Maternal IgG Affects Brain Development of Mouse Pups

Project description:Maternal IgG is an immunoglobulin present in breast milk and has been shown to play a role in the development of the immune system in infants. The aim of this study was to investigate the effects of maternal IgG on brain development in mouse pups. Maternal IgG immunoreactivity was observed in microglial cells of the pup brain, peaking at postnatal 8 day. In particular, strong IgG immunoreactivity was observed in the microglia in the corpus callosum and cerebellar white matter. IgG stimulation of primary cultured microglia resulted in activation of the type I interferon feedback loop by Syk. Among the IgG subclasses, IgG2a was found to have a strong influence on microglia. Analysis of FcRn KO mice that were unable to take up IgG from their mothers revealed that these mice had abnormalities in the proliferation and/or survival of microglia, oligodendrocytes, and some types of interneurons. FcRn KO mice also exhibited abnormalities in social behavior and adaptation to novel environments. This study suggests that maternal IgG affects brain development in pups.

Effect of gut microbiome transplant from neonates of particle-exposed mothers on the epigenome of dendritic cells

Project description:In our model the newborns of asthmatic mother mice or of mothers exposed to air pollutant particles are born with a predisposition to asthma. Gut microbiome of these pups is altered, and the transplant of the pups’ microbiome (GMT) has conferred the asthma predisposition to naïve recipients. We hypothesized that bacteria alter metabolomic profile in the gut, which polarizes the dendritic cells (DC) in the recipient by affecting epigenetic regulation in these key decision-maker cells. Here we examined DNA methylation profiles in the recipient host’s DCs to test the prediction that GMT confers alterations in DNA methylation (not seen with sterilized GMT).

Sex specific gene expression differences in mouse pups from obese dams

Project description:Maternal obesity is increasingly common and negatively impact offspring health. Children born to mothers with obesity are at higher risk of developing diseases associated with abnormalities within the hematopoietic system such as atypical immune profiles, hematopoiesis, and metabolic diseases such as type 2 diabetes. Interestingly, disease risks are often dependent on the offspring’s sex, suggesting sex-specific reprogramming effect of maternal obesity on offspring hematopoietic stem and progenitor cell (HSPC) function. However, the impact of maternal obesity exposure on offspring HSPC function is largely unknown, and the capability of HSPC to regulate offspring metabolic health has not been studied. Here we examined differential transcriptomes of hematopoietic stem and progenitor cells from male and female pups (postnatal day 21) from dams given western diet or control diet. RNA-seq revealed inflammatory gene pathways in female MatOb offspring that potentially protect HSPC function.
