Project description:The aim of this study was to analyse the transcriptional regulation in the cornea of rats that overexpress the human NRC32 gene coding for mineralocorticoid receptor (P1 hMR). Six-month-old male Wild-Type (WT) and transgenic P1 hMR rats were euthanized and the corneas were exised for RNA-sequencing. Total RNA samples were sequenced at the iGenSeq transcriptomic platform of the Brain and Spine Institute (ICM, Paris, France). RNA quality was checked by capillary electrophoresis (Agilent 2100 Bioanalyzer system) and RNA with integrity numbers (RIN) ranging from 7.8 to 8.2 was accepted for library generation. Quality of raw data has been evaluated with FastQC. Illumina DRAGEN bio-IT Platform (v3,8,4) was used to align reads on reference genome rd6 and quantification with genecode vM25 annotation gtf. Library orientation, library composition and coverage along transcripts were checked with Picard tools. differential analysis has been also conducted with edgeR. Multiple hypothesis adjusted p-values were calculated with the Benjamini-Hochberg procedure to control FDR. We identified genes differentially regulated in P1 hMR rats as compared to WT rats.

We observed low grade inflammation, RPE abnormatilites and anatomical signs of choroidal neuropathy in these rats. Bulk-transcriptomic analysis of neuroretinas was peformed to identify potential mechanisms.

Project description:Aldosterone, the main mineralocorticoid hormone in Humans, has a major role in maintaining the hemo-electrolytique homeostasis, by acting in the distal part of nephron. This steroid hormone mainly acts through its binding to a ligand-induced transcription factor, the mineralocorticoid receptor (MR). MR binds to specific genomic sequences, where it recruits transcriptional coregulators, to activate or repress target gene transcription. The aim of this work was to access the whole aldosterone-dependand MR target genes, by comparing sequenced data from aldosterone or vehicle-treated samples.
