Project description:To investigate the role of the VCX gene family in cancer cells at the transcriptomic level, we selected VCX gene family-positive A375 (melanoma) and PC9 (lung adenocarcinoma) cell lines. Stable cell lines with lentiviral shRNA targeting the conserved sequences shared by all VCX family members were generated, and cells treated with non-targeting shRNA were used as controls. Total RNA was extracted from cells at the logarithmic growth phase for RNA-seq library construction and sequencing. Differential expression genes and pathway analysis were performed to assess the transcriptomic alterations following VCX gene family knockdown. This dataset provides insights into the potential role of the VCX gene family in multiple cancer types.

RNA Sequencing Analysis of NCAPD2 Knockdown in Lung Adenocarcinoma Cell Line PC9

Project description:This study investigates the effect of NCAPD2 knockdown on lung adenocarcinoma (LUAD) by performing RNA sequencing on PC9 cell lines. The experiment included three control PC9 cells and three PC9 cells in which NCAPD2 was knocked down using shRNA. This RNA sequencing experiment aims to elucidate the role of NCAPD2 in regulating gene expression associated with tumor proliferation, migration, and survival in LUAD. Analysis of differential gene expression provides insights into the pathways affected by NCAPD2 knockdown, with an emphasis on signaling pathways critical to LUAD progression, such as the PI3K/AKT pathway.

Transcriptome-wide map of m6A RNAs in A375 NC cells and shMETTL3 cells

Project description:The newly discovered dynamic N6-methyladenosine (m6A) modification plays a critical role in gene expression from a post-transcriptional level. we profiled the transcriptome-wide m6A modification in mRNAs and lncRNAs in non-targeting control A375 cells(NC) and METTL3 knockdown A375 cells(shMETTL3). Methylated RNA immunoprecipitation sequencing results revealed that the RNA m6A modification is conserved. METTL3 knockdown altered the expression of RNAs m6A modified sites in A375 cells. Finally, we show that the transcriptome-wide m6A alterations occurring in mRNAs and lncRNAs following METTL3 knockdown suggest this process plays important regulatory roles during A375 growth. This study provides a framework for applying the m6A modification regulated by METTL3 to melanoma research.
