Project description:Here, we report the impact of the dose of the MHC-II restricted neoantigens on the phenotypic and the effector function of tumor-specific CD4 T cells. Whereas vaccines containing low doses of MHC-II-restricted neoantigen (LDVax) generated helper CD4 T cells that promote the proliferation and the cytotoxic functions of CD8 T cells, vaccines containing high doses of MHC-II restricted neoantigen (HDVax) elicited type 1 regulatory T cells that suppress the antitumor immunity.

Project description:Cold tumors are resistant to immune checkpoint inhibitors (ICIs) due to their poor T cell infiltration and immunosuppressive tumor microenvironment (TME). While the role of CD4+ T cell response in anti-tumor immunity has been emphasized, it remains unclear whether the MHC II-restricted neoantigen vaccines targeting CD4+ T cell can reverse the immune microenvironment and overcome resistance to ICIs. Here we observed an enrichment of immune inflammation-related signaling pathways in TME following administration of MHC II-restricted neoantigen vaccines, indicating their potent role in reversing immunosuppressive TME. An increase in the tumor infiltration of CD4+ T cells and CD8+ T cells, an enhancement of their effector functions, and a tendency towards exhaustion were observed, providing prerequisites for the effectiveness of ICI treatment. Furthermore, NicheNet analysis indicated an enrichment of the inhibitory immune checkpoint signaling axis PVR-TIGIT following vaccination. The combination of the vaccines and TIGIT blockade exhibited synergistic anti-tumor efficacy. Mechanistically, the combination therapy promoted the differentiation of CD8+ T cells into effector memory phenotypes while delaying their exhaustion. Our study demonstrates that MHC II-restricted neoantigen vaccines can remodel the inhibitory TME with insufficient T cell infiltration and synergistically inhibit tumor growth with TIGIT antibody, providing a novel strategy for treating cold tumors.

Project description:We investigated transcriptional changes in CD4CD8aa and CD4 intraepthelial lymphocytes. TCRαβ thymocytes differentiate to either CD8αβ cytotoxic T lymphocytes or CD4 T helper cells. This functional dichotomy is controlled by key transcription factors, including the T helper master regulator, ThPOK, which suppresses the cytolytic-program in MHC class II restricted CD4 thymocytes. ThPOK continues to repress CD8-lineage genes in mature CD4 T cells, even as they differentiate to T helper effector subsets. Surprisingly, we show here that the T helper-fate is not fixed and that mature antigen-stimulated CD4 T cells can switch off Thpok expression and reactivate CD8-lineage genes. This unexpected plasticity results in the post-thymic termination of the T helper program and the functional differentiation of distinct MHC class II restricted CD4 cytotoxic T lymphocytes.

Project description:The immune response against tuberculosis relies, at least in part, on CD4+ T cells. Protective vaccines require the induction of antigen-specific CD4+ T cells via mycobacterial peptides presented by MHC class-II in infected macrophages. We have purified MHC class-I and MHC-II peptides and analysed them by mass spectrometry. We have successfully identified 97 mycobacterial peptides presented by MHC-II and 54 presented by MHC-I, from 76 and 41 antigens, respectively. The sequences of selected peptides were confirmed by spectral match validation and immunogenicity evaluated by IFN-gamma ELISpot against peripheral blood mononuclear cells from volunteers vaccinated with BCG, M.tb latently infected subjects or patients with tuberculosis disease. Three antigens were expressed in viral vectors, and evaluated as vaccine candidates alone or in combination in a murine aerosol M.tb challenge model. When delivered in combination, the three candidate vaccines conferred significant protection in the lungs and spleen compared with BCG alone, demonstrating proof-of-concept for this unbiased approach to identifying novel candidate antigens.

Project description:Aberrant O-glycosylation is a hallmark of cancer, but its contribution to immune evasion is unclear. Using CRISPR–Cas9 to delete GalNAc-transferases, particularly Galnt7, in tumor cells, we reduced mucin-type O-glycan truncation. This remodeling activated dendritic cell– and T cell–dependent immunity, eradicating established tumors, preventing metastasis, and generating durable memory. It enhanced clearance of MHC class I–positive tumors via coordinated CD8⁺ and CD4⁺ T cell responses, and uniquely enabled CD4⁺ T cell–mediated elimination of MHC class I–deficient tumors, which evade cytotoxic T lymphocyte surveillance. Tumor cells with reduced O-glycan truncation served as potent whole-cell vaccines, with efficacy further improved by radiotherapy. These findings establish glycosylation as a central regulator of tumor immune evasion and define a broadly applicable vaccine platform that bypasses the limitations of neoantigen-based approaches for low-immunogenicity cancers.

Two sample set of CD4CD8aa and CD4 intraepthelial lymphocytes (IEL) from small intestine of RAG knockout mice ( 8 weeks after transfer of naive CD4 cells, adoptive tranfer model of colitis), were prepared via cell sorting, and RNA was prepared by TRIZol (Invitrogen, USA) . Data were analyzed in GeneSpring GX10. For microarray analysis, RNA was labeled and hybridized to GeneChip Mouse Genome 430 2.0 arrays according to the Affymetrix protocols. Data were analyzed in GeneSpring GX10.

Project description:No currently licensed vaccine reliably prevents pulmonary tuberculosis, a leading cause of infectious disease mortality with a growing rate of extensive drug resistance. Developing effective new vaccines will likely require identifying Mycobacterium tuberculosis (Mtb)-derived antigens presented on MHC class II (MHC-II) by infected phagocytes that CD4+ T cells can recognize to mediate protective immunity. To prime T cells capable of recognizing infected cells, vaccines will also need to deliver antigens to appropriate processing pathways. Here, we use mass spectrometry (MS) to identify 27 Mtb-derived MHC-II peptides presented by infected human dendritic cells, derived from 13 source proteins. Using targeted quantitative MS, we assess and optimize the ability of vaccines to drive presentation of MHC-II epitopes that match those presented by Mtb-infected cells, including the existing Bacillus Calmette Guerin (BCG) vaccine and new mRNA-encoded immunogens. Our results identify potential antigenic targets and immunogen design principles for tuberculosis vaccine development.

Project description:Mouse thymocytes can be classified into four major subsets based on expression of CD4 and CD8 co-receptors. CD4-CD8- (double negative, DN) cells become CD4+CD8+ (double positive, DP) cells following productive T cell receptor (TCR) beta chain rearrangement. A small proportion of DP cells are selected through interaction of clonal TCRalpha/beta and MHC self peptide complex expressed on thymic stromal cells. DP cell expressing MHC class I-restricted TCR become CD4-CD8+ cells, which will finally differentiate into cytotoxic T cells, while MHC class II restricted selection generates CD4+CD8- helper lineage T cells. We used microarrays to identify genes important for thymocyte differentiation and lineage determination by profiling gene expression in different thymocyte subsets.

Project description:CD4 T cells are key for priming and regulating immune recognition of infected and cancer cells, but predictions of class II epitopes have limited accuracy. We profiled over 100’000 HLA-II ligands by Mass Spectrometry and developed a novel motif deconvolution algorithm to analyze these data. Our work demonstrates substantial improvements in the definition of HLA-II binding motifs and enhanced accuracy in HLA-II ligand and class II epitope predictions.
