Project description:Blood collection is essential for assessing the physiological status of laboratory animals, however, it inflicts pain, stress, tissue damage, and behavioral disorder to the animal. The administration of analgesics is one way to reduce the distress of animals suffering after experiments. Here we compared efficacious blood collection methods from the facial vein (FV), retroorbital plexus (RP), and lateral tail vein (TV) and the effects of ibuprofen administration based on animal welfare aspects. We evaluated body condition, behavioral activity, stress, pain, immunological profiles, and genetic changes in serial blood collections with or without ibuprofen administration for 8 weeks. We found that RP method reduced body weight, food intake, and water intake. RP method increased the grimace scales of orbital tightening and ear position. RP and TV methods reduced nest-building behaviors. RP and TV methods increased the concentrations of corticosterone, prostaglandin E2, and pro-inflammatory cytokines in peripheral blood. Infiltration of effector immune cells was increased in RP methods. Gene expressions associated with stress, pain, and immune system regulation were upregulated in all RP groups and ibuprofen non-administrated TV group. In contrast, the changed features were restored in FV method and ibuprofen administration. These data indicate that the FV blood collection and ibuprofen administration are suitable methods to mitigate physiologically harmful factors for mice depending on animal welfare considerations.

Project description:Several pharmaceuticals persist in wastewater plants and contaminate increasingly natural water bodies, even ground water. In this study, different bacteria with known capabilities to degrade aromatic environmental pollutants – belonging to the genera Bacillus, Paenibacillus, Mycobacterium and Cupriavidus - were screened for their ability to transform the anti-inflammatory drug ibuprofen. Bacillus megaterium SBUG 518 as one of the most efficient strains tested was almost completely inhibited by 0.05% ibuprofen at pH 6 but was able to transform 0.005% ibuprofen within 4 h up to 92%. Four products were identified by HPLC and LC/MS. The drug was transformed via three different biotransformation reactions involving (A) conjugate formation of ibuprofen with a sugar molecule (B) the hydroxylation of the isobutyl side chain at two positions, and (C) glycosylation of 2-hydroxyibuprofen. The ibuprofen conjugate (ibuprofen pyranoside) was considerably less toxic to B. megaterium than the parent compound. Its reversible formation can lead to a temporary remarkable underestimation of ibuprofen concentrations in environmental “elimination” studies. Proteomic analyses including 1264 identified proteins suggest that cytochrome P450 BM3 plays a pivotal role in ibuprofen hydroxylation. Additionally, several proteins involved in stress response as well as proteins with similarity to multidrug efflux pump proteins were found in higher abundance in ibuprofen-incubated bacteria.

Project description:Purpose: To investigate the involvement of Nfat5 in the regulation of an ibuprofen transporter Method: Reverse transfection with siRNA against Nfat5 was used to knockdown Nfat5 in MDCK I cells. The uptake of both radiolabelled taurine and ibuprofen was measured in MDCK I cells, first treated with siRNA against Nfat5 for 24 h and afterwards cultivated with raffinose-supplemented normal growth medium (500 mOsm) for 24 h. The knock down of NFat5 was investigated by qpCR and western blotting. A transcriptome analysis of MDCK I cells treated with siRNA against Nfat5 was performed. Validation of the transcriptome analysis was perform with qPCR and radiolabelled uptake of ibuprofen. Results: The siRNA transfection resulted in a knock down of Nfat5, and the uptake of both taurine and ibuprofen was significant decreased in the transfected MDCK I cells. The transcriptome analysis of MDCK I cells treated with siRNA against Nfat5 revealed genes regulated by Nfat5 during hyperosmotic exposure. qPCR and the transciptome analysis showed an involvement of Nfat5 in the hyperosmotic regulation of Tmem184 and uptake in Tmem184b-siRNA treated MDCK I cells showed that Tmem184b might be involved in uptake of ibuprofen. Conclusion: The regulation of Tmem184b was found regulated by Nfat5 and could be a potential possible candidate for the gene product responsible for the uptake of ibuprofen in MDCK I cells

Project description:It has long been recognized that there is substantial inter-individual variability in the analgesic efficacy of non-steroidal anti-inflammatory drugs (NSAIDs), but the mechanisms underlying this variability are not well understood. In order to characterize the factors associated with heterogeneity in response to ibuprofen, we performed functional neuroimaging, pharmacokinetic/pharmacodynamic assessments, biochemical assays, and gene expression analysis in twenty-nine healthy subjects who underwent third molar extraction. Subjects were treated with rapid-acting ibuprofen (400 mg; N=19) or placebo (N=10) in a randomized, double-blind design. Compared to placebo, ibuprofen-treated subjects exhibited greater reduction in pain scores, alterations in CBF in brain regions associated with pain processing, and inhibition of ex vivo COX-1 and COX-2 activity and urinary prostaglandin metabolites (p<0.05). Ibuprofen-treated subjects could be stratified into partial responders (N=9, required rescue medication) and complete responders (N=10, no rescue medication). This variability in analgesic efficacy was not associated with demographic/clinical characteristics, markers of systemic inflammation, or ibuprofen pharmacokinetics. Complete responders exhibited less suppression of urinary prostaglandin metabolites and greater induction of serum tumor necrosis factor-α and interleukin 8, compared to partial responders (p<0.05). Partial responders exhibited more alterations in gene expression in peripheral blood mononuclear cells after surgery, with an enrichment in inflammatory pathways. These findings suggest that activation of the prostanoid biosynthetic pathway and regulation of the inflammatory response to surgery differs between partial and complete responders. Future studies are necessary to elucidate the molecular mechanisms underlying this variability and identify biomarkers that are predictive of ibuprofen response.
