Project description:Safeners are an important group of agrochemicals that can extend the selective use of existing herbicides into a wider range of cereals by protecting the crops from chemical injury. The recently released aromatic sulphonamide safeners cyprosulfamide and metcamifen were tested for their ability to protect rice plants from the phytotoxicity caused by clodinafop-propargyl, a herbicide normally only used in wheat. Transcriptome studies in rice cultures demonstrated that whereas cyprosulfamide had a negligible effect on gene expression over a 4 h exposure, metcamifen perturbed the abundance of 590 transcripts. Analysis of the inducible genes suggested that safening elicited similar gene families to those associated with specific biotic and abiotic stresses, notably those elicited by abscisic acid, salicylic acid and methyl-jasmonate.

Project description:Indica rice seedlings of IR64 variety were grown hydroponically for 7-days in a culture room with a daily photoperiodic cycle of 14h light and 10h dark. Seedlings were incubated in 0.1% dimethyl sulfoxide (control) or 50 micromolar solutions of indole-3-acetic acid (IAA treatment) and benzyl aminopurine (BAP treatment) for 1h and 3h. Equal amounts of 1h and 3h samoles were pooled for each treatment before RNA isolation. Cytosolic and plasma membrane proteome analysis of rice leaves in response to MSP1 treatment

Project description:MSP1 is a Magnaporthe oryzae secreted protein that elicits defense responses in rice. However, the molecular mechanism of MSP1 action is largely elusive. Here, we employed a TMT-based quantitative proteomic analysis of cytoplasmic as well as plasma membrane proteins to decipher the MSP1 induced signalling in rice. This approach led to the identification of 6691 proteins of which 3049 were identified in the plasma membrane (PM) while 3642 were identified in the cytoplasmic fraction. A parallel phosphoproteome analysis led to the identification of 1906 phosphopeptides and integration of proteome and phosphoproteome data showed activation of proteins related to the proteolysis, jasmonic acid biosynthesis, redox metabolism and MAP kinase signaling pathways in response to MSP1 treatment. Further, MSP1 induced phosphorylation of some of the key proteins including RBOHB, MEKK1, MPK3/6, CDPK and CaM suggest activation of PAMP-triggered immunity (PTI) in response to MSP1 treatment. In essence, our results further support the functioning of MSP1 as a PAMP and provide an overview of the MSP1 induced signaling in rice leaves.

Project description:Rice blast disease is a major threat to rice production worldwide, but the mechanisms underlying rice resistance to the causal agent Magnaporthe oryzae remain elusive. In this whole-genome transcriptome study of rice early defense response to M. oryzae, we applied Affymetrix Rice Genome Genechip to compare the compatible and incompatible rice-M. oryzae interactions in 24 hours post-inoculation. Leaf samples were harvested from three biological replicates of fungal- and mock-inoculated seedlings at 24 hours post-inoculation, from which RNA were extracted and analyzed with Genechip Rice Genome Array.

Cellular heterogeneity and therapeutic response profiling of IDH-wildtype glioma stem cell cultures

Project description:Gliomas are the most common type of primary malignant adult brain tumor. They appear to originate from neuroglial stem or progenitor cells, and are therapeutically challenging due to an invasive growth pattern and the absence of effective therapies. We have analyzed cellular, molecular and proteomic features and defined the therapeutic response profiles of four IDH1-wildtype glioma stem cell (GSC) cultures. All four GSC cultures were established from Grade IV glioblastoma (GBM) surgical resection tissue, can be continuously propagated and are highly enriched for stem/tumor repopulating cells. Integrated genomic and proteomic analysis of all four cultures was performed, together with use of a dual molecular bar-coding strategy to assess GSC population heterogeneity and the response to ionizing radiation. These well-characterized, bar-coded GSC cultures provide an experimentally tractable resource for investigating glioma biology, and to use to identify new and potentially more effective GBM therapies and treatment regimens.
