Project description:Non-small cell lung cancer (NSCLC) is the leading life-threatening cancer in the world. Gamboge, a type of desiccating resin excreted from gamboge stalk, has been reported to have antitumor effects in various malignancies. However, little is known about the underlying mechanism in lung cancer. In this study, we discovered a novel compound in gamboge, epigambogic acid A, which significantly inhibited lung cancer cell growth with 99.94% inhibition. In addition, epigambogic acid A suppressed cell invasion and metastasis, and promoted apoptosis in NSCLC cells. The combinatorial analysis of transcriptome sequencing and The Cancer Genome Atlas (TCGA) database indicated that epigambogic acid A could remarkably decrease the expression of oncogenes such as disrupted in renal cancer 1 (DIRC1) and collagen type III alpha 1 chain (COL3A1). Moreover, DIRC1 promoted cell proliferation and clone formation in NSCLC cells. Overall, these results identify a key role of epigambogic acid A in tumor suppression and uncover the mechanism of action of epigambogic acid A and DIRC1, which provides potential implications in NSCLC therapy.

Merlin/NF2 Suppresses Tumorigenesis by Inhibiting the E3 Ubiquitin Ligase CRL4DCAF1 in the Nucleus

Project description:Current models imply that the FERM domain protein Merlin, encoded by the tumor suppressor NF2, inhibits mitogenic signaling at or near the plasma membrane. Here, we show that the closed, growth inhibitory form of Merlin accumulates in the nucleus, binds to the E3 ubiquitin ligase CRL4DCAF1, and suppresses its activity. Depletion of DCAF1 blocks the promitogenic effect of inactivation of Merlin. Conversely, enforced expression of a Merlin-insensitive mutant of DCAF1 counteracts the antimitogenic effect of Merlin. Re-expression of Merlin and silencing of DCAF1 induce a similar, tumor-suppressive program of gene expression. Tumor-derived mutations invariably disrupt Merlinâs ability to interact with or inhibit CRL4DCAF1. Finally, depletion of DCAF1 inhibits the hyperproliferation of Schwannoma cells from NF2 patients and suppresses the oncogenic potential of Merlin-deficient tumor cell lines. We propose that Merlin suppresses tumorigenesis by translocating to the nucleus to inhibit CRL4DCAF1. To examine if Merlin controls gene expression through inhibition of CRL4DCAF1, and define the general function of this ligase, we compared the gene expression program activated by expression of Merlin or by depletion of DCAF1 in Merlin-null FC-1801 mouse Schwannoma cells

PKM2 methylation by CARM1 activates aerobic glycolysis to promote tumorigenesis

Project description:Metabolic reprogramming is a hallmark of cancer. Herein we discover that the key glycolytic enzyme pyruvate kinase M2 isoform (PKM2), but not the related isoform PKM1, is methylated by co-activator-associated arginine methyltransferase 1 (CARM1). PKM2 methylation reversibly shifts the balance of metabolism from oxidative phosphorylation to aerobic glycolysis in breast cancer cells. Oxidative phosphorylation depends on mitochondrial calcium concentration, which becomes critical for cancer cell survival when PKM2 methylation is blocked. By interacting with and suppressing the expression of inositol-1,4,5-trisphosphate receptors (InsP3Rs), methylated PKM2 inhibits the influx of calcium from the endoplasmic reticulum to mitochondria. Inhibiting PKM2 methylation with a competitive peptide delivered by nanoparticles perturbs the metabolic energy balance in cancer cells, leading to a decrease in cell proliferation, migration and metastasis. Collectively, the CARM1-PKM2 axis serves as a metabolic reprogramming mechanism in tumorigenesis, and inhibiting PKM2 methylation generates metabolic vulnerability to InsP3R-dependent mitochondrial functions.
