Project description:Background: The pathogenesis of hepatitis B virus (HBV)-caused hepatocellular carcinoma (HCC) is complex and not fully understood. In clinical, the effective prevention and treatment of HCC rely on the accurate diagnosis. We developed a biology network approach to investigate the potential mechanisms and biomarkers of each stages from HBV infection to HCC. Methods Global gene profiling of healthy individuals (HC), HBV carriers (HBVC), chronic hepatitis B patients (CHB), liver cirrhosis (LC) and HCC was analyzed by gene array. Differentially expressed genes (DEG) were found by RVM (Random variance model) corrective ANOVA and STC (Series Test of Cluster) analysis.
Project description:In order to identify genes expression in different types of human NK cells, we sorted normal decidual NK cells during the first trimester, NK cells from peripheral blood and cord blood for analysis using the Agilent Whole Human Genome Microarray. Samples were collected from healthy adult donors after obtaining informed conset according to the Ethics Committee of the University of Science & Technology of China. CD56+CD3- NK cells have been sorted from normal decidua, cord blood and peripheral blood by FACS Aria. Then Whole Human Genome Microarray was employed as a discovery platform to identify genes differentially expressed in each types of NK cells.

RNA was extracted from snap fresh tissue collected from resected HCC tumor and adjacent non-tumor liver tissues. All HCC tumors were HBV-associated HCC.
