Impact of parental obesity on adipose progenitor programming

Project description:Parental obesity (PO) increases the risk of obesity and metabolic syndromes in offspring. To investigate how PO influences adipose progenitors, we generated offspring from lean (Ln-F1) or obese (Ob-F1) parents and isolated the stromal vascular fraction (SVF) from inguinal white adipose tissue (iWAT) depots at postnatal day 21 (P21). Bulk RNA sequencing of the SVF was performed to uncover the molecular mechanisms by which PO programs adipose progenitors.

Project description:Infants experience distinct ketogenesis during the preweaning period, but its physiological significance remains unclear. Here, we show that preweaning ketosis enhances beige fat biogenesis and improves metabolic health in adulthood. To elucidate the underlying mechanisms, we performed multi-omics analyses of inguinal white adipose tissue (iWAT) and its stromal vascular fraction (SVF). Bulk RNA-seq of iWAT was conducted in 8-week-old Hmgcs2 knockout (KO) and control mice housed at room temperature (23 °C) or exposed to cold (4 °C). To further examine the effects of elevated ketosis, we administered oral 1,3-butanediol (1,3BD) or water to pups from postnatal day 2 to 21, and analyzed the iWAT SVF using bulk RNA-seq, single-cell RNA-seq, and ChIP-seq targeting H3K9ac, H3K14ac, and H3K9bhb. These integrative analyses reveal how early-life ketosis programs adipose progenitors to promote beige adipocyte development.

scRNA-seq of stromal vascular cells from 48 week (aged) mouse subcutaneous adipose tissue compared to control 10 week (young) mouse SVF

Project description:Adipose tissue mass and adiposity change throughout the lifespan. During aging, while visceral adipose tissue (VAT) tends to increase, peripheral subcutaneous adipose tissue (SAT) decreases significantly. Unlike VAT, which is linked to metabolic diseases, SAT has beneficial effects. However, the molecular details behind aging-associated loss of SAT remain unclear. Here we compare scRNA-seq of total SVF of SAT from young and aging mice to identify a novel Aging-dependent Regulatory Cell (ARC) that emerges in SAT of aged mice. Inguinal white adipose tissue (iWAT) was used as a representative SAT; iWAT pads of 2 mice from each age group were subjected to collagenase digestion and treated with a hypotonic buffer to remove red blood cells before subjection to scRNA-seq by 10X Genomics Chromium Single Cell Kit. The findings showed that ARCs express adipogenic markers but lack adipogenic capacity and inhibit differentiation of neighboring adipose precursors.

Project description:To assess the influences of a crude medicinal herb extract (MHE) on the immune composition and function in inguinal white adipose tissue (iWAT), we isolated iWAT stromal vascular fractions (SVFs) from control (PBS) and MHE-treated mice and performed RNA-seq analysis. iWAT SVF of mice treated with MHE was defined as the treatment group. Phosphate-buffered saline (PBS) treatment was used as the control group. Then RNA-Seq experiment was performed by OE Biotech Co., Ltd. (Shanghai, China) to analyze gene expression changes in iWAT SVF.

Transcriptomic profiling of adipose tissue in a mouse model of chronic social isolation-induced glucose metabolism dysfunction

Project description:Social isolation is a growing global public health concern, associated not only with mental health disorders but also with metabolic diseases such as type 2 diabetes. This study investigates the molecular mechanisms by which chronic social isolation disrupts glucose homeostasis via sympathetic nervous system activation. To elucidate adipose tissue transcriptional responses under socially isolated conditions, inguinal white adipose tissue (iWAT) was collected from male C57BL/6J mice subjected to either standard group housing or single housing for 8 weeks (n = 3 per group). Bulk RNA sequencing was performed to characterize differential gene expression. Analysis revealed substantial transcriptional reprogramming in iWAT from isolated mice, including upregulation of thermogenic and lipid metabolic pathways, such as Ucp1, and activation of key transcriptional regulators like Pparg. These findings suggest that social isolation induces sympathetic-driven metabolic remodeling in adipose tissue. The dataset provides valuable insight into how psychosocial stress influences peripheral metabolic gene networks and may contribute to stress-related metabolic disorders.

Spatial Recruitment of Cardiolipins in White Adipose Tissue after Cold Stimulation is Independent of UCP1

Project description:Abstract Brown and brite adipocytes are the key cells performing uncoupling protein 1 (UCP1) dependent non-shivering thermogenesis (NST) induced by cold exposure. Several lipid species are associated to NST in brown and white adipose tissue. Studies investigating the association of the lipid profile with NST rely on the analysis of whole organ homogenates or on the differentiation of pre-adipocytes in vitro. These approaches have so far not addressed the heterogeneity of white adipose tissue. Aim of this study was to characterize the lipid composition of white adipose tissue on a region-specific level in an in vivo context. We applied MALDI mass spectrometry imaging (MALDI-MSI) in combination with immunohistochemistry and high-resolution mass spectrometry on sections of inguinal white adipose tissue of 129S6/SvEvTac and C57BL6/N-UCP1 knockout and wildtype mice acclimatized to cold to identify lipids specific to areas of UCP1 expression. Based on the analysis of cold exposed 129S6/SvEvTac mice we identified cardiolipins (CL) and diacylglycerols (DG) species to be specific for areas expressing UCP1 and triacylglycerols (TG) to be the main lipid class characteristic for UCP1 negative regions within inguinal white adipose tissue. Investigation of C57BL6/N-UCP1 knockout and wildtype mice housed at either room temperature or acclimatized to cold, demonstrated that CL content in white adipose tissue is increased upon cold stimulation, independent of UCP1. We introduce a MALDI-MSI based approach to identify lipids associated to thermogenic adipocytes in adipose tissues demonstrating a clear regional cold dependent upregulation of CL independent of UCP1.
