Project description:The use of new natural eco-sustainable products is becoming an interesting option in order to reduce the use of chemical fertilizers and increase crop yields. Seaweed extracts are gained major attention as plant biostimulants due to their positive effect on plant-growth as well as on improving plants’ tolerance against abiotic stresses. Among the horticulture crops, lettuce (Lactuca sativa L.) is a major fresh vegetable crop in the Mediterranean area, which often requires the use of natural biostimulants to improve both the quantity and quality of production. The aim of this work was to assess the effect of either Chlorella vulgaris or Scenedesmus quadricauda extracts on lettuce seedlings (Lactuca sativa L.) by motoring the induced transcriptomic modifications using a RNASeq approach. The results showed that both C. vulgaris and S. quadricauda extracts positively influence the growth of lettuce seedlings. However, a higher reprogramming of the gene expression occurred in the case of C. vulgaris treatment than in S. quadricauda extract. Considering the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway functional enrichments, the main KEGG terms are in the “Biosynthesis of secondary metabolites”, “Metabolic pathways”, “Carbon metabolism” and “Biosynthesis of amino acids” categories. This study lays the basis for understanding the mechanisms and processes triggered by the use of microalgal extracts, which can represent an easier-to-handle and cheaper method for an eco-sustainable cultivation of lettuce plants than the application of chemicals.

Project description:Strawberry cultivation is associated with high mineral fertilizer doses and excessive use of chemical plant protection products. Based on previous research, we expected that chitin application to the growing medium will increase the nutrient availability and activate the plant’s systemic defense response, resulting in higher strawberry yields and less disease symptoms. We set up two experiments in which the temporal variability and differences in initial nutrient concentrations of the growing media were taken into account. Chitin induced the plant’s shoot biomass, explained by elevated N concentration in the growing medium and/or the attraction of plant-growth promoting fungal genera towards the plant root, such as Mortierella and Umbelopsis. The over-excessive N concentration and P and K deficiencies in the chitin treatment led to nutrient disbalances. This may explain the decreased fruit yield and disease resistance of the fruits towards Botrytis cinerea. In contrast, chitin caused a clear defense priming effect of the strawberry leaves, with a strong induction of the jasmonic acid response, resulting in less disease symptoms. Chitin caused positive effects on shoot growth and disease resistance of the leaves, but caution needs to be taken for nutrient disbalances leading to negative influences on fruit production and disease susceptibly.

We exploited this phenomenon to develop a method for increased lipid yield in stationary phase Chlorella sorokiniana cultures by 25% in just 24 hours, caused by a reproducible metabolic response that includes up-regulation of >30 lipid metabolism genes. This approach avoids the need to modify the strain or cultivation conditions, and does not affect cell viability or biomass.

Project description:Chlorella sp. HS2 is a halotolerant microalga exhibiting relatively high biomass productivity and substantially high lipid accumulation in marine growth media, which suggests this alga as an important crop for industrial algal cultivation systems. To determine pathways leading to HS2's acclimation responses to salt stress, we performed RNA-seq analysis with triplicated cultures grown in freshwater and marine media at both exponential and stationary growth phases. We then run de novo assembly to obtain HS2 transcriptome, which in turn was annotated and processed to extract dysregulated pathways. Results showed a large proportion of down-regulated genes, for instance photosynthesis and TCA pathways. Photosynthesis appeared, however, to recover at the stationary phase, while the general down-regulation pattern was maintained.

Project description:The use of biofertilizers is becoming an economical and environmentally friendly alternative to promote sustainable agriculture. Biochar from microalgae can be applied to enhance the productivity of food crops through soil improvement, slow nutrient absorption and release, increased water uptake, and long-term mitigation of greenhouse gas sequestration. Therefore, the aim of this study was to evaluate the stimulatory effects of biochar produced from Spirulina platensis biomass on the development and seed production of rice plants. Biochar was produced by slow pyrolysis at 300°C, and characterization was performed through microscopy, chemical, and structural composition analyses. Molecular and physiological analyses were performed in rice plants submitted to different biochar concentrations (0.02, 0.1, and 0.5 mg mL-1) to assess growth and productivity parameters. Morphological and physicochemical characterization revealed a heterogeneous morphology and the presence of K and Mg minerals in the biochar composition. Chemical modification of compounds post-pyrolysis and a highly porous structure with micropores were observed. Rice plants submitted to 0.5 mg mL-1 of biochar presented a decrease in root length, followed by an increase in root dry weight. The same concentration influenced seed production, with an increase of 44% in the number of seeds per plant, 17% in the percentage of full seeds per plant, 12% in the weight of 1,000 full seeds, 53% in the seed weight per plant, and 12% in grain area. Differential proteomic analyses in shoots and roots of rice plants submitted to 0.5 mg mL-1 of biochar for 20 days revealed a fine-tuning of resource allocation towards seed production. These results suggest that biochar derived from Spirulina platensis biomass can stimulate rice seed production.

Project description:Lettuce (Lactuca sativa L.) is a highly perishable horticultural crop with a relatively short shelf life due to leaf senescence that limits its commercial value and contributes to food waste. Postharvest senescence varies with influences of both environmental and genetic factors. Preharvest genetic factors can be indicative of postharvest quality. Discovery of additional preharvest markers to assess lettuce shelf life is an important step towards increasing the efficiency of lettuce breeding efforts for improved shelf life. We selected and evaluated three romaine lettuces with variable shelf lives with the aim of identifying preharvest markers of lettuce postharvest shelf life. We evaluated leaf morphological characteristics for each of the three cultivars. To assess molecular indicators of shelf life, we used an RNA sequencing approach to construct transcriptomic profiles of two of the cultivars, a short shelf life (SSL) breeding line 60184 and a long shelf life (LSL) cultivar ‘Okeechobee’ at maturity. We identified 552 upregulated and 315 downregulated differentially expressed (DE) genes between the genotypes. We found that 27 % of the DE lettuce genes had an Arabidopsis thaliana ortholog characterized as senescence-associated, indicating that variable expression of senescence-associated genes (SAGs) could serve as a tool for preharvest markers of postharvest shelf life. Notably, we identified several SAGs and functional groupings with highly differential expression between the cultivars. This includes several jasmonate ZIM-domain (JAZ), jasmonic acid (JA) signaling genes, chlorophyll a-b binding (CAB) chloroplast-associated genes, and cell wall modification genes including pectate lyases (PL) and expansins (EXP). This study presented an innovative approach for identifying molecular markers for preharvest factors linked to postharvest traits for prolonged shelf. These genes could potentially be developed further as preharvest predictors of shelf life for lettuce breeding

Project description:Green hydra (Hydra viridissima) harbors endosymbiotic Chlorella and have established a mutual relation. To identify the host hydra genes involved in the specific symbiotic relationship, transcriptomes of intact H. viridissima colonized with symbiotic Chlorella strain A99, aposymbiotic H.viridissima and H. viridissima artificially infected with other symbiotic Chlorella were compared by microarray analysis. The results indicated that genes involved in nutrition supply to Chlorella were upregulated in the symbiotic hydra. In addition, it was induced by supply of photosynthates from the symbiont to the host, suggesting cooperative metabolic interaction between the host and the symbiotic algae.
