Project description:This study focuses on the molecular mechanisms underlying the response of duckweed (Landoltia punctata) to high nitrogen and phosphorus stress in biogas slurry from pig farms. Through integrated transcriptomic and proteomic analyses, this study systematically elucidates, for the first time, how duckweed employs remarkable post-transcriptional regulatory strategies to remodel ribosomal function and photosynthetic carbon metabolism, thereby significantly enhancing protein synthesis and accumulation. The work provides novel insights into the adaptive mechanisms of aquatic plants under nutrient stress.

Project description:Reduction in visceral adipose tissue (VAT) mass reduces body weight and metabolic disease risk in obese patients. However surgical removal of VAT is highly invasive and thus not clinically feasible. We developed an injectable ice slurry for selective reduction of adipose tissue through cryolipolysis. The aim of this study was to investigate safety, feasibility and mechanism of ice slurry-induced cryolipolysis of VAT. Perigonadal VAT in diet-induced obese mice and rats was subjected to slurry or sham treatment. Body weight and blood chemistry were monitored for 56 days post-treatment. Histological analysis and molecular studies were performed to elucidate mechanisms of fat reduction. Treatment of VAT was well tolerated in all animals. Slurry induced adipocyte cell death via selective cryolipolysis; significant weight loss was noted at day 21 post-treatment. RNA sequencing from treated VAT samples showed increased expression of genes involved in inflammation, immune response, collagen biosynthesis and wound healing, and decreased expression of adipokines. This study demonstrates that slurry treatment is safe and effective in inducing cryolipolysis of VAT and subsequent weight loss in rodents. Ice slurry is promising as a minimally-invasive treatment to reduce visceral adipose tissue.

Project description:The use of biofertilizers is becoming an economical and environmentally friendly alternative to promote sustainable agriculture. Biochar from microalgae can be applied to enhance the productivity of food crops through soil improvement, slow nutrient absorption and release, increased water uptake, and long-term mitigation of greenhouse gas sequestration. Therefore, the aim of this study was to evaluate the stimulatory effects of biochar produced from Spirulina platensis biomass on the development and seed production of rice plants. Biochar was produced by slow pyrolysis at 300°C, and characterization was performed through microscopy, chemical, and structural composition analyses. Molecular and physiological analyses were performed in rice plants submitted to different biochar concentrations (0.02, 0.1, and 0.5 mg mL-1) to assess growth and productivity parameters. Morphological and physicochemical characterization revealed a heterogeneous morphology and the presence of K and Mg minerals in the biochar composition. Chemical modification of compounds post-pyrolysis and a highly porous structure with micropores were observed. Rice plants submitted to 0.5 mg mL-1 of biochar presented a decrease in root length, followed by an increase in root dry weight. The same concentration influenced seed production, with an increase of 44% in the number of seeds per plant, 17% in the percentage of full seeds per plant, 12% in the weight of 1,000 full seeds, 53% in the seed weight per plant, and 12% in grain area. Differential proteomic analyses in shoots and roots of rice plants submitted to 0.5 mg mL-1 of biochar for 20 days revealed a fine-tuning of resource allocation towards seed production. These results suggest that biochar derived from Spirulina platensis biomass can stimulate rice seed production.

Project description:The utilization of biogas slurry is critical for the sustainable development of animal husbandry. Biomass carbon adsorption is a feasible method for the recycling of nutrients from biogas slurry. However, research on the co-adsorption of ammonia nitrogen and phosphate is scarce. Herein, soybean straw was utilized as the raw material to prepare Ca2+-modified biochar (CaSSB), which was investigated for its ammonia nitrogen and phosphate adsorption mechanisms. Compared with natural biochar (SSB), CaSSB possesses a high H/C ratio, larger surface area, high porosity and various functional groups. Ca2+-modified soybean straw biochar exhibited excellent adsorption performance for NH4+-N (103.18 mg/g) and PO43--P (9.75 mg/g) at pH = 6, using an adsorbent dosage of 2 g/L. The experimental adsorption data of ammonia nitrogen by CaSSB corresponded to pseudo-second-order kinetics and the Langmuir isotherm model, suggesting that the adsorption process was homogeneous and that electrostatic attraction might be the primary adsorption mechanism. Meanwhile, the adsorption of phosphate conformed to pseudo-second-order kinetics and the Langmuir-Freundlich model, whose mechanism might be attributed to ligand exchange and chemical precipitation. These results reveal the potential of CaSSBs as a cost-effective, efficient adsorbent for the recovery of ammonium and phosphate from biogas slurry.
