Project description:Global liver gene expression in mouse treated with either chow diet or high-fat diet was compared. Results provide insight into mechanisms underlying effects of high-fat diet on gene expression in mouse liver. Male C57/BJ mice aged 6 weeks were randomly assigned to a high-fat diet or control chow diet. All mice were sacrificed under anesthesia 20 weeks after treatment and livers were harvested. 3-4 total RNA samples per group were analyzed and gene expression was compared between groups.

Project description:Systemic acute inflammatory signals can cause profound anorexia by disrupting the physiological appetite regulation in the hypothalamic milieu. Conversely, obesity related chronic inflammation of the hypothalamus can disturb anorexigenic signals and promote abnormal body weight control. The aim of the present study was to compare the global hypothalamic endophenotype in C57/Bl6 mice exposed to a high-fat diet or with acute illness mediated by LPS. Ten-week old male C57/Bl6 mice (n=18) were randomly divided into four groups; the control 1 group (n =3) was fed a normal diet whereas the high-fat diet (HFD) group (n =6) was fed a high-fat diet for eight weeks. The control 2 group (n=3) received an intraperitoneal injection of saline whereas the LPS group (n=6) received an intraperitoneal injection of LPS. Mice were sacrificed 18-hr post-injection. Both control 2 and LPS groups were fed a normal diet for eight weeks before the injection. The hypothalamic regions were removed and analysed using a 2D LC-MS methodology. The proteomic analysis profiled 9,235 proteins (q<0.05) across all biological states, of which 522 proteins were found modulated in the HFD group and another 579 in the LPS group. The proteomic profiles demonstrated that the systemic acute inflammation linked with anorexia induced a negative feedback loop of appetite control in the hypothalamus, suggesting an effort to re-establish homeostasis. By contrast, the chronic inflammation associated with obesity initiated a “perpetual cycle” of positive feedback enhancement of appetite regulation further exacerbating positive energy balance.

Project description:In this study, human particulated dentin (HPD) was developed. The characteristic of HPD was examinated by XRD, EDX and cytotoxicity test. HPD was used to implant in the calvarial defects of New Zealand White rabbits. bone tissue and blood samples were taken at 3, 5 and 7 days post-implantation for microarray experiment. The gene expression profiles and signaling pathways were monitored using GeneSpring software and Ingenuity Pathways Analysis (IPA), respectively. We used microarray and IPA to elucidate the global programme of gene expression and identified distinct classes of up- and down- regulated genes during HPD treated healing process.

Project description:Pomegranate skin extract could prevent fatty liver due to high fat diet in adult male Sprague Dawley rat. There are 3 groups of rats, feeding chow diet, high fat diet and high fat diet combined with pomegrante skin extract. After 8 weeks feeding, high fat diet group developped fatty liver but the other two groups still have healthy liver. We used microarrays to detail the global programme of gene expression underlying the fatty liver development and preventive effect of pomegranate skin extract on fatty liver. There are 3 treatment groups and 6 replicates for each group. The 3 treatment groups are control group feeding chow diet, high fat group feeding high fat diet and PE group feeding high fat diet and pomegranate skin extract. Rat livers were collecteded for RNA extraction and hybridization on Affymetrix microarrays.

Project description:Dietary protein restriction has been shown to increase energy expenditure and to improve insulin sensitivity in mice, but it is unknown whether humans exhibit similar phenotype to a prolonged eucaloric protein-restricted diet that meet daily protein minimum requirements. We hypothesize that a protein-restricted diet would necessitate an increase in energy intake in order to maintain body weight in healthy, lean men. We designed an overall amino acid diluted diet meeting the requirement for daily protein intake and essential amino acids. Healthy, young, lean men adhered to a protein-restricted, high-carbohydrate diet (LPHC: protein 9E%, 70E% carbohydrate, and 21E% fat) or a protein-restricted, high-fat diet (LPHF: 9E% protein, 50E% fat, and 40E% carbohydrate) for 5 weeks, followed by another 5 weeks on a higher, standard protein diet (HPD: 18E% protein), reflecting their habitual diet. The diets were eucaloric, and energy provision was adjusted to maintain body weight throughout the interventions. In addition, wild type (WT), and FGF21 knockout mice were also fed LPHC, LPHF diets, or a standard higher diet (HPD) for a total of 10 weeks. Our results showed that prolonged eucaloric LPHC and LPHF diets necessitated a daily increase of 20-21% (2.5 MJ) in food intake to maintain body weight compared to pre-intervention in healthy, lean men. Additionally, fasting plasma FGF21 levels increased from 90±125 pg/ml and 78±34 pg/ml to 257±99 pg/ml and 160±52 pg/ml at the end of the LPHC and LPHF, respectively. Furthermore, proteomic analysis revealed adaptations in the respiratory chain in human adipose tissue after 5-week protein-restricted diets. This was found to be dependent on FGF21 in mice, indicating increased energy utilization through alternative UCP1-independent futile cycle pathways likely mediated by FGF21. Moreover, whole-body insulin sensitivity, measured by a hyperinsulinemic-euglycemic clamp, was increased by 16% after the LPHC intervention while maintained after the LPHF intervention, despite the high fat intake. These findings suggest that a protein-restricted diet could serve as a promising approach to prevent weight gain and comorbidities associated with obesity.

Project description:The periodontal ligament tissue plays a crucial role in oral functions by providing structural support to the tooth. Due to the presence of undifferentiated mesenchymal cells, research into its regenerative capacity is ongoing. Pathological conditions can affect these functions and protein composition. The aim of this study was to evaluate through proteomic analysis the protein profile of the periodontal ligament (PDL) tissues of rats submitted to experimental periodontitis induction. The study was approved by the Ethical Committee on Animal Experimentation of the Sao Paulo State University, School of Dentistry at Araraquara, Brazil (protocol number 16/2015) and followed the Animal Research: Reporting of In Vivo Experiments guidelines. A total of eight male adult Holtzman rats, average weight of 300 g, were used and housed in plastic cages properly identified, with a 12-hours lights/dark cycle, under controlled temperature (22-25 °C), receiving water ad libitum and standard laboratory diet (Labina/Purina, Ribeirão Preto, Brazil). The animals were randomly assigned into two groups: control without any intervention (C, n=3) and experimental periodontitis (P, n=5). The animals of the group P were anesthetized with intramuscular injections of 10% ketamine chlorhydrate (0.08 ml/100 g body weight) and 2% xylazine chlorhydrate (0.04 ml/100 g body weight). To induce periodontitis, a cotton thread was placed bilaterally around the upper first molars and maintained with a knot for 20 days. After this period, all animals were euthanized by anesthetic overdose and had their maxillary jaws collected for subsequent analyses.
