Project description:The fifth generation (5G) of mobile communications promotes human exposure to electromagnetic fields exploiting the 3.5 GHz frequency band. We analyzed behaviors, cognitive functions, and gene expression in mice submitted to asymmetrical head exposure to a 5G-modulated 3.5 GHz signal. The exposures were applied for 1 h daily, 5 days per week over a six-week period, at a specific absorption rate (SAR) averaging 0.19 W/kg over the brain. Locomotor activities in an open field, object location, and object recognition memories were assessed repeatedly after four weeks of exposure and did not reveal any significant effect on the locomotion/exploration, anxiety level, or memory processes. mRNA profiling was performed at the end of the exposure period in two symmetrical areas of the right and left cerebral cortex, in which the SAR values were 0.43 and 0.14 W/kg, respectively. We found significant changes in the expression of less than 1% of the expressed genes, with over-representations of genes related to glutamatergic synapses. The right cortical area differed from the left one by an over-representation of responsive genes encoded by the mitochondrial genome. Our data show that repeated head exposures to a 5G-3.5 GHz signal can trigger mild transcriptome alterations without changes in memory capacities or emotional state.

Project description:The potential health risks of exposure to radiofrequency electromagnetic fields from mobile communications technologies have raised societal concerns. Guidelines have been set to protect the population (e.g. non-specific heating above 1 °C under exposure to radiofrequency fields), but questions remain regarding the potential biological effects of non-thermal exposures. With the advent of the fifth generation (5G) of mobile communication, assessing whether exposure to this new signal induces a cellular stress response is one of the mandatory steps on the roadmap for a safe deployment and health risk evaluation. Using the BRET (Bioluminescence Resonance Energy-Transfer) technique, we assessed whether continuous or intermittent (5 min ON/ 10 min OFF) exposure of live human keratinocytes and fibroblasts cells to 5G 3.5 GHz signals at specific absorption rate (SAR) up to 4 W/kg for 24 h impact basal or chemically-induced activity of Heat Shock Factor (HSF), RAt Sarcoma virus (RAS) and Extracellular signal-Regulated Kinases (ERK) kinases, and Promyelocytic Leukemia Protein (PML), that are all molecular pathways involved in environmental cell-stress responses. The main results are (i), a decrease of the HSF1 basal BRET signal when fibroblasts cells were exposed at the lower SARs tested (0.25 and 1 W/kg), but not at the highest one (4 W/kg), and (ii) a slight decrease of As2O3 maximal efficacy to trigger PML SUMOylation when fibroblasts cells, but not keratinocytes, were continuously exposed to the 5G RF-EMF signal. Nevertheless, given the inconsistency of these effects in terms of impacted cell type, effective SAR, exposure mode, and molecular cell stress response, we concluded that our study show no conclusive evidence that molecular effects can arise when skin cells are exposed to the 5G RF-EMF alone or with a chemical stressor.

Testing a 5G Communication System: Kriging-Aided O2I Path Loss Modeling Based on 3.5 GHz Measurement Analysis.

Project description:Deep knowledge of how radio waves behave in a practical wireless channel is required for the effective planning and deployment of radio access networks in outdoor-to-indoor (O2I) environments. Using more than 400 non-line-of-sight (NLOS) radio measurements at 3.5 GHz, this study analyzes and validates a novel O2I measurement-based path loss prediction narrowband model that characterizes and estimates shadowing through Kriging techniques. The prediction results of the developed model are compared with those of the most traditional assumption of slow fading as a random variable: COST231, WINNER+, ITU-R, 3GPP urban microcell O2I models and field measured data. The results showed and guaranteed that the predicted path loss accuracy, expressed in terms of the mean error, standard deviation and root mean square error (RMSE) was significantly better with the proposed model; it considerably decreased the average error for both scenarios under evaluation.

In vivo evaluation of the 94 GHz-chronic exposure effect on Hairless rat skin gene expression [C2]

Project description:Millimeter waves are new broadband frequencies that start to be used in several applications such as future wireless communications, medical, and also non-lethal weapons (the 94-GHz band in Active Denial Systems). However, low information are available on their potential effects on human. These frequencies belong to radiofrequency and they have the property to be stopped by the first layer of the skin. Therefore our studies have aimed to evaluate 94 GHz effects on skin cells whole gene expression. Chronic long term 94 GHz MMW exposures were performed on two rat populations constituted by 17 young animals and 14 adult ones. Each group of animals were equally split in Exposed and Sham exposed subgroup. This two independent exposure experiments were conducted during 5 months, with rats exposed 3 h per day, 3 days per week, to an incident power density of 10 mW/cm², which correspond to twice the ICNIRP limit of exposure for professional. At the end of the experiment, skin explant were taken and RNA were extracted. Then, the modification of the whole gene expression were analyzed with gene expression microarray. Without modification of the animal’s temperature, long term chronic 94 GHz-MMW exposure did not significantly modify the rat’s skin gene expression on both young and adult rats.

Project description:The rapid deployment of the fifth-generation (5G) spectrum by the telecommunication industry is intended to promote better connectivity and data integration among various industries. However, concerns among the public about the safety and health effects of radiofrequency radiations (RFRs) emitted from the newer-generation cell phone frequencies remain, partly due to the lack of robust scientific data. Previously, we used developmental zebrafish to model the bioactivity of 3.5 GHz RFR, a frequency used by 5G-enabled cell phones, in a novel RFR exposure chamber. With RFR exposures from 6 h post-fertilization (hpf) to 48 hpf, we observed that, despite no teratogenic effects, embryos showed subtle hypoactivity in a startle response behavior assay, suggesting abnormal sensorimotor behavior. This study builds upon the previous one by investigating the transcriptomic basis of RFR-associated behavior effects and their persistence into adulthood. Using mRNA sequencing, we found a modest transcriptomic disruption at 48 hpf, with 28 differentially expressed genes. KEGG pathway analysis showed that biochemical pathways related to metabolism were significantly perturbed. Embryos were grown to adulthood, and then a battery of behavioral assays suggested subtle but significant abnormal responses in RFR-exposed fish across the different assays evaluated that suggest potential long-term behavioral effects. Overall, our study suggests the impacts of RFRs on the developing brain, behavior, and the metabolome should be further explored.
