Laboratory mice engrafted with natural gut microbiota possess a wildling-like phenotype

Project description:Conventional laboratory mice housed under specific pathogen-free (SPF) conditions are the standard model in biomedical research. However, in the past years, many published results involving rodents have been considered irreproducible, raising concerns about the suitability of mice as model organisms. Emerging evidence indicates that variations in SPF microbiota contribute significantly to data variability across different laboratories. Despite efforts to standardize the microbiota, current microbial consortia lack the complexity and resilience needed to replicate interactions in free-living mammals. We present a standardizable and feasible approach for transplanting natural gut microbiota from wildlings into conventional laboratory mice. After engraftment, these TXwildlings adopt a structural and functional wildling-like microbiota and host physiology toward a more mature immune system, with characteristics similar to those of adult humans. We anticipate that using wild mouse-derived microbiota as standard for laboratory mouse models will improve the reproducibility and generalizability of basic and preclinical biomedical research.

RNA-seq Analysis of wildtype (N2) C. elegans at the L4 stage on diets from the laboratory environment and natural environment

Project description:We analyzed RNA-seq data from C. elegans on six bacterial diets, 3 E. coli diets (OP50, HT115, HB101) found in the laboratory setting and 3 found in C. elegans natural environment (Methylobacterium, Xanthomonas, Sphingomonas). We compared gene expression between L4 worms on the OP50 diet to the other 5 bacterial diets. We found that diet influences many gene expression changes, leading to genes being both up regulated and down regulated in a diet-dependent manner.

Project description:Aquatic ecosystems are subjected to a variety of man-induced stressors but also vary spatially and temporally due to variation in natural factors. In such complex environments, it remains difficult for ecotoxicologists to evaluate the effects of contaminants in wild organisms. The aim of the present study was to test the possibility to detect and unravel the effects of different anthropogenic and natural factors on wild fish by the use of a DNA microarray. Transcriptomic profiles from laboratory-exposed and wild fish sampled along a contamination gradient were compared. During laboratory experiments, fish were exposed to pollutants or natural factors under levels that were closed to those found in the sampling sites. A strong difference was observed between the transcriptomic patterns of wild and laboratory-exposed animals, suggesting a general stress induced by captivity in laboratory. Surprisingly, animals acclimatized to cold temperature clustered with fish from the most contaminated sites and shared common genes involved notably in epigenetic mechanisms. A possible energy saving strategy was suggested in cold-acclimated animals. This result may suggest that animals chronically exposed to contaminants develop a general defense/adaptation strategy against pollution through a general shunt in their basal metabolism, whatever the type of contaminants.
