Project description:To determine whether the expression levels of circular RNAs were altered and lay a foundation for future work, we used high-throughput microarray analysis to screen circular RNAs expression patterns in the spinal cord of adult rats after traumatic spinal cord injury (SCI), finally to evaluate the potential rat models as a platform for the development of novel therapeutic targets for spinal cord injury in future clinical studies. Overall six rats at 3 days post-SCI in two groups were used to perform the microarray.

RNA-seq Reveals Paxillin as a Key Facilitator of Endothelial Cell Migration and Angiogenesis after Spinal Cord Injury

Project description:Spinal cord injury (SCI) is a catastrophic condition resulting in significant neurological deficits. To map the altered pathways within the lesion epicenter and the surrounding rostro-caudal segments, we employed RNA-seq on injured spinal cord tissue. We sequenced samples from three level (rostral, epicenter, and caudal) at days 1, 14, and 28 post-injury to systematically define the transcriptomics profile, aiming to identify pathways related to angiogenesis after SCI. Gene set enrichment analysis (GSEA) revealed enriched pathways, including hepatocyte growth factor (HGF) receptor signalling and alpha-6 beta-4 signaling pathway, suggesting an active angiogenic signature. Furthermore, the participating genes in HGF receptor signaling were validated by qPCR analysis, demonstrating that HGF receptor signaling is involved in the pathological remodeling following SCI. Subsequently, we investigated paxillin as a candidate gene promoting endothelial cell migration through HGF receptor signaling. Immunohistochemistry data suggest paxillin’s role in mediating the endothelial cell migration and proliferation post-SCI. In summary, our findings indicate that paxillin is a key mediator of endothelial cell proliferation and migration in response to angiogenic factors like HGF following SCI. However, extensive mechanistic studies are required to fully establish paxillin’s role in endothelial cell migration and proliferation after SCI.

Circular RNAs expression profiles and potential key molecules incompletely transected spinal cord injury.

Project description:Circular RNA is implicated in numerous diseases and conditions, including traumatic injury to the central nervoussystem. However, we know little regarding their role in completely transected spinal cord injury (SCI). Our studyused high-throughput sequencing to analyze circular RNA expression in rats after experimental completetransection of spinal cord. We found differential expression in 400 circular RNAs after SCI, with 249 significantlyup-regulated and 151 significantly down-regulated. We then selected five circular RNAs for qRT-PCR validation,and the results were in agreement with RNA sequencing. Additionally, we predicted the function of circular RNAin SCI through GO and KEGG analyses, as well as the construction of circRNA/microRNA interaction networks.Furthermore, we identified circular RNA chr9:1829226-1834212 and mRNA Hdac11 as key molecules after SCI.In conclusion, this study is the first to characterize circular RNA function in rats after completely transected SCI,through identifying their differential expression. Our results provide insight into the mechanism and therapeutictargets of SCI.

Harnessing developmental dynamics of spinal cord extracellular matrix improves regenerative potential of spinal cord organoids

Project description:Neonatal spinal cord tissues exhibit remarkable regenerative capabilities as compared to adult spinal cord tissues after injury, but the role of extracellular matrix (ECM) in this process has remained elusive. Here, we found that early developmental spinal cord had higher levels of ECMproteins associated with neural development and axon growth, but fewer inhibitory proteoglycans, compared to those of adult spinal cord. Decellularized spinal cord ECM from neonatal (DNSCM) and adult (DASCM) rabbits preserved these differences. DNSCM promoted proliferation, migration, and neuronal differentiation of neural progenitor cells (NPCs) and facilitated axonal outgrowth and regeneration of spinal cord organoids more effectively than DASCM. Pleiotrophin (PTN) and Tenascin (TNC) inDNSCMwere identified as contributors tothese abilities. Furthermore,DNSCMdemonstrated superior performance as a delivery vehicle forNPCs and organoids in spinal cord injury (SCI)models. This suggests that ECMcues from early development stages might significantly contribute to the prominent regeneration ability in spinal cord.

Compared to DASCM, DNSCM promoted proliferation, migration, and neuronal differentiation of neural progenitor cells (NPCs), as well as facilitated the long-distance axonal outgrowth and axon regeneration of spinal cord organoids. Pleiotrophin (PTN) and Tenascin (TNC) in DNSCM were identified as contributors to the remarkable neural regeneration ability. Furthermore, DNSCM demonstrated superior performance when used as a delivery vehicle for NPCs and organoids in rats with spinal cord injury (SCI). It suggests that ECM cues derived from different development stage might contribute to the distinct regeneration ability of spinal cord.
