Project description:Liver fibrosis, marked by excessive extracellular matrix deposition, remains a significant challenge due to its complex and not fully understood mechanisms. This study explores the role of Minichromosome Maintenance Complex Component 7 (MCM7) in the development of liver fibrosis, with a focus on its regulatory network and interactions. We knocked down MCM7 in primary hepatocytes and exposed them to IL-6 to simulate inflammatory conditions. RNA sequencing was then utilized to identify the differentially expressed genes and pathways influenced by MCM7 depletion. The results revealed notable alterations in gene expression, particularly in fibrosis- and inflammation-related pathways. Our study identified MCM7 as a key regulator of IL11 expression through the SHCBP1-RACGAP1-STAT3 signaling pathway, which contributes to the activation of hepatic stellate cells and the advancement of liver fibrosis. These findings underscore the critical role of MCM7 in liver fibrosis by promoting IL11 expression, thereby driving the activation of hepatic stellate cells and the progression of fibrosis. Overall, this research sheds light on a mechanistic pathway where MCM7 plays a pivotal role in liver fibrosis progression through the regulation of IL11 expression under inflammatory conditions.

Project description:Liver fibrosis, characterized by the excessive deposition of extracellular matrix, remains a formidable challenge due to its complex and poorly understood mechanisms. In this study, we aimed to elucidate the role of Minichromosome Maintenance Complex Component 7 (MCM7) and SHC SH2-Domain Binding Protein 1 (SHCBP1) in liver fibrosis, focusing on their regulatory network and interaction. Stable hepatocyte cell lines with knockdown of MCM7 and SHCBP1 were constructed and subsequently stimulated with IL-1β to mimic inflammatory conditions. RNA sequencing was conducted to identify differentially expressed genes and pathways affected by the knockdown. Our analysis revealed significant changes in gene expression profiles, particularly in pathways related to fibrosis and inflammation. We discovered that MCM7 interacts with SHCBP1, which in turn binds to RACGAP1. This interaction activates the SHCBP1-RACGAP1-STAT3 signaling axis, leading to increased IL11 expression. Elevated IL11 levels contribute to the activation of hepatic stellate cells and the progression of liver fibrosis. The findings demonstrate that MCM7 is an important regulatory factor in liver fibrosis and promotes IL11 expression through the SHCBP1-RACGAP1-STAT3 signaling pathway, driving hepatic stellate cell (HSC) activation and fibrosis progression. Overall, our findings highlight a mechanistic pathway where MCM7 and SHCBP1 collaborate to regulate IL11 expression, thereby influencing liver fibrosis progression.

Project description:Cytokines such as TNF-alpha and IL-1beta are known for their contribution to inflammatory processes in liver . In contrast, the cytokine IL-17 has not yet been assigned a role in liver diseases. IL-17 can cooperate with TNF-alpha to induce a synergistic response on several target genes in different cell lines, but no data exist for primary hepatocytes. To enhance our knowledge on the impact of IL-17 alone and combined with TNF-alpha in primary murine hepatocytes a comprehensive microarray study was designed. IL-1beta was included as this cytokine is suggested to act in a similar manner as the combination of TNF-alpha and IL-17, especially with respect to its role in mRNA stabilization. Results: The present microarray analysis demonstrates that primary murine hepatocytes responded to IL-17 stimulation by upregulation of chemokines and genes, which are functionally responsible to increase and sustain inflammation. Cxcl2, Nfkbiz and Zc3h12a were strongly induced, whereas the majority of the genes were only very moderately upregulated. Promoter analysis revealed involvement of NF-kappaB in the activation of many genes. Combined stimulation of TNF-alpha/IL-17 resulted in enhanced induction of gene expression, but significantly synergistic effects could be applied only to a few genes, such as Nfkbiz, Cxcl2, Zc3h12 and Steap4. Comparison of the gene expression profile obtained after stimulation of TNF-alpha/IL-17 versus IL-1 proposed a IL-1beta-like effect of the latter cytokine combination. Moreover, evidence was provided that modulation of mRNA stability may be a major mechanism by which IL-17 regulates gene expression in primary hepatocytes. This assumption was exemplarily proven for Nfkbiz mRNA for the first time in hepatocytes. Our studies also suggest that RNA stability can partially be correlated to the existence of AU rich elements, but further mechanisms like the RNase-activity of the upregulated Zc3h12a have to be considered. We explored the expression of miR-93/106b in leiomyoma and paired myometrium (N=62) from untreated and patients exposed to hormonal therapies (GnRHa, Depo-Provera and oral contraceptives) from African Americans and Caucasians, and their regulatory functions in isolated paired (N=15) leiomyoma and myometrial smooth muscle cells (LSMC and MSMC) and leiomyosarcoma cell line (SKLM-S1). At tissue level leiomyomas expressed significantly lower levels of miR-93 and elevated MCM7 as compared to myometrium with limited racial influence or hormonal exposure on their expression. Assessing the regulatory function of miR-93/106b through doxycycline-inducible lentiviral transduction in microarray analysis, tissue factor (F3) and IL-8 were identified as their possible targets. At tissue level leiomyomas expressed a significantly lower level of F3 and an elevated IL-8 which exhibited an inverse relationship with miR-93, but with limited racial or hormonal influences. Gain-of-function of miR-93/106b in LSMC, MSMC and SKLM-S1 dose-dependently repressed F3 and IL-8 through direct interactions with their respective 3M-bM-^@M-^YUTRs and indirectly through F3 repression inhibited IL8, CTGF and PAI-1 expression, confirmed by using siRNA silencing or factor Vlla (FVIIa) activation of F3, as well as reducing the rate of proliferation, while increasing caspase 3/7 activity. We concluded that differential expression of miR-93/106b and their direct and/or indirect regulatory functions on F3, IL-8, CTGF and PAI-1 expression, with key roles in inflammation and tissue turnover may be of significance in the outcome of leiomyoma growth and associated symptoms. Total RNA isolated from TF324 cells transfected with DOX-inducible lentiviral construct carrying miR-106b~25 cluster with and without Dox treatments for 6 days was subjected to gene expression profiling using Sentirx Beadchip Array HumanHT-12_v4.
