Project description:BRD-K34222889 is a structural analog of the anti-inflammatory and senolytic drug piperlongumine. Piperlongumine inhibits GSTP1, an enzyme that places glutathione moieties on cysteine residues of various protein targets and for modulation of protein stability. The objective of this experiment was to determine the effect of a Piperlongumine analog BRD-K34222889 in endothelial dysfunction. Interestingly, BRD-K34222889 was found to inhibit glutathione S-transferase P (GSTP1), which increased ISCU protein stability via preventing glutathionylation and thus increased oxidative metabolism and decreased PAEC apoptosis. Activites of BRD-K34222889 reversed gene transcripts altered in PAECs with hypoxia that more than half of these genes were categorized in pathways relevant to cell cycle, cell death, and metabolism and all known to be controlled by ISCU. We use Affymetrix Clairom S platform to determine the whole gene expression.

Project description:Pulmonary arterial hypertension (PAH) is a fatal disease characterized by a proliferative endothelial cell phenotype, inflammation and pulmonary vascular remodeling. BMPR2 loss-of-function has been linked to pathologic plexiform lesions with obliteration of distal pulmonary arteries distal pulmonary arteries BMPR2 silencing inprimary human pulmonary artery ECs (HPAECs) recapitulate important aspects of cellular dysfunction and deregulated signaling associated with PAH. Primary HPAECs were transfected with gene-specific siRNA pools targeting BMPR2 or control siRNA followed PMA or control stimulation.

Project description:In this study, we analzyed differences in the effect of hypoxia on the transcriptomic profile of cultured human pulmonary artery endothelial cells (HPAECs) vs. human brain microvascular endothelial cells (HBMVECs). To determine which hypoxia-regulated genes were dependent on hypoxia inducible factor 1-alpha (HIF-1alpha), some cells were transfected with an siRNA against HIF-1alpha prior to treatment with hypoxia. At the completion of the treatment protocol, mRNA was isolated using standard methodology and analyzed further by RNA-seq.

Project description:A number of mediators that have been found to be involved in the pathogenesis of hypoxic pulmonary hypertension, these mediators including mRNA, microRNAs and long noncoding RNAs. Endothelial dysfunction plays a major role in the initiation of pulmonary vascular remodeling and is thought to be a major contributor to this process. However, mRNA and no-coding RNAs expression profiles and their biological functions in pulmonary artery endothelial cells (PAECs) exposed to hypoxia need to be investigated further. Pulmonary artery endothelial cells (PAECs) were exposed to normoxia condition (21% O2, 5% CO2, 74% N2) or hypoxia condition (3% O2, 5% CO2, 92% N2) (n=3 for each group).PAECs were harvested after 24 hours and performed to further analysis.
