Project description:The gut bacterium Parabacteroides spp. has been increasingly recognized for its therapeutic potential in treating metabolic disorders. However, the role of Parabacteroides johnsonii in metabolic disorders has never been reported. Here, we found that the abundance of P. johnsonii in the feces was negatively correlated with the blood glucose and lipid levels of obese patients. Oral administration of live P. johnsonii improved the metabolic dysfunction in high fat diet (HFD)-fed mice, accompanied by the alleviation of leaky gut and the systemic inflammation. P. johnsonii enhanced the catabolism of branched-chain amino acids (BCAAs) to branched-chain short-chain fatty acids (BSCFAs) in gut. Particularly, the conversion of valine to isobutyrate was correlated to the symptoms of obese patients. Isobutyrate intervention mirrored the favourable effects of P. johnsonii on HFD-fed mice. Isobutyrate increased H3K14 acetylation at Fgf1b promoters and activated its transcription through inhibition of HDAC3 in colon, thereby maintaining the intestinal barrier integrity. The natural product stachyose exhibited its anti-obesity effects by promoting the growth of P. johnsonii. Our findings provided mechanistic insights into the therapeutic potential of P. johnsonii, isobutyrate and stachyose in treating metabolic disorders.

In these experiments, the fluorescence ratio distributions obtained with L. iatae and L. gasseri showed characteristic inter-species profiles. The percentage of conserved L. johnsonii NCC533 genes was about 83% in the L. johnsonii strains comparisons and decreased to 51% and 47% for L. iatae and L. gasseri, respectively. These results confirmed the separate status of L. iatae from L. johnsonii at the level of species, and also that it is closer to L. johnsonii than L. gasseri.

Project description:<p>The gut microbiota plays a crucial role in the onset and progression of inflammatory bowel disease (IBD). β-glucan (BG), a soluble dietary fiber widely present in oats, barley, fungi, yeast and some bacteria, has multiple bioactive properties such as mitigating intestinal inflammation; however, the underlying mechanisms remain unclear. Here, we demonstrate that BG protects against colitis by increasing the abundance of Lactobacillus johnsonii in the gut, which promotes the production of indole-3-lactic acid (ILA) to activate the aryl hydrocarbon receptor (AhR) and increase the secretion of IL-22. Interestingly, L. johnsonii can not utilize BG but requires a cross-feeding interaction with Bacteroides uniformis. B. uniformis-derived nicotinamide (NAM) via the cobB gene promotes the growth of L. johnsonii. Deletion of cobB from B. uniformis results in inefficacy in L. johnsonii growth and fighting against colitis. In support of our findings, we conducted a clinical trial of the BG intervention, which demonstrated that BG increases the abundance of B. uniformis and L. johnsonii in the human gut along with increasing ILA levels. Our findings reveal the mechanism by which BG ameliorates colitis via L. johnsonii-ILA-AhR axis and provide new insights into cross-feeding between bacteria.</p>

Project description:Exposure to dogs in early infancy has been shown to reduce the risk of childhood allergic disease development and dog-ownership is associated with a distinct house dust microbial exposure. Here we demonstrate, using murine models, that exposure of mice toM-BM- dog-associated house dust protects against ovalbumin or cockroach allergen mediated airway pathology. Protected animals exhibited significant reductions in the total number of airway T cells, down-regulation of Th2-related airway responses as well as mucin secretion. Following house dust exposure, the cecal microbiome of protected animals was extensively restructured with significant enrichment of, amongst others, Lactobacillus johnsonii. Supplementation of wild type animals with L. johnsonii protected them against both airway allergen challenge or infection with respiratory syncytial virus. L. johnsonii mediated protection wasM-BM- associated with significant reductions in the total number and proportion of activated CD11c+/CD11b+ and CD11c+/CD8+ cells, as well as significantly reduced airway Th2 cytokine expression. Our results reveal that exposure to dog-associated household dust results in protection against airway allergen challenge and a distinct GI microbiome composition. Moreover the study identifies L. johnsonii as a pivotal species within the gastrointestinal tract capable of influencing adaptive immunity at remote mucosal surfaces in a manner that is protective against a variety of respiratory insults. The G2 PhyloChip microarray platform (commercially available from Second Genome, Inc.) was used to profile cecal gut bacteria from 29 mice: 7 controls, 5 gavaged with dust from homes with pets, 5 gavaged with dust from homes with no pets, 4 CRA-challenged, 4 gavaged with L. johnsonii, and 4 gavaged with L. johnsonii prior to CRA challenge.
