Project description:NR1D1 (also known as Rev-erbα) is a nuclear receptor involved in circadian rhythm and metabolic regulation. Its role in vascular biology is not fully understood. We performed CUT&Tag (Cleavage Under Targets and Tagmentation) to investigate the genome-wide binding profile of the transcription factor NR1D1 in primary vascular smooth muscle cells (VSMCs) isolated from mice. Two experimental conditions were included: cells treated with vasostatin-2 and untreated controls. The goal of this study was to identify potential transcriptional regulatory targets of NR1D1 under basal and vasostatin-2-stimulated conditions. CUT&Tag was carried out using a specific antibody against NR1D1, and sequencing was performed on the Illumina NovaSeq 6000 platform.

Project description:This study employs CUT&Tag technology to meticulously map the chromatin-binding landscape of the transcription factor Arid3a in Vascular Smooth Muscle Cells (VSMCs), thereby highlighting its crucial function in the intricate regulation of gene expression. The CUT&Tag assay illuminates Arid3a's presence at key promoter regions, exerting a profound impact on transcriptional modulation and suggesting the epigenetic governance of 260 genes. By integrating RNA-seq with sophisticated bioinformatic analyses, our research elucidates Arid3a's integral role in fundamental cellular activities, particularly in the dynamics of actin filaments, as well as its potential regulatory nexus with critical pathways, including cAMP signaling and calcium signaling. These insights unveil the intricate molecular choreography of Arid3a in the functioning of VSMCs and shed new light on its role in the development of abdominal aortic aneurysm (AAA).

Project description:Crotonylation of histones is discovered of late as one of post-translational modification that can regulate gene expression. However, the function of crotonylation on non-histone proteins in vascular smooth muscle cells (VSMC) is unclear. Here, we aim to use modification and proteomic analysis to find the cellular characteristic of crotonylated non-histone proteins and the crosstalk with ubiquitinated proteins in vascular smooth muscle cell (VSMC) phenotypic remodeling. We performed modification and proteomic analysis of VSMCs before and after stimulated with platelet-derived growth factor-BB (PDGF-BB). The crotonylated and ubiquitinated pan-antibody was used to enrich the protein and then subjected to high-throughput mass spectrometry analysis. The enrichment analysis was performed within differentially modified proteins in regards to GO terms, KEGG and protein domain.

Gene expression profiling using microarray in rat aortic vascular smooth muscle cells in response to Angiotensin II stimulation

Project description:Increased level of the hormone angiotensin II (AngII) contributes to the development of several cardiovascular issues, including atherosclerosis, vascular remodeling, and hypertension by altering gene expression. In our study, we use microarray analysis to investigate the effect of AngII on primary vascular smooth muscle cells derived from rat thoracic aorta. We have identified genes upregulated in response to AngII and are investigating their contribution to these cardiovascular conditions.
