Project description:Plant nucleosome assembly protein-related proteins (NRPs) are histone chaperons involved in nucleosome turnover. Despite this basic cellular function, the Arabidopsis nrp1-1 nrp2-1 knock out mutant has been reported to exhibit only relatively mild seedling root phenotypes and to significantly affect the expression of only few hundred genes (Zhu et al., 2006). Here we report that NRP loss-of-function as well as the ectopic overexpression of At NRP1 significantly affected the growth, development, and the pathogen response of Arabidopsis plants under short day conditions. The nrp1-1 nrp2-1 mutant grew faster and flowered weeks earlier than the wild type and the overexpressor. The latter developed slower and flowered at a lower number of leaves than the mutant and the wild type. Moreover, the mutant was more sensitive, the overexpressor was more tolerant to pathogen infection correlating with their more adult and juvenile character, respectively. Transcriptomic comparison of mature non-bolting plants agreed with the phenotypes. The presented and published data indicate that although NRPs might not be absolutely required for plant growth and development, they contribute to the epigenetic coordination of metabolic, growth, defence and developmental processes during adaptation.

Project description:The biological basis of afternoon nap, a widespread yet poorly understood phenomenon, has remained elusive. Here we identify NPAS2, among core circadian regulators, as a sex-independent determinant of the nap behavior in mice. Specifically, medial prefrontal cortex (mPFC)-expressed NPAS2 orchestrates nap regulation through circadian modulation of local dopaminergic activity. We demonstrate that tyrosine hydroxylase-positive (TH+) neurons in mPFC exhibit time-of-day dependent wake-promoting activity, showing minimal excitation precisely during nap hours. Mechanistically, NPAS2 achieves this circadian suppression through a POU2F2-TH regulatory pathway: 1) transcriptional activation of the transcription repressor POU2F2, and 2) consequent downregulation of TH expression (a rate-limiting enzyme for dopamine synthesis) and dopamine production in mPFC TH+ neurons. These findings establish an endogenous circadian mechanism where mPFC NPAS2 periodically inhibits wake-promoting dopaminergic activity to drive nap behavior, providing fundamental insights into the neural and molecular regulation of nap biology.

Project description:Photoperiod, the main physical synchronizer of seasonal functions, is now acknowledged as a key player for the modulation of molecule access to cerebrospinal fluid (CSF). From previous work showing a different transfer rate from steroids or protein hormones from blood to CSF according to photoperiod and, a modulation of choroid plexus (CP) tight junction protein content with photoperiod, we hypothesized that CSF proteome under long days (LD) would differ from that of CSF sampled under short days (SD) in ovariectomized-estradiol replaced ewes. CSF protein expression variability between SD- or LD-treated ewes was therefore studied from pools of CSF collected for 48H from ewes of similar age and weight.

Project description:Increased expression of SIRT1 extends the lifespan of lower organisms and delays the onset of age-related diseases in mammals. Here, we show that SRT2104, a synthetic small molecule activator of SIRT1, extends both mean and maximal lifespan of mice fed a standard diet. This is accompanied by improvements in health, including enhanced motor coordination, performance, bone mineral density and insulin sensitivity associated with higher mitochondrial content and decreased inflammation. Short-term SRT2104 treatment preserves bone and muscle mass in an experimental model of atrophy. These results demonstrate it is possible to design a small molecule that can slow aging and delay multiple age-related diseases in mammals, supporting the therapeutic potential of SIRT1 activators in humans. Project description:The Dr_ebfC protein in D. radiodurans, encoded by the dr_0199 gene, was initially characterized as a NAP with DNA-protective and histone-like properties, involved in safeguarding the DNA from damage. To study the role of this protein in chromosomal organization and transcriptional regulation, we constructed dr_0199 deletion mutant in D. radiodurans. The mutant strain was viable, but compared with wild-type cells, the growth rate was moderately inhibited and the cell density during the stationary phase was lower than that of the wild-type. Through RNA-seq analysis of the mutant strain, we identified a total of 1077 differentially expressed genes, with 636 upregulated genes and 441 downregulated genes compared to the wild-type. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses indicated that the upregulated genes in the mutant were mainly associated with environmental stress response, DNA recombination, regulation of metabolic activities, catalytic activities, and DNA binding processes. Notably, several DNA damage repair-related genes (e.g., ddrA, ddrB, ddrC, ddrD, pprA) regulated by PprI/DdrO proteins showed significantly increased expression levels in the mutant. This observation suggests that the absence of the Dr_ebfC protein might lead to cellular stress, implying a potential role of this NAP in DNA protection. Enrichment analysis of the downregulated genes in the mutant revealed their involvement in vital cellular processes closely related to respiration, translation, RNA metabolism, and electron transfer, corroborating the observed slow growth phenotype of the mutant.

