Galectin-3-Binding Protein, A Potential Biomarker from Central Obesity to Type 2 Diabetes

Project description:Diabetes has emerged as a significant public health concern. The existing diabetes diagnostic markers, such as fasting glucose and glycated hemoglobin, have limitations in accuracy and sensitivity in epidemiological investigations. This study identified new serum biomarkers for diabetes through a three-step process: screening, validation, and correlation analysis. By excluding proteins that exhibit opposing trends in DDA and DIA results, we identified 170 differentially expressed proteins. Subsequent quantification of six differential proteins in the plasma of 38 diabetic patients and 32 controls through ELISA and immunoturbidimetry showed a significant upregulation of Adipocyte plasma membrane-associated protein (APMAP), Galectin-3-binding protein (LG3BP), Cathepsin D (CATD), and Beta-2-microglobulin (B2MG) in the diabetic group. Notably, LG3BP demonstrated a strong association with established diabetes markers and risk factors in the correlation analysis, particularly waist circumference, suggesting its potential as a serum/plasma biomarker for diabetes closely related to central obesity. Those findings not only contribute to the repertoire of diagnostic biomarkers for diabetes but also provide a robust analytical framework for the identification of specific protein markers in large-scale serum/plasma samples.

Oxidative Stress Biomarker of Type 2 Diabetes Mellitus

Project description:In this project, using discovery-based mass spectrometry approach and adopting data-dependent LC-MS/MS, we detected and identified modified peptides in complex clinical plasma samples and developed first-level biomarker verification strategy. Further, using liquid chromatography-multiple reaction monitoring mass spectrometry (LC-MRM-MS) based targeted approach which virtually quantifies irreversible oxidation at any targeted cysteine, we quantified cysteine tri-oxidation in HSA in diabetes patients. Site-specific tri-oxidized HSA could be a potential biomarker of diabetes and provide directive of disease mechanism.

Identification of novel long non-coding RNAs as biomarker for patients with renal cell carcinoma

Project description:Long non-coding RNAs (lncRNA) play an important role in carcinogenesis, but knowledge of lncRNA expression in renal cell carcinoma is rudimental. We screened 32,183 lncRNA transcripts in malignant and adjacent normal renal tissue and determined dysregulation of approximately 4% of the lncRNA transcripts in clear cell renal cell carcinoma tissue. The distinct changes of lncRNA expression may be used to develop a non-invasive biomarker, because lncRNAs are detectable in bodily fluids. Long non-coding RNA screening and identification of potential biomarkers for type 2 diabetes.

Project description:BackgroundTo investigate new lncRNAs as molecular markers of T2D.MethodsWe used microarrays to identify differentially expressed lncRNAs and mRNAs from five patients with T2D and paired controls. Through bioinformatics analysis, qRT-PCR validation, ELISA, and receiver operating characteristic (ROC) curve analysis of 100 patients with T2D and 100 controls to evaluate the correlation between lncRNAs and T2D, and whether lncRNAs could be used in the diagnosis of T2D patients.ResultsWe identified 68 and 74 differentially expressed lncRNAs and mRNAs, respectively. The top five upregulated lncRNAs are ENST00000381108.3, ENST00000515544.1, ENST00000539543.1, ENST00000508174.1, and ENST00000564527.1, and the top five downregulated lncRNAs are TCONS_00017539, ENST00000430816.1, ENST00000533203.1, ENST00000609522.1, and ENST00000417079.1. The top five upregulated mRNAs are Q59H50, CYP27A1, DNASE1L3, GRIP2, and lnc-TMEM18-12, and the top five downregulated mRNAs are GSTM4, PODN, GLYATL2, ZNF772, and CLTC. Examination of lncRNA-mRNA interaction pairs indicated that the target gene of lncRNA XR_108954.2 is E2F2. Multiple linear regression analysis showed that XR_108954.2 (r = 0.387, p < 0.01) and E2F2 (r = 0.368, p < 0.01) expression levels were positively correlated with glucose metabolism indicators. Moreover, E2F2 was positively correlated with lipid metabolism indicators (r = 0.333, p < 0.05). The area under the ROC curve was 0.704 (95% CI: 0.578-0.830, p = 0.05) for lncRNA XR_108954.2 and 0.653 (95% CI: 0.516-0.790, p = 0.035) for E2F2.ConclusionsThis transcriptome analysis explored the aberrantly expressed lncRNAs and identified E2F2 and lncRNA XR_108954.2 as potential biomarkers for patients with T2D.
