Project description:Cerebral hemodynamics are crucial for brain homeostasis and serve as a key proxy for brain activity. Although this process involves coordinated interaction between vessels, neurons and glial cells, its dysregulation in neuroinflammation is not well understood. We used in vivo mesoscopic functional ultrasound imaging to monitor cerebral blood volume changes during neuroinflammation in rats injected with lipopolysaccharide (LPS) in the visual cortex, under resting-state or visual stimulation, combined to advanced ex vivo techniques for glial cell reactivity analysis. Cortical neuroinflammation induced large oscillatory hemodynamic traveling waves in the frequency band of vasomotion (~0.1 Hz) in both anesthetized and awake rats. Vasomotor waves traveled through large distances between adjacent penetrating vessels, spanning the entire cortex thickness, and even extending to subcortical areas. Moreover, vasomotion amplitude correlated with microglial morphology changes and was significantly reduced by astrocytic toxins, suggesting that both microglia and astrocytes are involved in the enhancement of vasomotion during neuroinflammation. Notably, functional connectivity was increased under this oscillatory state and functional hyperemia was preserved or even exacerbated. These findings reveal new spatiotemporal properties and cellular mechanisms of cerebral vasomotion, and suggest that this is an important component of brain hemodynamics in pathological states. Moreover, reactive microglia and astrocytes are participating to increased vasomotion during neuroinflammation. These results call for a reassessment of vasomotion and traveling waves as major phenomena when imaging brain hemodynamic activity, particularly in conditions associated with neuroinflammation.

Circulating traveling waves rapidly pace and mature hiPSC-derived cardiomyocytes in self-organized tissue ring

Project description:Directed differentiation methods allow acquisition of high-purity cardiomyocytes (CMs) differentiated from human induced pluripotent stem cells (hiPSCs); however, their immaturity characteristic limits their application for drug screening and regenerative therapy. The rapid electrical pacing of cardiomyocytes have been used for efficiently promoting the maturation of cardiomyocytes, here we describe a simple device in modified culture plate on which hiPSC-derived CMs can form three-dimensional self-organized tissue rings (SOTRs). Using calcium imaging, we show that within the ring, traveling waves (TWs) of action potential spontaneously originated and ran robustly at a frequency up to 4 Hz. After 2 weeks, SOTRs with TW training show matured features including structural organization, increased cardiac-specific gene expression, enhanced Ca2+-handling properties, an increased oxygen-consumption rate, and enhanced contractile force. We subsequently use a mathematical model to interpret the origination, propagation, and long-term behavior of the TWs within the SOTRs. The TW could also potentially be used for pacing the electrical excitable cells such as neuron and retina cells for various applications.

Melanin Nanoparticles Alleviates Thalamic Stroke Damage by Modulating Glial Activation and Neuroinflammation

Project description:Thalamic strokes represent a major cause of post-stroke cognitive impairment, characterized by glial activation and disrupted iron homeostasis. Microglia and astrocytes play pivotal roles in the inflammatory response, but the mechanisms connecting glial activation to ferroptotic cell death remain unclear. This study explores the neuroprotective effects of melanin nanoparticles (MMPP) in thalamic stroke, focusing on glial activation and neuronal ferroptosis. Using a photothrombotic stroke model, MMPP nanoparticles suppress microglial activation, which subsequently mitigates astrocytic reactivity. Transcriptomic analysis reveals that MMPP nanoparticles regulate the MKK4/JNK signaling pathway, linking glial activation to ferroptosis. Reactive astrocytes secrete lipocalin-2 (LCN2), which triggers ferroptosis in neurons through the 24p3 receptor, causing iron accumulation and ROS production. Additionally, MMPP restores key ferroptosis markers by upregulating GPX4 and downregulating PTGS2, interrupting the pathological cascade, preserving neuronal antioxidant defenses, and reducing ferroptotic cell death. These findings highlight MMPP’s dual antioxidant and anti-inflammatory effects, particularly through modulation of the microglia-astrocyte-LCN2 axis. This study suggests that MMPP provides a promising therapeutic strategy to alleviate neuroinflammation and promote functional recovery following ischemic stroke.

Mitochondrial Microprotein MOCCI Controls Neuroinflammation by Altering Glial Activation States [scRNA-seq]

Project description:MOCCI, discovered in a screen for mitochondrial peptides that modify inflammatory outcomes, has emerged as a significant regulator of neuroinflammation. Here, we show that MOCCI is upregulated in microglia and astrocytes in both the mouse and human brain upon neuroinflammation, and is required for orchestrating proper, complete, and beneficial activation of microglia and astrocytes. Induction of MOCCI triggers the transition of glial into a neuroprotective state and promotes the resolution of inflammation. In vitro, MOCCI deficiency leads to reduced migration, phagocytosis and cytokine secretion in glia. In the cuprizone mouse model of Multiple Sclerosis, MOCCI plays a role in both demyelination and remyelination: MOCCI deficiency protects against cuprizone-induced oligodendrocyte death and prevents remyelination by inhibiting the clearance of cellular debris.

Project description:Deposition of amyloid beta (Aβ) and hyperphosphorylated tau along with glial cell-mediated neuroinflammation are prominent pathogenic hallmarks of Alzheimer’s disease (AD). In recent years, impairment of autophagy has been found to be another important feature contributing to AD progression. Therefore, the potential of the autophagy activator spermidine, a small body-endogenous polyamine often used as dietary supplement, was assessed on Aβ pathology and glial cell-mediated neuroinflammation. Oral treatment of the amyloid prone AD-like APPPS1 mice with spermidine reduced neurotoxic soluble Aβ and decreased AD-associated neuroinflammation. A subsequent proteome analysis of isolated microglia confirmed the anti-inflammatory and revealed cytoskeletal effects of spermidine in APPPS1 mice. Our data highlight that the autophagy activator spermidine holds the potential to enhance Aβ degradation and to counteract microglia-mediated neuroinflammation in AD pathology.

Controlling human organoid symmetry breaking reveals signaling gradients drive segmentation clock waves

Project description:Axial development of mammals is a dynamic process involving several coordinated morphogenetic events, including axial elongation, somitogenesis, and neural tube formation. To gain insight into the signals control the dynamics of human axial morphogenesis, we generated hundreds of axially elongating organoids by inducing anteroposterior symmetry breaking of spatially coupled epithelial cysts derived from human pluripotent stem cells. Each organoid was composed of a neural tube flanked by presomitic mesoderm sequentially segmented into somites. Periodic activation of the somite differentiation gene MESP2 coincided in space and time with anteriorly traveling segmentation clock waves in the presomitic mesoderm of the organoids, recapitulating critical aspects of somitogenesis. Through timed perturbations of organoids, we demonstrated that FGF and WNT signaling play distinct roles in axial elongation and somitogenesis and that FGF signaling gradients drive the segmentation clock waves. By generating and perturbing organoids that robustly recapitulate the architecture and dynamics of multiple axial tissues in human embryos, this work offers a means to dissect complex mechanisms underlying human embryogenesis.
