Project description:Streptococcus pyogenes, or Group A Strep (GAS), a significant human pathogen, employs quorum sensing (QS) systems to coordinate its behavior and genetic regulation in order to enhance survival. Our previous research established that one such QS system, the Rgg2/3 system, has the ability to suppress macrophage NFkB activity and production of pro-inflammatory cytokines. Yet, the scope of suppression was undetermined, and the mechanism remained elusive. In this study, we used transcriptomic and phosphoproteomic approaches to elucidate the extent and mechanisms of QS-mediated immune suppression. We found QS-ON GAS broadly suppressed most inflammatory transcriptional pathways including those of NFkB, type I and type II interferon responses, and intracellular stress responses. Yet, we found no alternative transcriptional programs being activated instead. Additionally, phosphoproteomics showed no disruption upstream in typical inflammatory pathways such as those related to NFkB and MAPK activation, which we successfully confirmed. Instead, the proteomic data highlighted a potential role for epigenetic mechanisms of inflammatory regulation. Despite this, measuring DNA methylation and inhibiting important histone and chromatin modifiers did not reveal which if any epigenetic regulation protein might be involved in the mechanisms of suppression. Still, these findings expand our understanding of QS-mediated suppression and of GAS virulence strategies that appear to target irregular means of restricting inflammation. Uncovering this mechanism will offer invaluable insight into GAS, itself, as well as understudied immunological pathways.

Global H-NS counter-silencing by LuxR activates quorum sensing gene expression

Project description:Bacteria coordinate cellular behaviors using a cell-cell communication system termed quorum sensing. In Vibrio harveyi, the master quorum sensing transcriptional factor LuxR directly regulates >100 genes in response to changes in population density. Here, we show that LuxR derepresses quorum sensing loci by competing with H-NS, a global transcriptional repressor that oligomerizes on DNA to form filaments and bridges. We first identified H-NS as a repressor of bioluminescence gene expression, for which LuxR is a required activator. In an hns deletion strain, LuxR is no longer necessary for transcription activation of the bioluminescence genes, suggesting that the primary role of LuxR is to displace H-NS to derepress gene expression. Using RNA-seq and ChIP-seq, we determined that H-NS and LuxR co-regulate and co-occupy 28 promoters driving expression of 63 genes across the genome. ChIP-PCR assays show that as autoinducer concentration increases, LuxR protein accumulates at co-occupied promoters while H-NS protein disperses. LuxR is sufficient to evict H-NS from promoter DNA in vitro, which is dependent on LuxR DNA binding activity. From these findings, we propose a model in which LuxR serves as a counter-silencer at H-NS-repressed quorum sensing loci by disrupting H-NS nucleoprotein complexes that block transcription.

Project description:Yersinia pestis, the etiological agent of plague, is able to sense cell density by quorum sensing. The function of quorum sensing in Y. pestis is not clear. Here, the process of quorum sensing was investigated by comparing transcript profiles when three quorum-sensing synthase genes are knocked out. Two strains, ∆pgm (pigmentation-negative) mutant R88 as treatment and quorum sensing null strain R115 with mutations (∆pgm, ∆ypeIR, ∆yspIR, and ∆luxS) as control, are used in this analysis.

Project description:Quorum sensing is a cell to cell communication process that involves chemical signaling. Yersinia pestis, the agent of plague, has two functional AHL quorum sensing systems Ysp and Ype. For several reasons, it was not clear what effect AHL pathways have on virulence gene expression and survival in the two different hosts, flea and human. To investigate to what effect AHL quorum sensing has on gene expression, we conducted microarray studies comparing Yersinia pestis CO92 (âpgm) to a double AHL mutant strain (âpgm ÎypeIR ÎyspIR) at 37Â°C. Six independent RNA samples from Y. pestis CO92 R114 AHL deficient cultures were paired with six independent RNA samples from control Y. pestis CO92 R88 cultures for hybridization to six two-color microarrays. For three arrays, the control RNA sample was labeled with Alexa 555 dye and the experimental RNA sample was labeled with Alexa 647 dye; the dyes were reversed for the other three arrays to account for any dye bias.
