Project description:Novel target genes for the two transcription factor isoforms Pax6 and Pax6(5a) were identified by generating mouse embryonic fibroblast cell lines stably expressing either Pax6 or Pax6(5a), and comparing their gene expression pattern with the original mouse embryonic fibroblast cell line. Flp-In-3T3 cell lines were generated according to the manufacturers instruction (Invitrogen), using the mouse Pax6 and Pax6(5a) cDNA as insert. Correct inserts were verified by PCR, sequencing and Westernblot.

Project description:We identify mammalian TRIM71 as repressor of mRNAs that inhibits translation and affects mRNA stability. In this data set we compare the expression profile of human HEK293 Flp-In cells stably expressing TRIM71 to that of the parental cells, not expressing TRIM71 protein.
Project description:3T3 murine embryonic fibroblasts are used as feeder layers to support the proliferative potential of human primary keratinocytes during in vitro expansion for cell and gene therapy applications. To identify signals mediating this function, we analyzed fibroblast protein content using mass spectrometry. We compared the proteome of lethally irradiated fibroblasts used as feeder layers in two conditions: standard quality (SQ-FL) and low quality (LQ-FL) fibroblasts. Our findings highlight differences in extracellular matrix components between the two conditions
