KMT5A-Mediated Methylation of IRF3 Promotes Tumor Progression through Immune Suppression

Project description:Lysine methylation is a critical post-translational modification (PTM) involved in diverse physiological and pathological processes. Interferon regulatory factor 3 (IRF3) plays a pivotal role in antitumor immunity; however, the regulatory mechanisms and functional impact of IRF3 methylation within the tumor microenvironment remain incompletely understood. This study demonstrates that monomethylation of IRF3 at lysine 193 (K193) suppresses its phosphorylation-dependent activation. Mass spectrometry-based protein interactome analysis identified lysine methyltransferase 5A (KMT5A) as the key enzyme responsible for IRF3 K193 monomethylation. In colorectal cancer (CRC), aberrantly high expression of KMT5A impaired in vivo antitumor immune responses. Mechanistically, KMT5A catalyzes IRF3 monomethylation at K193, which impedes IRF3 phosphorylation and subsequent activation, thereby suppressing the production of type I interferons (IFN-I). Collectively, these findings elucidate KMT5A-mediated IRF3 K193 methylation as a critical regulatory axis promoting tumor immune evasion and progression. Furthermore, IRF3 K193 methylation represents a promising therapeutic target for CRC intervention.

Transcriptomic analysis of KMT5A-knockout healthy volunteer peripheral blood CD8+ T cells under activated conditions

Project description:This study aims to investigate the role of Lysine Methyltransferase 5A (KMT5A, also known as SETD8) in CD8+ T cell biology. CD8+ T cells were isolated from healthy volunteer PBMCs using magnetic bead-based negative selection. Isolated cells were subjected to CRISPR-Cas9 mediated gene editing targeting KMT5A (KMT5A-KO group) or a non-targeting control guide RNA (control group, CTRL) via ribonucleoprotein (RNP) complex electroporation. Cells were harvested at 72 hours post-electroporation for RNA extraction. RNA sequencing (RNA-seq) libraries were constructed using the Illumina TruSeq Stranded mRNA kit and sequenced on an Illumina platform to generate paired-end reads. Transcriptome profiles of KMT5A-KO CD8+ T cells were compared to those of CTRL cells to identify differentially expressed genes (DEGs) and elucidate potential pathways regulated by KMT5A in this cell type.

EGFR and EGFRvIII coopt host defense pathways, promoting progression in glioblastoma

Project description:Co-amplification of EGFR and EGFRvIII, a tumor-specific truncation mutant of EGFR, represent hallmark genetic lesions in glioblastoma. We report that EGFR and EGFRvIII stimulate the innate immune defense receptor Toll-like Receptor 2 (TLR2); and that knockdown of TLR2 led to a dramatic survival advantage in glioblastoma xenografts. EGFR and EGFRvIII activated TLR2 in a ligand-independent manner, promoting tumor growth and immune evasion. We show that EGFR and EGFRvIII cooperate to activate the Rho-associated protein kinase ROCK2, modulating malignant progression both by activating TLR2 and WNT signaling, and through remodeling the tumor microenvironment.

EGF-Mediated Transcriptional Coregulation of EpICD-LEF1 Alters Nanomechanical Properties of Endometrial Cancer Cells for Promoting Cell Migration

Project description:Epithelial cell adhesion molecule (EpCAM), a membrane protein known to modulate cell-cell adhesion, is also a signaling molecule internalized into the nucleus for transcriptional regulation. Here we demonstrate that activated EGF/EGFR is a signaling factor to drive the proteolysis of EpCAM. Cleavage of the extracellular fragment EpEX results in topographic fading of cell-surface EpCAM detected by antibody-conjugated cantilevers of atomic force microscope (AFM). As a result, internalization of the cytoplasmic domain EpICD forms a transcription factor complex with LEF1 that regulates gene transcription for enhanced cell-mobility functions. Comprehensive probing of cell surface using AFM tip (without antibody) reveals increased elasticity and non-stickiness of these cells, promoting epithelial to mesenchymal transition. While EpCAM cleavage may contribute to the loss of cell-surface adhesiveness, its internalized EpICD additionally regulates targets for promoting cell migration. Thus, this EGF/EGFR-modulated action on structural EpCAM and regulatory EpICD can enhance invasion potential of transformed cells. RL95-2 were stimulated with EGF for 0,12,24,and 48 hr.Immunoprecipitation was carried out using antibodies against EpCAM and Lef-1, sequenced by Illumina HiSeq 2000
