Influence of seagrass decline on the metabolic profile of sediment microbial communities

Project description:Seagrass meadows are highly productive ecosystems that are considered hotspots for carbon sequestration. The decline of seagrass meadows of various species has been documented worldwide, including that of Cymodocea nodosa, a widespread seagrass in the Mediterranean Sea. To assess the influence of seagrass decline on the metabolic profile of sediment microbial communities, metaproteomes from two sites, one without vegetation and one with a declining Cymodocea nodosa meadow, were characterised at monthly intervals from July 2017 to October 2018. The differences in the metabolic profile observed between the vegetated and nonvegetated sediment before the decline were more pronounced in the deeper parts of the sediment and disappeared with the decay of the roots and rhizomes. During the decline, the protein richness and diversity of the metabolic profile of the microbial communities inhabiting the nonvegetated sediment became similar to those observed for the vegetated communities. Temporal shifts in the structure of the metabolic profile were only observed in the nonvegetated sediment and were also more pronounced in the deeper parts of the sediment. The assessment of the dynamics of proteins involved in the degradation of organic matter, such as ABC transporters, fermentation-mediating enzymes, and proteins involved in dissimilatory sulphate reduction, reflected the general dynamics of the metabolic profile. Overall, the metabolic profile of the microbial communities inhabiting the nonvegetated sediment was influenced by the decline of seagrass, with stronger shifts observed in the deeper parts of the sediment.

Cymodocea nodosa response to simulated CO2-driven ocean acidification

Project description:We experimentally manipulate CO2 levels and investigate the effects of high-CO2/low pH on seagrass metabolism and underlying molecular mechanisms. Cymodocea nodosa plants were collected in Cadiz Bay at the end of January 2014 and transported to the mesocosm facility. After a one week acclimation period, C. nodosa were either kept under normal (400 ppm) or elevated (1200 ppm) CO2 concentration for 12 days. RNA extraction and sequencing was performed in all phases to test differential expression of genes in different conditions.

Dual-compartment shotgun proteomics unravel thermal stress responses in reef-calcifiers and their photosymbionts

Project description:The proliferation of coral reef organisms, such as the key ecological engineers and carbonate producers’ large benthic foraminifera (LBF), greatly relies on the mutual association with photosymbiotic algae. With respect to global change impacts such as ocean warming, evaluating stress and acclimatization responses is highly important, but probing proteins and partitioning host and symbionts are major obstacles for studying LBF. We therefore applied a label-free quantitative proteomics approach to LBF holobionts in order to detect changes in the relative abundances of proteins in response to different thermal-stress scenarios. Subsequent identification allowed for the partial assignment of 1,618 proteins to either foraminiferal host or endosymbiotic diatoms. While single and episodic thermal-stress events only induced minor metabolic impacts, chronic thermal stress diminished photosynthesis-related proteins among the symbionts. Stress response and proteolysis-related proteins increased in abundance, illustrating the extent of damage. In contrast to the symbionts, host homeostasis was maintained through extensive repair mechanisms and high protein turnover. Metabolic pathways were adjusted to the symbiont loss by using alternative energy resources. However, linking proteome to physiological data revealed that resource redistribution impairs growth and certainly other functions that could disturb carbonate production. We show how compartment partitioning can give insights into interactions and stress response mechanisms in photosymbiotic reef-calcifiers.

Amino acid catabolism fuels microalgal lipogenesis

Project description:Microalgal lipid, a feasible substrate for biofuel, is typically accumulated during the stationary growth phase. Generating strains which trigger lipogenesis from the exponential growth phase will enhance lipid productivity, reduce cost of biofuel production. We characterized a lipid-rich microalgal mutant, Dunaliella tertiolecta, which exhibited a six-fold enhancement of neutral lipids production in the exponential growth phase with marginal compromise on growth (4%). Using transcriptomics and metabolomics, regulatory mechanisms of the mutant were uncovered.
