Project description:We have employed a single nucleus sequencing approach using 10x Genomics snRNA-seq to study the process of skeletal muscle hypertrophy. We examined the plantaris muscle of control adult mice (Ctl) and adult mice after 7 days of induced overload (7OV). This study highlights a new subpopulation of myonuclei in the muscles of control and hypertrophied mice.

Skeletal muscle specific overexpression of miRNA-1 impairs mechanical overload-induced skeletal muscle hypertrophy

Project description:MicroRNAs (miRNAs) are small, non-coding RNAs that play a critical role in regulating gene expression post-transcriptionally. Skeletal muscle-specific miRNAs, including miR-1, are important for skeletal muscle development and maintenance. In response to mechanical loading, skeletal muscle levels of miR-1 decrease by approximately 50%, suggesting a potential involvement in muscle hypertrophy. In the current investigation, we hypothesized that a reduction of miR-1 levels in response to mechanical loading would be necessary for skeletal muscle growth to occur. By significantly elevating miR-1 levels during the hypertrophic process via lentiviral delivery, we observed a blunted growth response in the plantaris muscle subjected to synergist ablation. A deeper RNA-based integrative analysis (transcriptomics and RNA eCLIP) indicates that miR-1 inhibits the expression of Itm2a and Melusin, two membrane-related proteins. While their exact mechanism in muscle hypertrophy is yet to be identified, our results suggest that miR-1-regulated membrane proteins are important for skeletal muscle hypertrophy.

Role of PDK1 in Skeletal Muscle Hypertrophy Induced by Exercise Load

Project description:Skeletal muscle mass is an important determinant of whole-body glucose disposal. We here show that mice (M-PDK1KO mice) with skeletal muscle–specific deficiency of 3'-phosphoinositide–dependent kinase 1 (PDK1), a key component of the phosphatidylinositol 3-kinase (PI3K) signaling pathway, manifest a reduced skeletal muscle mass under the static condition as well as impairment of exercise load–induced muscle hypertrophy. Whereas exercise load-induced changes in gene expression were not affected, the phosphorylation of ribosomal protein S6 kinase (S6K) and S6 induced by exercise load was attenuated in skeletal muscle of M-PDK1KO mice, suggesting that PDK1 regulates muscle hypertrophy not through changes in gene expression but through stimulation of protein synthesis via the S6K-S6 axis.

Targeted inhibition of platelet-derived growth factor signaling prevents muscle fiber growth during skeletal muscle hypertrophy

Project description:The platelet-derived growth-factor receptors alpha and beta (PDGFRα and PDGFRβ) mark fibroblasts and pericytes in skeletal muscle, respectively. While the role that these cells play in muscle growth has been evaluated, it was not known whether the receptors that mark these cells play a role in controlling transcriptional and functional changes during skeletal muscle hypertrophy. To evaluate this, we inhibited PDGFR signaling in mice subjected to a synergist ablation muscle growth procedure, and measured changes 3 and 10 days later. The results indicated that PDGF signaling was required for fiber hypertrophy and ECM production that occur during muscle growth.

Transcriptome analysis of satellite cells isolated from an animal model of skeletal muscle hypertrophy

Project description:Skeletal muscle is a post-mitotic tissue with unique plasticity. It has an extraordinary ability to adjust to changes in its physiological environment by altering its metabolic phenotype and its mass. Satellite cells are the most important myogenic progenitor cells involved in muscle regeneration and remodeling. The activation of these cells is governed by genetic programs involving complex transcriptional responses and can be involved in muscle hypertrophy adaptations. We described here gene expression variations evaluated by microarray analysis in satellite cells isolated from wild type and Magic-F1 transgenic mice, a model of skeletal muscle hypertrophy. The transcriptome analysis shows an altered gene signature in proliferating Magic-F1 satellite cells compared to controls, involving genes associated to muscle growth and vasculogenesis.

Microarray analysis of skeletal muscle hypertrophy induced by heat-stress in healthy humans

Project description:This study was aimed at examining the effects of long-term of heat-stress on the gene expression of skeletal muscle hypertrophy. Heat- and stream-generating (HSG) sheets were placed on thigh laterally. The HSG sheets (heat-stress) were applied 8-hrs/day, once a day, 4 days/weeks, for 10 weeks. A muscle biopsy was taken from the vastus lateralis muscle (2 cm depth) of the treated leg before and after the experiment. Oligonucleotide microarray revealed that genes related to ATP-synthesis, protein synthesis and the molecular chaperonic activity were increased by heat stress. These results suggest that heat-stress might be a useful countermeasure for muscular atrophy during aging. Overall design: Five subjects were applied by heat- and stream-generating (HSG) sheets for 10 weeks. Total RNAs obtained from the skeletal muscle before and after the experiment were used for the analysis.
