Project description:Myogenesis in amniotes unfolds through two consecutive waves. The primary myotube lineage is characterized by the expression of slow myosin, sometimes in combination with fast myosin, and may serve as a scaffold for the secondary lineage, which expresses exclusively fast myosin. The embryonic origin of these two lineages, their relationship, and their connection to adult muscle stem cells are unknown. Here, we employed innovative strategies, combining novel TCF-LEF/β-catenin signaling reporters with the precise spatiotemporal control of in vivo electroporation in avian embryos, to track limb muscle progenitors from early migration to late fetal stages. Strikingly, we uncovered two distinct progenitor populations co-existing from the earliest stages of limb myogenesis, with specific developmental fates: reporter-positive progenitors exclusively form primary myotubes, while reporter-negative progenitors generate secondary myotubes and adult muscle stem cells. Furthermore, we uncovered a novel function of TCF-LEF/β-catenin signaling in regulating the spatial organization of the primary myotube lineage via CXCR4-mediated control of myoblast migration, likely contributing to its proposed organizing function. By redefining the embryonic origins of these myogenic populations, our findings not only resolve a longstanding question in muscle biology but also provide a crucial molecular entry point for understanding the cellular and molecular underpinnings of muscle fiber type diversity and function.

Early lineage restriction and regional segregation during mammalian heart development

Project description:Cardiac development arises from two sources of mesoderm progenitors, the first (FHF) and the second heart field (SHF). Mesp1 has been proposed to mark the most primitive multipotent cardiac progenitors common for both heart fields. Here, using clonal analysis of the earliest prospective cardiovascular progenitors in a temporally controlled manner during the early gastrulation, we found that Mesp1 progenitors consist of two temporally distinct pools of progenitors restricted to either the FHF or the SHF. FHF progenitors were unipotent, while SHF progenitors, were either uni- or bipotent. Microarray and single cell RT-PCR analysis of Mesp1 progenitors revealed the existence of molecularly distinct populations of Mesp1 progenitors, consistent with their lineage and regional contribution. Altogether, these results provide evidence that heart development arises from distinct populations of unipotent and bipotent cardiac progenitors that independently express Mesp1 at different time points during their specification, revealing that the regional segregation and lineage restriction of cardiac progenitors occurs very early during gastrulation. We used microarrays to characterize the molecular mechanisms that control Mesp1 progenitor specification and lineage segregation during the early stage of cardiac mesoderm formation, 50 Mesp1 H2B-GFP+ or Mesp1 H2B-GFP- cells at E6.5 or E7.5 from mouse embryos were sorted for RNA extraction, amplification and hybridization on Affimetrix microarrays. Microaarrays were performed on Mouse Genome 430 PM strip Affymetrix array. The overall design was repeated in two different biological samples.

Project description:The mammary gland (MG) is composed of basal cells (BCs) and luminal cells (LCs). While it is generally believed that MG arises from embryonic multipotent progenitors (EMPs), it remains unclear when lineage restriction occurs and what are the mechanisms responsible for the switch from multipotency to unipotency during MG morphogenesis. Here, we performed multicolor lineage tracing and assessed the fate of single embryonic progenitors during mouse MG development. We demonstrated the existence of a developmental switch from multipotency to unipotency during embryonic MG development. Molecular profiling and single cell RNA-seq revealed the hybrid gene expression of EMPs, and the gene trajectory and lineage segregation occurring during MG development. In situ characterization showed that one of the earliest signs of lineage segregation consists in the restricted expression of p63 in the future BCs. Sustained p63 expression during MG development promotes unipotent BC fate in EMPs. Altogether, this study identifies the timing and the mechanisms mediating the switch from multipotency to unipotency during MG development.
