARF27 expression in both species. Rice and Medicago represent distinct phylogenetic lineages that split ~150 million years ago. Therefore we conclude that regulation of strigolactone biosynthesis by NSP1 and NSP2 is an ancestral function conserved in higher plants. Since strigolactone biosynthesis is highly regulated by environmental conditions like phosphate starvation, NSP1 and NSP2 will be important tools in future studies on the molecular mechanisms by which environmental sensing is translated into regulation of strigolactone biosynthesis. As NSP1 and NSP2 are single copy genes in legumes, it implies that a single protein complex fulfills a dual regulatory function of different downstream targets; symbiotic and non-symbiotic, respectively.