Project description:As in Eukaryotes, bacterial genomes are not randomly folded. Bacterial genetic information is generally carried on a circular chromosome with a single origin of replication from which two replication forks proceed bidirectionaly towards the opposite terminus region. Here we investigate the higher-order architecture of the Escherichia coli genome, showing its partition into two structuraly distinct entities by a complex and intertwined network of contacts: the replication terminus (ter) region and the rest of the chromosome. Outside ter, the condensin MukBEF and the ubiquitous nucleoid-associated protein (NAP) HU promote DNA contacts in the megabase range. Within ter, the MatP protein prevents MukBEF activity and contacts are restricted to ~280 kb creating a domain with distinct structural properties. We also show how other NAPs contribute to nucleoid organization, such as H-NS that restricts short-range interactions. Combined, these results reveal the contributions of major, evolutionary conserved proteins in a bacterial chromosome organization.

Project description:We sequenced mRNA from 16 hypothalamus (3-week-old= 8 and 6-week old=8) and 16 pitutary (3-week-old= 8 and 6-week old=8) of Ross308 fast growing-chicken line. In which groups were chickens characterized with high growth rate (n=4) and low growt rate (n=4). The cockerels were reared in a deep-litter facility in experimental farm of the National Research Institute of Animal Production located in Aleksandrowice till 22 or 42 days of age, under the same, optimal, electronically controlled environmental conditions (temperature, lighting regime, air humidity) and were fed ad libitum with the same complete starter (1-21 days of age), grower (22-35 days of age) and finisher (36-42 days of age) diets containing 22, 20.5 and 20.5% crude protein and 2990, 3130 and 3130 kcal/kg metabolic energy.

Project description:Protein synthesis and decay rates must be tightly coordinated to ensure proteome homeostasis. How this coordination is established is unknown. Here we use quantitative live cell imaging combined with computational inference and proteomics to determine how changes in global protein synthesis rates alter protein decay rates. A passive adaptation of protein degradation and dilution rates operates in cells tightly coupling proliferation rate to protein synthesis rates, allowing to partially buffer changes in proteome concentrations with increased loss of short-lived proteins. In contrast, the proliferation rate of Mus musculus embryonic stem cells (mESCs) is hyposensitive to moderate changes in protein synthesis. mESCs uncouple protein degradation and dilution rates and robustly maintain their protein levels. Finally, we also show that terminally differentiated human glial cells strongly adapt protein degradation rates to altered protein synthesis. Our work illuminates the complex interplay between protein synthesis, degradation, and cell division to regulate proteome homeostasis.

Project description:Aging is accompanied by complex alterations in brain structure and functions, with growing evidence implicating the choroid plexus (CP) as a key regulator of cognitive and motor function. Here, we investigated age-related changes in the CP of the gray mouse lemur (Microcebus murinus), a non-human primate model that recapitulates human brain aging features. Lemurs of different ages underwent behavioral testing, followed by transcriptomic profiling and immunofluorescence analyses of lateral ventricle CP tissue. Behavioral assessments revealed age-related declines in motor coordination and exploratory drive, whereas working memory and visual discrimination remained largely preserved. Histological analysis showed no significant structural alterations in CP architecture. Transcriptomic profiling identified 1,519 upregulated and 1,682 downregulated genes with aging, highlighting increased interindividual heterogeneity, upregulation of immune- and transport-related pathways, and downregulation of signaling and intercellular communication processes. Functionally, AQP1 protein expression decreased with age without changes in mRNA levels, suggesting post-transcriptional regulation, whereas NKCC1 and TTR expression were largely maintained. Notably, reduced AQP1 expression correlated with age-related motor decline. These findings indicate that CP aging in the mouse lemur is characterized by selective functional vulnerability, immune activation, and transcriptional remodeling, yet essential barrier and secretory functions are largely preserved. Our results highlight a primate-like aging profile of the CP, providing insights into mechanisms by which CP dysfunction may contribute to age-related motor decline and altered brain homeostasis.
