Project description:This study investigates the effects of hyperbaric oxygen (HBO) on cellular senescence and aging-related phenotypes. In naturally aged mice, HBO treatment reduced systemic senescence burden and reversed aging-related transcriptomic signatures in skin, liver, and kidney tissues. Additionally, single-session HBO exposure was shown to preferentially suppress SASP propagation in human stromal cells. This dataset includes transcriptomic profiles of skin, liver, and kidney tissues from young, aged, and HBO-treated aged mice, as well as human primary prostate stromal cells (PSC27).

Hyperbaric Oxygen Promotes Both the Proliferation and Chemosensitization of Glioblastoma Cells by Inhibiting HIF1α/HIF2α-ABCG2

Project description:Hyperbaric oxygen enhances glioma chemosensitivity, but the mechanism remains unclear. Hypoxia is common in gliomas, and as the main effector molecules of hypoxia, HIF1α and HIF2α promote the malignant progression by inhibiting cell apoptosis and maintaining stemness. ABCG2 is a marker protein of tumor stem cells and drug efflux transporter protein. This study aims to reveal the detailed mechanism of hyperbaric oxygen promote both proliferation and chemosensitization. Under hyperbaric oxygen and hypoxic conditions, we investigated the differences in cell cycle, proliferation, apoptosis, LDH release, and the expression of proteins and mRNA. We also conducted studies on transcriptional regulation and performed in vivo experiments. It revealed that under hypoxic conditions, HIF1α, HIF2α, and ABCG2 are highly expressed, and both HIF1α and HIF2α promote ABCG2 expression. After hyperbaric oxygen treatment, the expression of HIF1α, HIF2α, and ABCG2 decreased, both cell proliferation and chemosensitivity increased. After knocking out HIF1α and HIF2α, cell proliferation and chemosensitivity increased, but the expression of stem cell marker proteins decreased. ChIP‒qPCR revealed that HIF1α and HIF2α target the ABCG2 promoter. Gain- and loss-of-function experiments suggested that ABCG2 can promote the expression of stem cell marker proteins, inhibit cell apoptosis, and promote tumor progression. This study confirmed that hyperbaric oxygen can inhibit ABCG2 expression through HIF1α and HIF2α, thereby promoting the proliferation and chemosensitization of gliomas.

Project description:RATIONALE: Radiation therapy can cause long-term adverse effects. Hyperbaric oxygen therapy may be effective in lessening gastrointestinal symptoms caused by radiation therapy given for pelvic cancer. It is not yet known whether high-pressure oxygen is effective in treating adverse effects caused by radiation therapy. PURPOSE: This randomized phase III trial is studying hyperbaric oxygen therapy to see how well it works in treating long-term gastrointestinal adverse effects caused by radiation therapy in patients with pelvic cancer.

Project description:Senescent is an irreversible form of cell cycle arrest initiated by damaged cell constituents and subsequent pro-oncogenic signaling. Replicative senescence in vitro can be considered a model for human aging. When fibroblasts are cultured under atmospheric oxygen conditions of 20%, typical of normal tissue culture procedure, fibroblasts generally reach their replicative capacity at 50-60 population doublings (PDs). When fibroblasts are cultured under normal physiological oxygen conditions of 3%, PDs increase about 30% relative to atmospheric levels. Hence while oxygen is a requirement for normal aerobic respiration, it can contribute to the total amount of oxidative stress to which cells are exposed to, leading to a long-term adverse effect in vitro. Inasmuch, cultures maintained under hyperoxic and hypoxic conditions provide a convenient model system for assessing the relationship between oxygen/oxidative stress and senescence. We used microarrays to profile the changes in global gene expression during aging and senescence of Imr90 cells under growth oxygen conditions of 3% and 20%. Imr90 cells at various population doubling timepoints (young, old, and senescent) grown separately under 3 and 20% oxygen growth conditions were selected for RNA extraction and hybridization on Affymetrix microarrays. Timepoints from cells grown under 3% and 20% oxygen conditions were age matched via population doublings to ensure accurate cross sample comparison.

Gene expression analysis of bleomycin-challenged mouse lung tissues with/without repetitive hyperbaric oxygen treatment

Project description:Idiopathic pulmonary fibrosis (IPF) is a dreadful, chronic, and irreversibly progressive disease leading to death with few effective treatments. Our previous study suggested that repetitive hyperbaric oxygen (HBO) treatment alleviates bleomycin-induced pulmonary fibrosis in mice, but the underlying mechanism is not clear. In this study, we compared the RNA-seq data from bleomycin challenged mouse lung tissues with or without repetitive HBO treatment, to figure out the affected biological processes and pathways. Combined with integrative analysis of public data from GEO, our study support HBO treatment as a viable strategy against pulmonary fibrosis.
