Project description:In order to investiage the morphogenetic control of early tube development in the Drosophila melanogaster embryo we performed single-cell-RNAseq-analysis of FACS-isolated early salivary gland placodal cells (stage 10-12), making up less than 0.4% of cells within the embryo to look for early tubulogeneic signatures. In order to identify salivary gland specific signals from epidermal signatures we also performed single-cell-RNAseq-analysis on FACS-isolated epidermal cells in stage 10-12 Drosophila melanogaster embryos for the purpose of differential expression anaylsis.

Project description:The goal of this study was to generate a spatiotemporal atlas of gene expression in the developing salivary gland. This will allow us to identify changes in gene expression that may result in the formation of the different domains of the salivary gland. At least 4 microarrays per location per time point were collected. The originating samples are rather small, so many clefts, ducts and buds had to be pooled to generate enough aRNA for hybridization. Gene expression of murine 3 major salivary gland organoids

Project description:Salivary glands that produce and secret saliva, which is essential for lubrication, digestion, immunity, and oral homeostasis, consist of diverse cells. The long-term maintenance of diverse salivary gland cells in organoids remains problematic. Here, we established long-term murine salivary gland organoids from 3 major salivary glands, including parotid gland (PG), submandibular gland (SMG), and sublingual gland (SLG). Murine salivary gland organoids expressed gland-specific genes and proteins of acinar, myoepithelial, and duct cells. Organoids were maintained in growth media (named GEM) and further underwent differentiation in differentiation media (named DAM). Our study will provide an experimental platform for the exploration of mechanisms involvled in tissue regeneration, development, or several salivary gland diseases.

Generation of functional salivary gland progenitor cells from extended pluripotent stem cells

Project description:The aim of this study was to use pluripotent stem cells (PSCs) as a starting point to progressively differentiate into induced salivary gland progenitor cells (iSGPCs) by mimicking salivary gland development. In this study, we further explored and optimized the passage strategy and organoid formation protocol of iSGPCs based on our previous studies. After 17 days of differentiation, PSC-derived iSGPCs exhibited a marker expression profile similar to that of adult salivary gland progenitors, including KRT5, KRT14, KRT19, and CD49F. Functionally, iSGPCs exhibit self-renewal capacity, are able to form salivary gland-like branching organoids, and have therapeutic potential in models of radiation-induced salivary gland hypofunction.
