Project description:N4-acetylation of cytidine (ac4C) in RNA are crucial for regulating the expression level and promoting the translate efficiency of mRNA. However, epitranscriptomic profiling of ac4C-related RNA (mRNA/ lncRNA/ circRNA) acetylation in renal cortex are largely unknown. Therefore, we profiled the transcriptome-wide ac4C modification in mRNA/ lncRNA/ circRNA in renal cortex from either ischemia reperfusion-acute kidney injury (I/R-AKI) mouse or Sham-operated mouse. At the same time, the expression levels of mRNA/ lncRNA/ circRNA were also evaluated by the sequencing experiment in input group. Therefore, this is an experiment for quantifying both the expression levels and the ac4C modification alterations in mRNA/ lncRNA/ circRNA within renal cortex from I/R-AKI mouse.
Project description:To elucidate the molecular mechanism and signaling pathways that PinX1 is involved in, a Human LncRNA Array V2.0 (Arraystar, USA)-based genome wide screen of the alterations in lncRNA and mRNA expression profiles was performed in MCF-7 cells stably overexpressing PinX1. MCF-7 breast cancer cells were transfected with the pcDNA3.1-PinX1 and pcDNA3.1 empty vector and the microarray analysis were performed after the clonal selection of cells with a high expression level.
Project description:we found a total of 570 circRNA that have difference, among which 155 are up-regulated and 415 are down-regulated; and 898 lncRNA that have difference, in which 414 are up-regulated and 484 are down-regulated; and 476 mRNA that have difference, in which 98 are up-regulated and 378 are down-regulated. In this work, we sequenced circRNA, lncRNA and mRNA from recurrent COVID-19 patients and healthy people, and compared the differences.
