Project description:To elucidate the underlying therapeutic mechanisms of Gui-qi-yi-shen granules (GQYS) in the treatment of IgAN, we have employed transcriptome sequencing of 9 mice kidney samples. 155 differentially genes were identified between IgAN and control groups, with 125 upregulated and 30 downregulated. Meanwhile, 403 genes differed between GQYS and IgAN groups, including 171 upregulated and 332 downregulated genes.

RNA-Seq expression analysis of chronic asthmatic mice with Bu-Shen-Yi-Qi formula treatment and prediction of regulated gene targets of anti-airway remodeling

Project description:Airway remodeling is one of the typical pathological characteristics of asthma. While, the structural changes of the airways in asthma are complex, which impedes the development of novel asthma targeted therapy. Our previous study have shown that Bu-Shen-Yi-Qi formula (BSYQF) could ameliorate airway remodeling in chronic asthmatic mice by modulating airway inflammation and oxidative stress in the lung. To further explore the molecular mechanism of BSYQF, in this study we analyzed the lung transcriptome of control, asthma mice with/without BSYQF treatment. Using RNA-sequencing analysis, we found that BSYQF treatment completely or partial reversed a group of genes that induced expression changes in asthma. Additionally, based on previous result we identified 21 differential expression genes (DEGs). Then through inputting 21 DEGs into Ingenuity Pathway Analysis (IPA) database to construct gene network, we inferred Adipoq, SPP1 and TNC which are located at critical nodes in the network, may be key regulators of BSYQF's anti-remodeling effect. Our results provide a preliminary clue to the molecular mechanism of anti-remodeling of BSYQF in asthma.

Project description:Research purpose: To explore the key targets and core signal pathways of XHT (Xinhuitong, XHT) in regulating coronary heart disease with Qi deficiency and blood stasis syndrome through transcriptomics, and reveal the transcriptional regulatory network of XHT in treating coronary heart disease with Qi deficiency and blood stasis syndrome. Research method: Using transcriptomics RNA-Seq technology, gene sequencing of myocardial tissues in the ischemic marginal zone of the Qi deficiency and blood stasis syndrome model group and the high-dose Yiqi Huoxue prescription group and searching for differential genes, from the perspective of gene regulation and expression patterns To study the possible effective drug targets and effect mechanism pathways of XHT in the treatment of coronary heart disease with Qi deficiency and blood stasis syndrome, and to further explore the treatment of coronary heart disease with Qi deficiency and blood by Chinese medicine compound prescriptions. The possible pharmacological mechanism of blood stasis syndrome, and the key molecular nodes and effect mechanism pathways suggested by it are verified by molecular biology methods.

Integrative Pharmacology Powers the Active Components Detection and Mechanism Exploration of Wang Bi Granules Acting on Rheumatoid Arthritis

Project description:Wang Bi Granule (WBG), composed of 16 botanical drugs and 1 animal drug, is clinically used to treat “Wang Bi” syndrome, which mainly refers to later rheumatoid arthritis (RA) in modern medicine, as well as ankylosing spondylitis, tuberculous arthritis, and Kashin-Beck disease characterized by joint pain and swelling deformation. However, its chemical composition and pharmacological mechanisms have not been clarified. Therefore, in the present study we aimed to characterize the chemical components of WBG and explore its mechanism in treating RA using integrative pharmacology, including chemical composition detection, efficacy evaluation, and mechanism exploration. We employed UPLC-QTOF-MS/MS to describe the chemical profile of WBG, TNF-α-stimulated RAW264.7 cells to simulate inflammatory process of RA and evaluate the anti-inflammatory effect of WBG, and network pharmacology to mine the mechanism of WBG acting on RA. As a result, a total of 278 chemical components were identified or tentatively characterized and the water extract of WBG improved the imbalance of inflammation by regulating 179 differential genes in RAW264.7 cells induced by TNF-α. 55 key active constituents were obtained based on the interactions among “components’ targets, RA-related genes, and differential genes (WBG vs TNF-α group)”, which may ameliorate RA conditions by regulating 161 hub genes that mainly involved in inflammation-immune related pathways. The present study, for the first time, employed integrative pharmacology to characterize the chemical profile of WBG and reveal its mechanism against RA through inflammation-immune regulation system.

Network pharmacology and experimental evidence reveal the protective mechanism of Yi-Qi Cong-Ming decoction on age-related hearing loss.

Project description:ContextYi-Qi Cong-Ming (YQCM) decoction has been widely used to prevent age-related hearing loss (ARHL), the most prevalent neurodegenerative disease in the elderly.ObjectiveTo explore the mechanism of YQCM decoction in the treatment of ARHL.Materials and methodsThe chemical constituents of YQCM were screened from the Traditional Chinese Medicine Systems Pharmacology Database. Potential targets of YQCM against ARHL were predicted by DrugBank, GeneCards, and OMIM database. Protein-protein network and enrichment analysis were used for exploring possible molecular mechanisms. Molecular docking and an in vitro model of ARHL by exposing auditory cells with 100 μM H2O2 for 3 h were applied. Cell viability and mitochondrial membrane potential (ΔΨM) were detected by CCK-8 and high-content analysis. γH2AX and cleaved caspase-3 were detected by Western blot.ResultsThe main compounds have good affinities with hub targets, especially AKT1, PTGS2, and CASP3. GO and KEGG analysis showed that the main biological process and key targets were related to negative regulation of the apoptotic process. H2O2 treatment could reduce the cell viability by 68% and impaired ΔΨM, while 90 μg/mL YQCM pre-treatment could restore the cell viability by 97.45% and increase ΔΨM (2-fold higher). YQCM pre-treatment also reduced γH2AX and cleaved caspase-3 protein levels.ConclusionsOur study suggested that YQCM prevents ARHL by modulating the apoptosis process in auditory hair cells. Moreover, this study proved that bioinformatics analysis combined with molecular docking and cell model is a promising method to explore other possible pharmacological interventions of ARHL.
