Project description:In our study, we detected activation of ACKR3 signaling in cardiac lymphatics adjacent to the site of ischemic injury of left anterior descending artery (LAD) ligation. Furthermore, we found that Ackr3∆Lyve1 mice exhibited better survival and were protected from the formation of acute tissue edema after ischemic cardiac injury, in addition to a denser cardiac lymphatic network after LAD ligation, especially in the injured tissues. To characterize the transcriptomic changes in cardiac lymphatic endothelial cell (LEC) and macrophage populations in the heart upon ischemic cardiac injury in Ackr3∆Lyve1 mice, CD68+ macrophages and CD68-/LYVE1+ LECs were immunoprecipitated from the ventricles of Ackr3fl/fl and Ackr3∆Lyve1 mice at baseline or 7 days post LAD ligation. We then performed microarray on the collected lymphatic and macrophage RNA samples.

RNA-sequencing of WT and P2Y4 KO pericardial adipose tissue (PAT) 24 hours after left anterior descending artery (LAD) ligation

Project description:We identified differentially regulated genes in the pericardial adipose tissue of wild type (WT) and P2Y4 KO mice, 24 hours after left anterior descending artery (LAD) ligation as a model of myocardial infarction (MI). Differentially regulated genes were involved in metabolism, brown fat cell differentiation, immune response and inflammation. P2Y4 KO mice displayed adipocyte beiging with increased PD-L1 expression and a higher number of regulatory leukocytes in their PAT 24 hours after LAD ligation. Up-regulated genes linked to adipocyte beiging included Ucp1, Cited1 and Prdm16.

Transcriptome analysis of ACKR3+ and ACKR3- Marginal Zone B cells (MZBs) in mice

Project description:The marginal zone (MZ) of the spleen contains a specialized set of innate-like B cells that are in direct contact with the blood. Here we show that expression of the atypical chemokine receptor 3 (ACKR3) defines two phenotypically, transcriptionally and functionally distinct, equal-sized populations of mouse MZ B cells (MZBs). The ACKR3-expressing population is required for optimal development and function of the MZ and for the differentiation and positioning of all MZBs. In the absence of ACKR3 expression on B cells the MZ is distorted and the MZBs fail to deliver blood-borne antigens to follicular dendritic cells (FDCs), leading to a reduced early humoral immune response. Effective reconstitution of MZ deficient CD19ko mice with either of the two MZB subsets shows that ACKR3- MZBs can differentiate into the more mature ACKR3+ MZBs, but not vice versa. The phenotype of CD19ko mice lacking a MZ is readily rescued by the adoptive transfer of ACKR3-sufficient, and to a much lower degree by ACKR3-deficient follicular B cells (FoBs), indicating that ACKR3 expression is crucial for the establishment of the MZ developmental niche. Similarly, the inability of CD19ko mice to respond to T-independent antigen is rescued when ACKR3-proficient, but not ACKR3-deficient FoBs are adoptively transferred. Accordingly, ACKR3-deficient FoBs are able to reconstitute the MZ if the niche is pre-established by ACKR3 proficient MZBs. Our data uncover a new unexpected role of ACKR3 on B cells for forming the splenic microarchitecture.

Adm and Ackr3 regulate cardiac lymphatic vessel formation during zebrafish heart development and regeneration

Project description:It is well established that lymphatic vessels regulate interstitial fluid homeostasis and facilitate the transport of macromolecules and immune cells. However, the molecular mechanisms underlying cardiac lymphatic vessel formation during heart development and regeneration remain unclear. Single-cell RNA sequencing revealed that, during heart regeneration, ackr3a mutants exhibit altered LEC subcluster compositions and differential expression of several candidate downstream genes. Our findings uncover the heterogeneity of cardiac LECs and demonstrate that the Adm–Ackr3 decoy receptor axis regulates cardiac lymphatic vessel formation during zebrafish heart development and regeneration.

ACKR3 promotes CXCL12/CXCR4-mediated migration of lymphoma cells

Project description:Chemotaxis is an essential physiological process, often harnessed by tumors for metastasis. CXCR4, its ligand CXCL12 and the atypical receptor ACKR3 are overexpressed in many human cancers. Interfering with this axis by ACKR3 deletion impairs lymphoma cell migration towards CXCL12. Here, we propose a model of how ACKR3 controls the migration of the diffused large B-cell lymphoma VAL cells in vitro and in vivo in response to CXCL12. VAL cells expressing full-length ACKR3, but not a truncated version missing the C-terminus, can support the migration of VAL cells lacking ACKR3 (VAL-ko) when allowed to migrate together. This migration of VAL-ko cells is pertussis toxin-sensitive suggesting the involvement of a Gi-protein coupled receptor. RNAseq analysis indicate the expression of chemotaxis-mediating LTB4 receptors in VAL cells. We found that LTB4 acts synergistically with CXCL12 in stimulating the migration of VAL cells. Pharmacologic or genetic inhibition of BLT1R markedly reduces chemotaxis towards CXCL12 suggesting that LTB4 enhances in a contact-independent manner the migration of lymphoma cells. The results unveil a novel mechanism of cell-to-cell-induced migration of lymphoma.

L1CAM overexpression in mouse lung endothelial cells (lECs)

Project description:In an attempt to elucidate the molecular mechanisms underlying the multiple roles of L1 in endothelium, we checked whether manipulating its expression affected the transcriptome of lECs. To this purpose, we compared the gene expression profiles of L1-overexpressing and control lECs by Affymetrix, which revealed a remarkable effect of L1 overexpression on lECs transcriptome. Mouse lung-derived EC (lECs), kindly provided by E. Dejana (Milan, Italy), were stably transfected with a pcDNA3.1/Hygro(-) vector containing mouse LICAM full length or with and empty pcDNA3.1/Hygro(-) vector as control. Three independent experimental replicates were performed.
