Project description:Laryngeal squamous cell carcinoma (LSCC) has an insidious onset and strong heterogeneity. The lack of effective biomarkers makes early diagnosis difficult. Liquid biopsy technology provides a new direction to break through this bottleneck. The co-culture model of CAFs and LSCC cells confirmed that CAFs can promote LSCC glycolysis through exosomes to promote its malignant progression. Further screening of differential lncRNAs that drive LSCC glucose metabolism reprogramming and promote tumor progression was performed using exosome lncRNA chips.

Project description:Laryngeal cancer is a sub-type of head and neck cancer, and its treatment frequently entails the use of chemotherapy drugs like docetaxel. Nevertheless, the development of docetaxel resistance mechanisms presents a substantial challenge. Analyzing the transcriptome has become a fundamental approach for understanding the underlying molecular factors contributing to resistance and for pinpointing potential treatment targets. This type of analysis provides a comprehensive overview of the changes in gene expression that occur in laryngeal cancer cells that have become resistant to docetaxel. By comparing the transcriptomes of resistant and sensitive cells, we can identify potential biomarkers and targets for therapy. We have effectively created a laryngeal cancer cell line that is resistant to docetaxel, as evidenced by the increased expression of genes associated with various processes, such as microtubules (TUBB3, EML4ALK), receptors (TRK, PDGFR, GPCR, NOTCH), epithelial adherence (CDH1), metabolism (PIK3CA, DEPTOR, GSK-3β, PLCβ, CYP24A1), hypoxia (HIF-1α, HIF-1β), apoptosis (TNFRSF11A, p21, MDM2), cytokine production (STAT2/4/6, iNOS, IL-6, IL-8, GMCSFR) and autophagy (CFLAR, GABARAP, ATG4A, RasGEF). Taken together, our data provide a framework that encompasses therapeutic targets for inhibition of upregulated gene expression of docetaxel resistance on laryngeal cancer cells.

Project description:In vitro studies revealed that AEG-1 is palmitoylated at cysteine at a.a. 75 position. In order to check how palmitoylation regulates AEG-1 function in vivo we generated a knockin moue using CRISPR/Cas9 taking in which cysteine 75 was mutated to serine (AEG-1-C75S). Hepatocytes were isolated from AEG-1-WT and AEG-1C75S littermates, total RNA was isolated and RNA-sequencing was performed.

Project description:Background: MicroRNA (miRNA) is an emerging subclass of small non-coding RNAs that regulates gene expression and has a pivotal role for many physiological processes including cancer development. Recent reports revealed the role of miRNAs as ideal biomarkers and therapeutic targets due to their tissue- or disease-specific nature. Head and neck cancer (HNC) is a major cause of cancer-related mortality and morbidity, and laryngeal cancer has the highest incidence in it. However, the molecular mechanisms involved in laryngeal cancer development remain to be known and highly sensitive biomarkers and novel promising therapy is necessary. Methodology/Principal Findings: To explore laryngeal cancer-specific miRNAs, RNA from 5 laryngeal surgical specimens including cancer and non-cancer tissues were hybridized to microarray carrying 723 human miRNAs. The resultant differentially expressed miRNAs were further tested by using quantitative real time PCR (qRT-PCR) on 43 laryngeal tissue samples including cancers, noncancerous counterparts, benign diseases and precancerous dysplasias. Significant expressional differences between matched pairs were reproduced in miR-133b, miR-455-5p, and miR-196a, among which miR-196a being the most promising cancer biomarker as validated by qRT-PCR analyses on additional 84 tissue samples. Deep sequencing analysis revealed both quantitative and qualitative deviation of miR-196a isomiR expression in laryngeal cancer. In situ hybridization confirmed laryngeal cancer-specific expression of miR-196a in both cancer and cancer stroma cells. Finally, inhibition of miR-196a counteracted cancer cell proliferation in both laryngeal cancer-derived cells and mouse xenograft model. Conclusions/Significance: Our study provided the possibilities that miR-196a might be very useful in diagnosing and treating laryngeal cancer. Total RNA including low molecular weight RNA from tissue samples was isolated using the mirVanaTM miRNA Isolation Kit (Ambion) according to the manufacturer's instructions. The quality of the RNA samples was assessed using an Agilent 2100 Bioanalyzer, and only the samples meeting the criteria of 28S/18S > 1 and RNA Integrity Number (RIN) M-bM-^IM-% 7.5 were used for all analyses. For microarray analysis, we used the Human miRNA Microarray Kit V2 (Agilent Technologies, Santa Clara, CA), which contains 20-40 features targeting each of 723 human miRNAs (Agilent design ID 019118) as annotated in the Sanger miRBase, release 10.1. Labeling and hybridization of total RNA samples were performed according to the manufacturer's protocol. One hundred ng of total RNA was used as an input into the labeling reaction, and the entire reaction was hybridized to each array for 20 hours at 55M-BM-0C. The results were analyzed using Agilent GeneSpring GX7.3. Normal controls and cancer samples were compared using Welch's t-test (p<0.05) and differentially expressed miRNAs with at least a 2-fold change in expression were considered to be potential biomarkers.

Project description:M1 macrophages induce protective immunity against infection, but also contribute to metabolic and inflammatory diseases. Here we show that he E3 ubiquitin ligase, MDM2, promotes the glycolytic and inflammatory activities of M1 macrophage by increasing the production of IL-1β, MCP-1 and nitric oxide (NO). Mechanistically, MDM2 triggers the ubiquitination and degradation of E3 ligase, SPSB2, to stabilize iNOS and increases production of NO, which s-nitrosylates and activates HIF-1α for triggering the glycolytic and pro-inflammatory programs in M1 macrophages. Myeloid-specific haplo-deletion of MDM2 in mice not only blunts LPS-induced endotoxemia and NO production, but also alleviates obesity-induced adipose tissue-resident macrophage inflammation. By contrast, MDM2 haplodeletion induces higher mortality, tissue damage and bacterial burden, and also suppresses M1 macrophage response, in the cecal ligation and puncture-induced sepsis mouse model. Our findings thus identify MDM2 as an activator of glycolytic and inflammatory responses in M1 macrophages by connecting the iNOS-NO and HIF-1α pathways.
