Project description:Water use efficiency has long been considered as an important target for the breeding of improved plant performance under drought. Minimizing leaf transpirational water loss via reduction of stomatal water conductance plays a key contributory role in drought resistance. In this study, we employed both guard cell (GC) targeted and constitutive ectopic overexpression of the Target of Rapamycin (TOR) kinase, a master regulator of multiple signaling networks in transgenic Arabidopsis thaliana, to investigate the impact of these expressed AtTOR transgenes in response to drought and water use efficiency. We performed genome-wide transcriptome analysis employing RNA-seq on the three Arabidopsis genotypes grown on the three water treatments, and further analysis will be used to elucidate the potential mechanism(s) contributing to differences in leaf stomatal physiology between WT and transgenic lines.

Project description:The overall objective was to document transcriptomic changes in the guard cells of Arabidopsis thaliana under short (1x) - and long-term (3x) saline growth conditions. Guard cells of Arabidopsis responded also to the intensity of the salt management by the microarray datasets clearly clustering in (-) salt, 1x salt and 3x salt. Similarly, more GO terms were significantly enriched in differentially expressed guard cell genes of 1 x salt than 3x salt treated plants.

Effect of Abscissic acid on Brassica napus guard cells

Project description:Plants acclimate to drought and water stress through diverse physiological responses, primarily mediated by the hormone abscisic acid (ABA). The closure of stomatal pores on aerial surfaces of plants is one of the rapid responses mediated by ABA to reduce transpirational water loss because plants lose the majority of their water through stomatal pores. Stomatal guard cells have been an important cellular system for studying ABA signaling. The dynamic changes in the transcriptome of stomatal guard cells in response to ABA have been investigated in the model plant Arabidopsis thaliana. In this study, we analyzed the dynamics of ABA-regulated transcriptomes in stomatal guard cells of Brassica napus.
