Project description:Obesity is predicted to become the largest modifiable risk factor for breast cancer in postmenopausal women, yet the mechanisms underlying this association are unclear. We identified a novel protective mechanism for lean adipocytes to suppress breast cancer by secreting the oxylipin 9S-HODE, which induces ferroptosis in breast cancer cells while sparing normal breast epithelial cells. Consequently, the inhibition of ferroptosis accelerates breast cancer in lean, but not obese, mice. Obese adipocytes fail to secrete 9S-HODE, suggesting that the loss of 9S-HODE significantly contributes to the acceleration of breast cancer in obesity. Further, 9S-HODE supplementation into tumors in obese mice is sufficient to reduce tumor burden and additionally can inhibit the growth of patient-derived breast cancer organoids, underscoring its potential as a therapeutic agent.

Project description:The progression of noninvasive ductal carcinoma in situ to invasive ductal carcinoma for patients with breast cancer results in a significantly poorer prognosis and is the precursor to metastatic disease. In this work, we have identified insulin-like growth factor–binding protein 2 (IGFBP2) as a potent adipocrine factor secreted by healthy breast adipocytes that acts as a barrier against invasive progression. In line with this role, adipocytes differentiated from patient-derived stromal cells were found to secrete IGFBP2, which significantly inhibited breast cancer invasion. This occurred through binding and sequestration of cancer-derived IGF-II. Moreover, depletion of IGF-II in invading cancer cells using small interfering RNAs or an IGF-II–neutralizing antibody ablated breast cancer invasion, highlighting the importance of IGF-II autocrine signaling for breast cancer invasive progression. Given the abundance of adipocytes in the healthy breast, this work exposes the important role they play in suppressing cancer progression and may help expound upon the link between increased mammary density and poorer prognosis.

Project description:The aim of the project was to compare global gene expression in adipocytes from obese patients and lean controls. Subcutaneous adipose tissue was collected from severely obese patients undergoing bariatric surgery (average body-mass index (BMI) of 45.5 kg/m2 (n = 12, thereof 4 men) and healthy lean patients undergoing hernia repairs (average BMI of 24.2 kg/m2 (n = 12, thereof 7 men), between 27 and 56 years of age. Adipocytes were isolated by collagenase treatment of adipose tissue, followed by filtering and centrifugation. Floating adipocytes were lysed in Qiazol before RNA purification and microarray analysis.

Secreted IL-1beta, in adipocytes culture medium, and insulin, in PANC-1 culture medium, were performed using ELISA assays. BPA was found to promote up-regulation of ER? and ERR?, and down-regulation of GPR30 expression modulating estrogen signaling and following a non-linear dose-response. Microarray data analysis demonstrated that BPA increases the gene expression of pro-inflammatory cytokines and lipid metabolism-related FABP4 and CD36 in adipocytes. PCSK1 resulted the most interesting gene being down-regulated by BPA thus impairing insulin production in pancreas. Project description:Type 2 diabetes is associated with obesity, and is characterized by insulin resistance in target tissues of the hormone combined with insufficient systemic insulin. Insulin resistance apparently begins in subcutaneous adipocytes that fail to further accumulate triacylglycerol. To understand the pathogenesis of transition from lean to obesity and to diabetes, we performed transcriptome profiling by RNA-sequencing of isolated primary human adipocytes. Half of identified transcripts were protein-coding and shared between individuals, while non-protein-coding transcripts were subject specific. Differential gene expression between lean and diabetes reflected corresponding changes in 7 of 12 characterized insulin-signaling proteins. In the tissue, adipocytes are surrounded by extracellular matrix (ECM) providing a dynamic scaffold. However, excess ECM is implicated in obesity and diabetes. Bioinformatic analyses of transcriptomes revealed major upregulation of genes related to ECM in obese compared with lean, which was reversed in diabetes. However, biochemical analysis of adipose tissue demonstrated reduced ECM in obesity or diabetes compared with lean. We conclude that marked changes in the adipocyte transcriptome related to ECM do not reflect global adipose tissue fibrosis, but likely control the pericellular milieu surrounding the adipocyte; and excess ECM is not a feature of subcutaneous adipose tissue in obesity or diabetes.
