Project description:Probiotics have become promising strategies for treating intestinal barrier dysfunction. However, the mechanisms by which probiotic bacteria interact with host cells to elicit beneficial effects remain unclear. Herein, we demonstrate that administration of E. coli Nissle 1917 (EcN) before Salmonella Typhimurium (STm) infection significantly suppressed STm pathogenesis in a mouse model. To address how bacterial interactions with intestinal epithelium affect STm infectivity, we developed a human gut-on-chip model coupled with computational fluid dynamics simulations, which recapitulated in vivo intestinal barrier protective phenotypes under physiologically relevant flow conditions. Using quantitative proteomics, we discovered that EcN-derived outer membrane protein MipA mediates protection by directly binding to intestinal epithelial integrin, inducing upregulation of integrin-linked kinase (ILK) to strengthen tight junctions and block STm invasion. Notably, expression of MipA in non-pathogenic STm conferred comparable barrier protection, establishing its pivotal role as a barrier enhancer. Our finding uncovers a novel mechanism whereby the bacterial membrane protein acts as a molecular trigger to enhance intestinal epithelial integrity, providing a significant framework for probiotic interventions against enteric infections.

Project description:This study aimed to investigate the systemic immunological effects of kimchi consumption in humans. We conducted a 12-week randomized, double-blind, placebo-controlled clinical trial involving thirteen overweight but otherwise healthy adults who consumed either placebo, spontaneously fermented kimchi (S-K), or starter kimchi fermented with Leuconostoc mesenteroides KCKM0828 (LMS-K). Peripheral blood mononuclear cells (PBMCs) were collected before and after the intervention and subjected to single-cell RNA sequencing (scRNA-seq) to profile immune responses at single-cell resolution.

Project description:This study generated 10x Genomics Visium spatial transcriptomic profiles from mouse liver sections to investigate the effects of high-fat diet and kimchi supplementation on hepatic gene expression organization. Samples were obtained from male C57BL/6J mice assigned to normal diet (ND), high-fat diet (HFD), high-fat diet supplemented with spontaneously fermented kimchi (S-K), and high-fat diet supplemented with starter kimchi fermented with Leuconostoc mesenteroides KCKM0828 (LMS-K). The dataset was designed to assess diet-associated transcriptional changes across the liver lobule and to evaluate how kimchi intervention modulates zonation-related hepatic programs along the portal-to-central axis. Raw FASTQ files were prepared for SRA submission through GEO, and processed outputs include filtered gene-barcode matrices, HDF5 matrices, spatial coordinate files, scale factor files, tissue images, and summary reports.

Project description:Paneth cells are important for maintaining epithelial cell renewal and modulating innate immune function in the intestine through secretion of growth factors and antimicrobial peptides, which help sustain epithelial stem and progenitor cells and contribute to the intestinal barrier and protection against pathogenic bacteria. Here we show that the intestine-enriched miR-802 is a central regulator of intestinal epithelial cell proliferation and Paneth cell function. Genetic ablation of mir-802 in mice leads to amplified ROS generation and Notch/Wnt signaling, increased intestinal epithelial turnover, impaired enterocyte differentiation and nutrient uptake, and increased enterocyte apoptosis. Mice lacking mir-802 in the intestine also exhibit Paneth cell expansion, increased antimicrobial peptide production, and protection against Salmonella infection. This phenotype relies on the miR-802 target gene Tmed9.

Project description:Intraepithelilal lymphocytes (IELs) are located at the intestinal barrier where they can offer swift protection against invading pathogens. However, they are kept in a heightened state of activation resembling effector T cells, but without cytokine production or clonal proliferation. They also posses altered metabolic pathways than CD8+ memory T cells from spleen. With differentially expressed gene analysis of intestinal IELs and CD8+ memory T cells from spleen, we confirmed increased expression of metabolic enzymes involved in lipid uptake and lipid metabolism in IELs compared with CD8+ memory T cells. This was particularly the case for enzymes involved in mevalonate, lanosterol and cholesterol synthesis pathways, suggesting increased lipid metabolite generation. Differential gene expression showed also strong predisposition for cytotoxic potential that IELs possess in comparison to CD8+ memory T cell.

Project description:Acute superior mesenteric vein thrombosis (ASMVT) decreases the expression of junction-associated proteins and the number of intestinal epithelial cells, which leads to intestinal epithelial barrier disruption. Thermoprotein deposition has also recently been identified as an important cause of mucosal barrier defects. However, the role and mechanism of thermoprotein deposition in asmvt is not fully understood. Differentially expressed microRNAs (miRNAs) in the intestinal tissues of ASMVT mice were detected by transcriptome sequencing (RNA-Seq). To identify miRNAs involved in intestinal barrier disruption, we performed RNA-Seq analysis of intestinal tissues from ASMVT mice . Sixty miRNAs were abnormally expressed in normal intestinal tissues compared with ASMVT-induced intestinal tissues.

Project description:The incidence and prevalence of inflammatory bowel disease (IBD) is gradually increasing. A high-fat diet (HFD) is known to disrupt intestinal homeostasis and aggravate IBD, yet the underlying mechanisms remain largely undefined. Here, a positive correlation between dietary fat intake and disease severity in both IBD patients and HFD-fed mice is observed. HFD induces a significant decrease in indole-3-acetic acid (IAA) and lead to intestinal barrier damage. Furthermore, IAA supplementation enhances the intestinal mucin sulfation and effectively alleviates colitis. Mechanistically, IAA upregulates key molecules involved in mucin sulfation, including Papss2 and Slc35b3, the synthesis enzyme and the transferase of 3'-phosphoadenosine-5'-phosphosulfate (PAPS), via the Aryl Hydrocarbon Receptor (AHR). More importantly, AHR can directly bind to the transcription start site of Papss2. Oral administration of Lactobacillus reuteri, which can produce IAA, contributes to protecting against colitis and promoting mucin sulfation, while the modified L. reuteri strain lacking the iaaM gene (LactobacillusΔiaaM) and the ability to produce IAA fails to exhibit such effects. Overall, IAA enhances intestinal mucin sulfation through AHR, contributing to the protection of intestinal homeostasis.

Project description:Group 3 innate lymphoid cells (ILC3s) protect the intestinal barrier by producing IL-22 and IL-17A, yet the molecular mechanisms that sustain their cytokine output during inflammation remain poorly defined. Here, we identify the cytosolic scaffold DAB2IP as a critical ILC3-intrinsic regulator of effector function. In human IBD, DAB2IP expression is reduced in inflamed tissues, correlating with impaired epithelial repair and decreased IL-22. In mice, DAB2IP expression in ILC3s declines during intestinal inflammation, and silencing DAB2IP markedly diminishes IL-22 and IL-17A production. Adoptive transfer of Dap2ip-deficient ILC3s exacerbates DSS-induced colitis and increases susceptibility to Citrobacter rodentium. Mechanistically, DAB2IP promotes NF-κB activation, sustaining transcription of Il22 and Il17a. These results reveal a context-dependent role for DAB2IP as a positive regulator of NF-κB in ILC3s, highlighting its previously unknown function in mucosal immunity and epithelial repair, and suggesting that restoring DAB2IP signaling could enhance barrier protection during intestinal inflammation.
